Nonspecific interactions alter lipopolysaccharide patterns and protein mobility on sodium dodecyl sulfate polyacrylamide gels
In testing whether bacterial lipopolysaccharide (LPS) was a natural substrate for an esterase from the soil amebae Dictyostelium discoideum, we observed altered banding patterns of the LPS and changed protein mobility on sodium dodecyl sulfate (SDS) polyacrylamide gels after incubation of LPS with the enzyme. The initial interpretation of these results was that the enzyme had removed ester-linked acyl chains from the LPS, leading to a change in its migration on gels. However, esterase inactivated by treatment with either dithiothreitol (DTT), heat, or SDS generated the same mobility shifts. Bovine serum albumin (BSA) also induced the same change in the electrophoretic pattern. We conclude that the altered LPS patterns and protein mobility on SDS gels were caused by nonspecific interactions between LPS and protein

Mutant glycyl-tRNA synthetase (Gars) ameliorates SOD1G93A motor neuron degeneration phenotype but has little affect on Loa dynein heavy chain mutant mice
Background:
In humans, mutations in the enzyme glycyl-tRNA synthetase (GARS) cause motor and sensory axon loss in the peripheral nervous system, and clinical phenotypes ranging from Charcot-Marie-Tooth neuropathy to a severe infantile form of spinal muscular atrophy. GARS is ubiquitously expressed and may have functions in addition to its canonical role in protein synthesis through catalyzing the addition of glycine to cognate tRNAs.
Methodology/Principal findings:
We have recently described a new mouse model with a point mutation in the Gars gene resulting in a cysteine to arginine change at residue 201. Heterozygous Gars^{C201R/+} mice have locomotor and sensory deficits. In an investigation of genetic mutations that lead to death of motor and sensory neurons, we have crossed the Gars^{C201R/+} mice to two other mutants: the TgSOD1^{G93A} model of human amyotrophic lateral sclerosis and the Legs at odd angles mouse (Dync1h1^{Loa}) which has a defect in the heavy chain of the dynein complex. We found the Dync1h1^{Loa/+}; Gars^{C201R/+} double heterozygous mice are more impaired than either parent, and this is may be an additive effect of both mutations. Surprisingly, the Gars^{C201R} mutation significantly delayed disease onset in the SOD1^{G93A}; Gars^{C201R/+} double heterozygous mutant mice and increased lifespan by 29% on the genetic background investigated.
Conclusions/Significance:
These findings raise intriguing possibilities for the study of pathogenetic mechanisms in all three mouse mutant strains

Effects of multi-stage dehumidified-air drying on the polyphenol content of Hydrocotyle bonariensis
Traditional drying methods involve high temperatures that degrade heat-sensitive compounds. Dehumidified-air drying, an alternative to traditional drying methods, is suitable for heat-sensitive compounds; however, it consumes a large amount of energy and is comparatively expensive. In this study, a multi-chamber dehumidified-air dryer was designed to dry Hydrocotyle bonariensis, and the retention of the polyphenol content of Hydrocotyle bonariensis under various drying conditions was examined. Multi-chamber dehumidified-air drying involves two chambers; each chamber was operated at temperatures of 30, 40, and 50°C with air volumetric flow rates of 30 and 50 L/min. The results indicated that the highest retention of total phenolic content and total flavonoid content, 24.67 mg of GAE/g dry weight (DW) and 2.204 mg of catechin/g DW, respectively, was obtained at 50°C with a 50 L/min air flow rate in the first drying chamber. Multi-stage dehumidified-air dryers have the potential to dry heat-sensitive products with reduced energy consumption
Genótipos de milho com potencial para comercialização no estágio verde de maturação no Estado do Amazonas.
Resumo: O milho no estágio verde de maturação - milho verde - representa uma alternativa viável de produção e agregação de valor para pequenas propriedades no Amazonas. Com o objetivo de avaliar a adaptabilidade de genótipos e verificar qual a melhor época para colheita para os diferentes cultivares, foi conduzido um experimento em Latossolo Amarelo distrófico no município de Manaus, utilizando o delineamento experimental de blocos casualizados com oito tratamentos e quatro repetições. Os tratamentos foram definidos por oito genótipos de milho, sendo seis variedades (Sol da Manhã, BR 106, BR 5110, Saracura, Cativerde, AG 1051, HTMV1 e Campeão) e dois híbridos (HTMV1 e AG 1051) que foram colhidos em cinco épocas diferentes e avaliados parâmetros fitotécnicos, organolépticos e mercadológicos dos diferentes genótipos. Os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Duncan a 5% de probabilidade. Os resultados evidenciaram variações no estande, altura de planta, altura de inserção das espigas, diâmetro do colmo, número de espigas totais, número de espigas comerciais, comprimento da espiga, diâmetro da espiga, número de carreiras, número de fileiras, brix e cor dos grãos em função dos cultivares utilizados. O número de espigas comerciais foi maior para os genótipos AG 1051 e BR 5110, com 42708 e 38542 espigas comerciais ha-1, respectivamente. Considerando o preço da "mão de milho", nome dado a 50 espigas comerciais de milho verde comercializada no Amazonas, custando R7,21), o produtor teria uma renda bruta variando de R 12.812,00
Phagosomal Proteins of \u3ci\u3eDictyostelium discoideum\u3c/i\u3e
In recognizing food particles, Dictyostelium cell-surface molecules initiate cytoskeletal rearrangements that result in phagosome formation. After feeding D. discoideum cells latex beads, early phagosomes were isolated on sucrose step gradients. Protein analyses of these vesicles showed that they contained glycoproteins and surface-labeled species corresponding to integral plasma membrane proteins. Cytoskeletal proteins also were associated with phagosomes, including myosin II, actin and a 30 kDa-actin bundling protein. As seen by the acridine orange fluorescence of vesicles containing bacteria, phagosomes were acidified rapidly by a vacuolar H+-ATPase that was detected by immunoblotting. Except for the loss of cytoskeletal proteins, few other changes over time were noted in the protein profiles of phagosomes, suggesting that phagosome maturation was incomplete. The indigestibility of the beads possibly inhibited further endocytic processing, which has been observed by others. Since nascent phagosomes contained molecules of both the cytoskeleton and plasma membrane, they will be useful in studies aimed at identifying specific protein associations occurring between membrane proteins and the cytoskeleton during phagocytosis
