    Fall cone test on biopolymer-treated clay

    Fall cone tests were conducted to evaluate the consistency variations of clay soils treated with six types of biopolymers, e.g. carrageenan kappa gum (KG), locust bean gum (BG), xanthan gum (XG), agar gum (AG), guar gum (GG) and sodium alginate (SA) at various concentrations (e.g. between 0.1% to 5% biopolymer to soil mass ratio). The dependences of shear viscosity on water content, and undrained shear strength on water content were established. The results indicated that KG and SA increased the liquid limit (LL) of treated soils after the biopolymer content exceeded a certain limit (e.g. 0.5%), BG and GG contributed to a peak point in LL at biopolymer concentration of 1% to 2%, while XG and AG almost did not change the LL at all. The plastic limit (PL) was about 25% to 50% of the LL, leading to a trend of plasticity index (PI) similar to liquid limit. In order to further simplify the testing procedure and get the Atterberg limits for biopolymer-treated soil, one-point method was adopted

    Effect of slickwater-alternate-slurry injection on proppant transport at field scales : a hybrid approach combining experiments and deep learning

    Proppant transport in underground fractures plays a key role in mitigating sand screen-out and enhancing the stimulated production for hydraulic fracturing. The effects of field pumping schedules, however, are not fully studied. We investigate the effect of slickwater-alternate-slurry injection on proppant transport at field-practical scales. A new hybrid approach is proposed to directly connect experimental studies with field operations, which consists of observation experiments, calculations, and deep learning (DL) workflow. The experiments reveal that the alternate injection induces the unexpected proppant ridge. The modified calculations (considering the ridge height) are proposed to extract features for training the DL algorithm. The workflow predicts the downhole pressure (mainly governed by proppant injection) for error analyses. Approximately 20.2% of the error is eliminated by considering the proppant ridge, thus demonstrating its effect on proppant injection. The predictions are significantly improved in early and late periods of fracturing operations when the fracture is initially created and highly filled. The sensitivity analysis suggests that the pump rate may dominate the ridge height compared with other hydraulic parameters. The study of proppant ridge complements the mechanisms of proppant transport, which is essential for controlling fracturing pressure and boosting the proppant injection

    Plasmodium falciparumvar Gene Is Activated by Its Antisense Long Noncoding RNA

    Plasmodium falciparum erythrocyte membrane protein 1, encoded by var gene, is an immunodominant antigen mediating immune evasion in humans. At a given time, only a single var gene is commonly expressed in one parasite. However, the regulation mechanism of var transcription remains largely unknown. In this study, we identified the antisense long non-coding RNA (aslncRNA) derived from var intron as an activation factor for the corresponding var gene. The exogenous artificial var aslncRNA transcribed by T7 RNA polymerase from episome can specifically activate the homologous var gene, and the exogenous aslncRNA activates transcription of both var mRNA and endogenous aslncRNA in a manner independent of the conserved intron sequence within the var gene family. Interestingly, the newly activated var gene and the previously dominant var gene then could be co-expressed in the same parasite nuclei, which suggests that the aslncRNA-mediated var gene activation could escape from the control of mutually exclusively expression of the var gene family. Together, our work shows that var aslncRNA is the activator responsible for var gene transcriptional regulation

    Strain regulating and kinetics accelerating of micro-sized silicon anodes via dual-size hollow graphitic carbons conductive additives

    Micro-sized silicon (mu Si) anode features fewer interfacial side reactions and lower costs compared to nanosized silicon, and has higher commercial value when applied as a lithium-ion battery (LIB) anode. However, the high localized stress generated during (de)lithiation causes electrode breakdown and performance deterioration of the mu Si anode. In this work, hollow graphitic carbons with tailored dual sizes are employed as conductive additives for the mu Si anode to overcome electrode failure. The dual-size hollow graphitic carbons (HGC) additives consist of particles with micrometer size similar to the mu Si particles; these additives are used for strain regulation. Additionally, nanometer-size particles similar to commercial carbon black Spheron (SP) are used mainly for kinetics acceleration. In addition to building an efficient conductive network, the dual-size hollow graphitic carbon conductive additive prevents the fracture of the electrode by reducing local stress and alleviating volume expansion. The mu Si anode with dual-size hollow graphitic carbons as conductive additives achieves an impressive capacity of 651.4 mAh g(-1) after 500 cycles at a high current density of 2 A g(-1). These findings suggest that dual-size hollow graphitic carbons are expected to be superior conductive additives for micro-sized alloy anodes similar to mu Si.Web of Scienc

    Cancer-associated fibroblast infiltration in osteosarcoma: the discrepancy in subtypes pathways and immunosuppression

    Introduction: Osteosarcoma (OS), the primary malignant bone tumor, has a low survival rate for recurrent patients. Latest reports indicated that cancer-associated fibroblasts (CAFs) were the main component of tumor microenvironment, and would generate a variable role in the progression of tumors. However, the role of CAFs is still few known in osteosarcoma.Methods: The processed RNA-seq data and the corresponding clinical and molecular information were retrieved from the Cancer Genome Atlas Program (TCGA) database and processed data of tumor tissue was obtained from Gene Expression Omnibus (GEO) database. Xcell method was used in data processing, and Gene set variation analysis (GSVA) was used to calculates enrichment scores. Nomogram was constructed to evaluate prognostic power of the predictive model. And the construction of risk scores and assessment of prognostic predictive were based on the LASSO model.Results: This study classified Cancer Genome Atlas (TCGA) cohort into high and low CAFs infiltrate phenotype with different CAFs infiltration enrichment scores. Then TOP 9 genes were screened as prognostic signatures among 2,488 differentially expressed genes between the two groups. Key prognostic molecules were CGREF1, CORT and RHBDL2 and the risk score formula is: Risk-score = CGREF1*0.004 + CORT*0.004 + RHBDL2*0.002. The signatures were validated to be independent prognostic factors to predict tumor prognosis with single-factor COX and multi-factor COX regression analyses and Norton chart. The risk score expression of risk score model genes could predict the drug resistance, and significant differences could be found between the high and low scoring groups for 17-AAG, AZD6244, PD-0325901 and Sorafenib.Discussion: To sum up, this article validated the prediction role of CAF infiltration in the prognosis of OS, which might shed light on the treatment of OS
