    Primary Hyperparathyroidism in Older People: Surgical Treatment with Minimally Invasive Approaches and Outcome

    Introduction. Elderly patients with primary hyperparathyroidism (pHPT) are often not referred to surgery because of their associated comorbidities that may increase surgical risk. The aim of the study was to review indications and results of minimally invasive approach parathyroidectomy in elderly patients to evaluate its impact on outcome. Materials and Methods. All patients of 70 years of age or older undergoing minimally approach parathyroidectomy at our Department from May 2005 to May 2011 were reviewed. Data collected included patients demographic information, biochemical pathology, time elapsed from pHPT diagnosis to surgical intervention, operative findings, complications, and results of postoperative biochemical studies. Results and Discussion. 37 patients were analysed. The average length of stay was 2.8 days. 11 patients were discharged within 24 hours after their operation. Morbidity included 6 transient symptomatic postoperative hypocalcemias while one patient developed a transient laryngeal nerve palsy. Time elapsed from pHPT diagnosis to first surgical visit evidences that the elderly patients were referred after their disease had progressed. Conclusions. Our data show that minimally invasive approach to parathyroid surgery seems to be safe and curative also in elderly patients with few associated risks because of combination of modern preoperative imaging, advances in surgical technique, and advances in anesthesia care

    Rock-face temperature at high-elevation sites: a new measuring approach

    The Alpine environment and in particular the cryosphere, is responding quickly and with great intensity to climate change. Temperature increase observed in the Alps urge the scientific community to study not only air temperature but also rock temperature, to deepen the knowledge about thermal properties of the potentially unstable geological materials. The metrological traceability of measurements is fundamental for data comparability in space and in time and this can be achieved by the use of calibrated instruments and with the evaluation of measurement uncertainties. Here we present some preliminary results of rock-face temperature analysis based on data acquired at high-elevation sites, by means of sensors with documented traceability to International System of Units Standards and evaluated measurement uncertainty.     H2AFZ:A Novel Prognostic Marker in Canine Melanoma and a Predictive Marker for Resistance to CDK4/6 Inhibitor Treatment

    Uncontrolled proliferation is a key feature of tumor progression and malignancy. This suggests that cell-cycle related factors could be exploited as cancer biomarkers and that pathways specifically involved in the cell cycle, such as the Rb-E2F pathway, could be targeted as an effective anti-tumor therapy. We investigated 34 formalin-fixed paraffin-embedded (FFPE) tissue samples of canine cutaneous melanocytoma, cutaneous melanoma, and oral melanoma. Corresponding clinical follow-up data were used to determine the prognostic value of the mRNA expression levels of several cell cycle regulated E2F target genes (E2F1, DHFR, CDC6, ATAD2, MCM2, H2AFZ, GINS2, and survivin/BIRC5). Moreover, using four canine melanoma cell lines, we explored the possibility of blocking the Rb-E2F pathway by using a CDK4/6 inhibitor (Palbociclib) as a potential anti-cancer therapy. We investigated the expression levels of the same E2F target gene transcripts before and after treatment to determine the potential utility of these molecules as predictive markers. The E2F target gene H2AFZ was expressed in 91.43% of the primary tumors and H2AFZ expression was significantly higher in cases with unfavorable clinical outcome. Among the other tested genes, survivin/BIRC5 showed as well-promising results as a prognostic marker in canine melanoma. Three of the four tested melanoma cell lines were sensitive to the CDK4/6 inhibitor. The resistant cell line displayed higher expression levels of H2AFZ before treatment compared to the CDK4/6 inhibitor-sensitive cell lines. The present results suggest that CDK4/6 inhibitors could potentially be used as a new anti-cancer treatment for canine melanoma and that H2AFZ could serve as a prognostic and predictive marker for patient selection.</p

    Agricultural abandonment in Mediterranean reclaimed peaty soils: Long-term effects on soil chemical properties, arbuscular mycorrhizas and CO2 flux

    In the last century, most peatlands were reclaimed for agricultural purposes, which led to peat degradation and to severe subsidence, and thus too wet conditions for crops. In some areas this has therefore led to wide agricultural abandonment. However, studies on the effect of agricultural abandonment as a potential restoration tool are lacking. In this study, the effectiveness and the restoration potential of agricultural abandonment in reducing peat degradation and in improving soil microbial biodiversity were evaluated. The main chemical parameters, arbuscular mycorrhizal (AM) fungal diversity and soil respiration partitioning were used to assess the long-term effect of 15 years of agricultural abandonment (Aband) in a Mediterranean reclaimed peatland. An intensive maize cultivation (Cult) in the same area was used as a comparison. Multivariate analyses showed that 15 years of agricultural abandonment: did not affect the main soil chemical parameters, except for NH4+ which was lower in the Aband than in the Cult; increased AM fungal root colonization and the diversity in terms of number of families of AM fungi retrieved in roots, but decreased soil AM fungal richness; reduced total soil respiration and its autotrophic component but increased respiration by heterotrophs; determined a lower fluctuation of soil CO2 flux response to air temperature than the Cult. Thus, although some soil quality parameters were significantly improved, 15 years of agricultural abandonment may not lead to an effective restoration. Consequently, alternative and sustainable solutions for the protection and preservation of Mediterranean peatlands need to be develope

    This review is dedicated to the memory of Prof. Domenico Mariotti, who significantly contributed to establishing the Italian research community in Agricultural Genetics and carried out the first experiments of Agrobacterium-mediated plant genetic transformation and regeneration in Italy during the 1980s. Following his scientific interests as guiding principles, this review summarizes the recent advances obtained in plant biotechnology and fundamental research aiming to: (i) Exploit in vitro plant cell and tissue cultures to induce genetic variability and to produce useful metabolites; (ii) gain new insights into the biochemical function of Agrobacterium rhizogenes rol genes and their application to metabolite production, fruit tree transformation, and reverse genetics; (iii) improve genetic transformation in legume species, most of them recalcitrant to regeneration; (iv) untangle the potential of KNOTTED1-like homeobox (KNOX) transcription factors in plant morphogenesis as key regulators of hormonal homeostasis; and (v) elucidate the molecular mechanisms of the transition from juvenility to the adult phase in Prunus tree species
