    Embryo splitting can increase the quantity but not the quality of blastocysts

    AbstractObjectiveIn this study, we investigated the developmental potential of single blastomeres that were obtained from 4-cell mice embryos that were split during the blastocyst stage.Materials and MethodsImprinting Control Region (ICR) mice (age: 6–8 weeks), were superovulated and mated with a single fertile male of the same strain. We obtained 2-cell embryos that were then cultured in 4 groups (×4) with Human tubal fluid (HTF) supplemented with 12% fetal bovine serum. When these embryos reached the 4-cell stage, their zonae pellucidae were removed and every single blastomere was isolated by repeated pipetting with Ca/Mg2+-free medium. The isolated blastomeres (study group) and the intact embryos (control group) were then cultured to determine the blastocyst formation rate and quality.ResultsWe collected a total of 936 embryos from 524 morphologically intact, top-grade embryos in the 4-cell stage from 80 stimulated mice. We used 356 of these embryos to isolate the blastomeres. The remaining 168 embryos were cultured as controls. A total of 1312 single blastomeres were obtained and cultured in vitro. Among these, 620 blastocysts were harvested from the original embryos compared with 136 blastocysts that were harvested from the control group. The overall blastocyst formation rate was 174.2% (620 blastocysts from 356 embryos) for the study group compared with 81.5% (136 blastocysts from 168 embryos) for the control group. The study group was 43.3% (268 of 620) top-grade blastocysts compared with 91% (152 of 168) of the control group. Taken together, the percentage of top-grade blastocysts obtained per original embryo in the split group was 75.4% (174.2%×43.3%) compared with 74.2% (81.5%×91%) for the control group.ConclusionsEmbryo splitting can increase the number of blastocysts. However, the percentage of available top-grade blastocysts is the same compared with nonsplit embryos. Embryo splitting may not be a cost-effective technique for the generation of high-quality mouse blastocysts

    The Impacts Of Presentation Modes And Product Involvements On “Line” Short Message Service (SMS) Advertising Effectiveness

    In today’s ubiquitous commerce (UC) era, short message service (SMS) advertisement has played an important role in the world of marketing. Convenience and economical reasons influence SMS usage frequency along with social involvement to influence attitudes towards SMS advertising. SMS advertising creates numerous opportunities for the marketers in promoting their products effectively. Adopting the competition for attention theory as the theoretical framework, we developed hypotheses to investigate the influences of presentation mode and involvement on SMS advertising performance (recall of advertising information). An experiment was conducted to examine the effects of three types of information presentation modes (text-only, image-text, and emoji-text) in the contexts of two product types (high- versus low-involvement products) in the “LINE” SMS environment. Specifically, in this current study, we allocate participants to six experimental environments (text-only for high-involvement products, text-only for low-involvement products, image-text for high-involvement products, image-text for low-involvement products, emoji-text for high-involvement products and emoji-text for low-involvement products) randomly to collected empirical data to examine the proposed hypotheses. The research findings are expected to provide instrumental guidelines for the practitioners to better achieve the goals of ads in the “LINE” SMS environment. Also, the empirical results may provide insights into the research of advertising interface design of SMS and integrating efforts from cognitive science and vision research to understand users’ involvement of SMS advertising processes

    Space Net Optimization

    Most metaheuristic algorithms rely on a few searched solutions to guide later searches during the convergence process for a simple reason: the limited computing resource of a computer makes it impossible to retain all the searched solutions. This also reveals that each search of most metaheuristic algorithms is just like a ballpark guess. To help address this issue, we present a novel metaheuristic algorithm called space net optimization (SNO). It is equipped with a new mechanism called space net; thus, making it possible for a metaheuristic algorithm to use most information provided by all searched solutions to depict the landscape of the solution space. With the space net, a metaheuristic algorithm is kind of like having a ``vision'' on the solution space. Simulation results show that SNO outperforms all the other metaheuristic algorithms compared in this study for a set of well-known single objective bound constrained problems in most cases.Comment: 12 pages, 6 figure

    Glycogen synthase kinase 3α and 3β have distinct functions during cardiogenesis of zebrafish embryo

    <p>Abstract</p> <p>Background</p> <p>Glycogen synthase kinase 3 (GSK3) encodes a serine/threonine protein kinase, is known to play roles in many biological processes. Two closely related GSK3 isoforms encoded by distinct genes: GSK3α (51 kDa) and GSK3β (47 kDa). In previously studies, most GSK3 inhibitors are not only inhibiting GSK3, but are also affecting many other kinases. In addition, because of highly similarity in amino acid sequence between GSK3α and GSK3β, making it difficult to identify an inhibitor that can be selective against GSK3α or GSK3β. Thus, it is relatively difficult to address the functions of GSK3 isoforms during embryogenesis. At this study, we attempt to specifically inhibit either GSK3α or GSK3β and uncover the isoform-specific roles that GSK3 plays during cardiogenesis.</p> <p>Results</p> <p>We blocked <it>gsk3α </it>and <it>gsk3β </it>translations by injection of morpholino antisense oligonucleotides (MO). Both <it>gsk3α</it>- and <it>gsk3β</it>-MO-injected embryos displayed similar morphological defects, with a thin, string-like shaped heart and pericardial edema at 72 hours post-fertilization. However, when detailed analysis of the <it>gsk3α</it>- and <it>gsk3β</it>-MO-induced heart defects, we found that the reduced number of cardiomyocytes in <it>gsk3α </it>morphants during the heart-ring stage was due to apoptosis. On the contrary, <it>gsk3β </it>morphants did not exhibit significant apoptosis in the cardiomyocytes, and the heart developed normally during the heart-ring stage. Later, however, the heart positioning was severely disrupted in <it>gsk3β </it>morphants. <it>bmp4 </it>expression in <it>gsk3β </it>morphants was up-regulated and disrupted the asymmetry pattern in the heart. The cardiac valve defects in <it>gsk3β </it>morphants were similar to those observed in <it>axin1 </it>and <it>apc</it><sup><it>mcr </it></sup>mutants, suggesting that GSK3β might play a role in cardiac valve development through the Wnt/β-catenin pathway. Finally, the phenotypes of <it>gsk3α </it>mutant embryos cannot be rescued by <it>gsk3β </it>mRNA, and vice versa, demonstrating that GSK3α and GSK3β are not functionally redundant.</p> <p>Conclusion</p> <p>We conclude that (1) GSK3α, but not GSK3β, is necessary in cardiomyocyte survival; (2) the GSK3β plays important roles in modulating the left-right asymmetry and affecting heart positioning; and (3) GSK3α and GSK3β play distinct roles during zebrafish cardiogenesis.</p

    Eddy-Kuroshio interaction processes revealed by mooring observations off Taiwan and Luzon

The definitive version was published in Geophysical Research Letters 42 (2015): 8098–8105, doi:10.1002/2015GL065814.The influence and fate of westward propagating eddies that impinge on the Kuroshio were observed with pressure sensor-equipped inverted echo sounders (PIESs) deployed east of Taiwan and northeast of Luzon. Zero lag correlations between PIES-measured acoustic travel times and satellite-measured sea surface height anomalies (SSHa), which are normally negative, have lower magnitude toward the west, suggesting the eddy-influence is weakened across the Kuroshio. The observational data reveal that impinging eddies lead to seesaw-like SSHa and pycnocline depth changes across the Kuroshio east of Taiwan, whereas analogous responses are not found in the Kuroshio northeast of Luzon. Anticyclones intensify sea surface and pycnocline slopes across the Kuroshio, while cyclones weaken these slopes, particularly east of Taiwan.     Ameliorate Effects of Li-Fu Formula on IL-6-Mediated Cardiac Hypertrophy in Hamsters Fed with a Hyper-Cholesterol Diet

    Hypercholesterolemia diets are considered as major sources to cause cardiac hypertrophy. This study intends to evaluate the effects of Li-Fu formula on cardiac hypertrophy induced by hypercholesterolemia diet. Twenty-four male Golden Syrian hamsters were randomly divided into control, cholesterol and Li-Fu formula groups and fed with different experimental diets for 2 months. Histopathological analysis and western blotting were performed to measure the myocardial architecture, and various cardiac hypertrophy-associated molecules in the excised left ventricle from hamsters. The ratios of whole heart weight/body weight (BW) and left ventricle weight/BW were significantly higher in the cholesterol group but significantly lower in the Li-Fu formula group. The protein levels of both atrial natriuretic peptide and brain natriuretic peptide were significantly increased in the cholesterol group but significantly reduced in the Li-Fu formula group. Additionally, significantly increased interleukin-6, STAT3, MEK5, p-ERK5 and non-cardiomyocyte proliferate signal molecules such as p-MEK and p-ERK, were detected in the cholesterol group but significantly reduced in the Li-Fu formula group. Notably, no significant variations of inflammatory signaling molecules, including p-P38 and p-JNK, were detected in all groups. Our experimental results demonstrated the significant reductions of cardiac hypertrophy and related eccentric hypertrophy signaling, non-cardiomyocyte proliferate signaling in the excised left ventricle of hamsters from the Li-Fu formula. We suggested the protective effects of Li-Fu formula on cardiac hypertrophy that may be useful in prevention or treatment of hypertrophy-associated cardiovascular diseases

    Sitagliptin and Fractures in Type 2 Diabetes: A Nationwide Population-Based Propensity-Matching Study

    Background: Sitagliptin, a dipeptidyl peptidase-4 inhibitor possibly affects bone turnover. We conducted this cohort study to determine whether sitagliptin is associated with an increased risk of fracture.Methods: The sitagliptin cohort included 1,578 patients aged 20 years and above. The nonsitagliptin cohort comprised propensity-score matched patients at a ratio of 1:1. The primary outcome was the incidence of fractures, which was evaluated using Kaplan–Meier survival analysis and proportional hazards modeling.Results: The mean age of patients in the sitagliptin and nonsitagliptin cohorts was 63.1 and 63.3 years, respectively. The incidence of fractures in the sitagliptin cohort was 46 per 1,000 person-years and that in the nonsitagliptin cohort was 40.8 per 1,000 person-years. Compared with patients in the nonsitagliptin cohort, those in the sitagliptin cohort who received sitagliptin for ≥250 days had a higher risk of fracture (aHR = 1.32, 95% CI = 1.06–1.64).Conclusion: Using sitaglipin ≥250 days was associated with an increased risk of fracture
