    Striped marlin in their Pacific Ocean milieu: Vertical movements and habitats vary with time and place

    Striped marlin, Kajikia audax, a top bycatch of the longline fishery, has been designated as being in overfished condition in the Western and Central North Pacific, and overfishing is still occurring. This prompts an urgent need to devise conservation and management measures based on the best, current information on the biology and ecology of this species. Despite decades of conventional tagging around the Hawaiian waters, ecological research on striped marlin in the Central North Pacific has been lacking since 2005, and little is known about striped marlin’s vertical habitat, diving behavior and bycatch vulnerability in this area. To address this knowledge void, 31 popup satellite archival tags (4 X-Tags; Microwave Telemetry, Inc. and 27 MiniPATs; Wildlife Computers Inc.) were deployed on striped marlin (138-192 cm eye fork length) between 2016 and 2019 via the Hawaii-based longline fleet. Transmitted time series records revealed striped marlin spent 38 and 81% of their day and night in the top 5 m, with median daytime and night depths of 44 m and 2 m, respectively. Temperatures experienced were 23.3°C, daytime median, and 24.6°C, nighttime median, to a minimum of 7.6°C at the deepest depth logged, 472 m. Striped marlin exhibited distinct swimming behaviors, including diel depth distributions, excursions around the top of the thermocline, and extended time at the surface, most likely reflecting the dynamic biophysical environment and intrinsic life history of this highly migratory predator. High post-release survivorship (86%) in tagged striped marlin, and their predominant use of the sea surface and mixed layer indicate that live release measures can be a viable bycatch reduction strategy

    Recent years have seen increased interest in early intervention in and prevention of mental health difficulties during adolescence In this pilot study a structured expressive textile art and fashion based ETAFB intervention was conducted to evaluate the beneficial effects on personal wellbeing and social interaction of youth with emotional distress A one-group pretest-posttest pilot study was conducted among 18 youth with emotional distress All the participants attended four sessions of ETAFB intervention All the participants were subjected to a preintervention test and a 4-week postintervention test using the Personal Wellbeing Index School Children PWI-SC 12-item General Health Questionnaire GHQ-12 and Social Interaction Anxiety Scale SIAS The study findings revealed that the ETAFB intervention was associated with significant improvements in some of the measurements This information provides the groundwork for a further study to investigate the effect of the ETAFB intervention on the relationship between youth well-being and facilitator

    A Randomized Controlled Trial of Auricular Transcutaneous Electrical Nerve Stimulation for Managing Posthysterectomy Pain

    Background. A patient- and assessor-blinded randomized controlled trial was conducted to examine the effectiveness of auricular transcutaneous electrical nerve stimulation (TENS) in relieving posthysterectomy pain. Method. Forty-eight women who had undergone a total abdominal hysterectomy were randomly assigned into three groups (n = 16 each) to receive either (i) auricular TENS to therapeutic points (the true TENS group), (ii) auricular TENS to inappropriate points (the sham TENS group), or (iii) 20 minutes of bed rest with no stimulation (the control group). The intervention was delivered about 24 hours after the operation. A visual analogue scale was used to assess pain while resting (VAS-rest) and upon huffing (VAS-huff) and coughing (VAS-cough), and the peak expiratory flow rate (PEFR) was assessed before and at 0, 15, and 30 minutes after the intervention. Result. As compared to the baseline, only the true TENS group reported a significant reduction in VAS-rest (P = .001), VAS-huff (P = .004), and VAS-cough (P = .001), while no significant reduction in any of the VAS scores was seen in the sham TENS group (all P > .05). In contrast, a small rising trend was observed in the VAS-rest and VAS-huff scores of the control group, while the VAS-cough score remained largely unchanged during the period of the study. A between-group comparison revealed that all three VAS scores of the true TENS group were significantly lower than those of the control group at 15 and 30 minutes after the intervention (all P < .02). No significant between-group difference was observed in PEFR at any point in time. Conclusion. A single session of auricular TENS applied at specific therapeutic points significantly reduced resting (VAS-rest) and movement-evoked pain (VAS-huff, VAS-cough), and the effects lasted for at least 30 minutes after the stimulation. The analgesic effects of auricular TENS appeared to be point specific and could not be attributed to the placebo effect alone. However, auricular TENS did not produce any significant improvement in the performance of PEFR

    Immunopanning purification and long-term culture of human retinal ganglion cells

    Purpose: To establish a robust method to isolate primary retinal ganglion cells (RGCs) from human fetal retina for long-term culture while maintaining neuronal morphology and marker protein expression. Methods: A total of six human retinas were obtained from aborted fetuses at 10 to 12 weeks of gestation with informed consent from mothers. RGCs were isolated and purified by a modified two-step immunopanning procedure. The cells were maintained in a serum-free defined medium supplemented with brain-derived neurotrophic factor, ciliary neutrophic factor, and forskolin. The viable RGCs and the extent of neurite outgrowth were examined by calcein-acetoxymethylester assay. Expression of RGC markers was studied by immunocytochemistry. Results: Primary RGCs from human fetal retinas were isolated and maintained in vitro for one month with substantial neurite elongation. In cell culture, almost 70% of the isolated cells attached, spread, and displayed numerous dendrites. They were immunoreactive to RGC-specific markers (Thy-1, TUJ-1, and Brn3a) and negative for glial fibrillary acidic protein and amacrine cells marker HPC-1.     Effect of Indocyanine Green and Illumination on Gene Expression in Human Retinal Pigment Epithelial Cells

    PURPOSE. To investigate the biological effects of indocyanine green (ICG) and acute illumination on human retinal pigment epithelial (RPE) cells. METHODS. Three concentrations (0, 0.25, and 2.5 mg/mL) of ICG were applied to ARPE19 cells for 1 minute. After isotonic rinsing, the cells were irradiated with a light beam with a wavelength spectrum from 400 to 800 nm and an output of 1850 lumens for 15 minutes. The cells were collected at timed intervals for the investigation of cell death and expression of stress-response genes by reverse transcription-polymerase chain reaction, immunofluorescence, and Western blot analysis. RESULTS. After ICG incubation, photoreactive changes were observed in the RPE cells. A reduction in cellular viability and considerable shrinkage of the cells were observed. The expressions of the apoptosis-related genes p53 and bax and the cell cycle arrest protein p21 were upregulated in cells treated with both ICG and light. Of the early-response genes, the expression of c-fos was specifically enhanced by light, with additive effects from the presence of ICG. Such stimulatory effects on these gene expressions were greater at 2.5 mg/mL than at 0.25 mg/mL ICG. CONCLUSIONS. ICG in the presence of acute illumination can elicit cell-cycle arrest and even apoptosis in RPE cells.
