    Magnetic Leakage Internal Detection Device and Series-Parallel Detection Method for Small Diameter Ferromagnetic Spiral Heat Exchanger Tubes

    Based on the calculation of magnetic circuit of the detection probe, this article designs a kind of magnetic leakage detection device and series-parallel detection method of small diameter spiral heat exchange tube to realize the safety detection of small diameter spiral heat exchange tube. The detection device includes a detection probe and a probe drive mechanism, which drives the detection probe to move in the spiral heat exchange tube. The detection probe includes an intermediate connector, a magnetizer which is arranged coaxially at both ends of the intermediate connector in turn, a steel ball support body and an end connector. The designed detection probes are mixed in series and parallel, and multiple detection probes are put together in series to be placed in a spiral heat exchange tube to form a group of detection probes in series. Then, multiple groups of detection probes in series are placed in each spiral heat exchange pipe, forming a series-parallel combination, which can detect multiple spiral heat exchange pipes at the same time and improve the detection efficiency greatly

    Spatiotemporal Evolution of Land Subsidence in the Beijing Plain 2003–2015 Using Persistent Scatterer Interferometry (PSI) with Multi-Source SAR Data

    Land subsidence is one of the most important geological hazards in Beijing, China, and its scope and magnitude have been growing rapidly over the past few decades, mainly due to long-term groundwater withdrawal. Interferometric Synthetic Aperture Radar (InSAR) has been used to monitor the deformation in Beijing, but there is a lack of analysis of the long-term spatiotemporal evolution of land subsidence. This study focused on detecting and characterizing spatiotemporal changes in subsidence in the Beijing Plain by using Persistent Scatterer Interferometry (PSI) and geographic spatial analysis. Land subsidence during 2003–2015 was monitored by using ENVISAT ASAR (2003–2010), RADARSAT-2 (2011–2015) and TerraSAR-X (2010–2015) images, with results that are consistent with independent leveling measurements. The radar-based deformation velocity ranged from −136.9 to +15.2 mm/year during 2003–2010, and −149.4 to +8.9 mm/year during 2011–2015 relative to the reference point. The main subsidence areas include Chaoyang, Tongzhou, Shunyi and Changping districts, where seven subsidence bowls were observed between 2003 and 2015. Equal Fan Analysis Method (EFAM) shows that the maximum extensive direction was eastward, with a growing speed of 11.30 km2/year. Areas of differential subsidence were mostly located at the boundaries of the seven subsidence bowls, as indicated by the subsidence rate slope. Notably, the area of greatest subsidence was generally consistent with the patterns of groundwater decline in the Beijing Plain

    Performance of criteria for selecting evolutionary models in phylogenetics: a comprehensive study based on simulated datasets

    <p>Abstract</p> <p>Background</p> <p>Explicit evolutionary models are required in maximum-likelihood and Bayesian inference, the two methods that are overwhelmingly used in phylogenetic studies of DNA sequence data. Appropriate selection of nucleotide substitution models is important because the use of incorrect models can mislead phylogenetic inference. To better understand the performance of different model-selection criteria, we used 33,600 simulated data sets to analyse the accuracy, precision, dissimilarity, and biases of the hierarchical likelihood-ratio test, Akaike information criterion, Bayesian information criterion, and decision theory.</p> <p>Results</p> <p>We demonstrate that the Bayesian information criterion and decision theory are the most appropriate model-selection criteria because of their high accuracy and precision. Our results also indicate that in some situations different models are selected by different criteria for the same dataset. Such dissimilarity was the highest between the hierarchical likelihood-ratio test and Akaike information criterion, and lowest between the Bayesian information criterion and decision theory. The hierarchical likelihood-ratio test performed poorly when the true model included a proportion of invariable sites, while the Bayesian information criterion and decision theory generally exhibited similar performance to each other.</p> <p>Conclusions</p> <p>Our results indicate that the Bayesian information criterion and decision theory should be preferred for model selection. Together with model-adequacy tests, accurate model selection will serve to improve the reliability of phylogenetic inference and related analyses.</p

    Positive selection on hemagglutinin and neuraminidase genes of H1N1 influenza viruses

    BACKGROUND: Since its emergence in March 2009, the pandemic 2009 H1N1 influenza A virus has posed a serious threat to public health. To trace the evolutionary path of these new pathogens, we performed a selection-pressure analysis of a large number of hemagglutinin (HA) and neuraminidase (NA) gene sequences of H1N1 influenza viruses from different hosts. RESULTS: Phylogenetic analysis revealed that both HA and NA genes have evolved into five distinct clusters, with further analyses indicating that the pandemic 2009 strains have experienced the strongest positive selection. We also found evidence of strong selection acting on the seasonal human H1N1 isolates. However, swine viruses from North America and Eurasia were under weak positive selection, while there was no significant evidence of positive selection acting on the avian isolates. A site-by-site analysis revealed that the positively selected sites were located in both of the cleaved products of HA (HA1 and HA2), as well as NA. In addition, the pandemic 2009 strains were subject to differential selection pressures compared to seasonal human, North American swine and Eurasian swine H1N1 viruses. CONCLUSIONS: Most of these positively and/or differentially selected sites were situated in the B-cell and/or T-cell antigenic regions, suggesting that selection at these sites might be responsible for the antigenic variation of the viruses. Moreover, some sites were also associated with glycosylation and receptor-binding ability. Thus, selection at these positions might have helped the pandemic 2009 H1N1 viruses to adapt to the new hosts after they were introduced from pigs to humans. Positive selection on position 274 of NA protein, associated with drug resistance, might account for the prevalence of drug-resistant variants of seasonal human H1N1 influenza viruses, but there was no evidence that positive selection was responsible for the spread of the drug resistance of the pandemic H1N1 strains

    Combination Therapy Strategy of Quorum Quenching Enzyme and Quorum Sensing Inhibitor in Suppressing Multiple Quorum Sensing Pathways of P. <i>aeruginosa</i>

    Abstract The threat of antibiotic resistant bacteria has called for alternative antimicrobial strategies that would mitigate the increase of classical resistance mechanism. Many bacteria employ quorum sensing (QS) to govern the production of virulence factors and formation of drug-resistant biofilms. Targeting the mechanism of QS has proven to be a functional alternative to conventional antibiotic control of infections. However, the presence of multiple QS systems in individual bacterial species poses a challenge to this approach. Quorum sensing inhibitors (QSI) and quorum quenching enzymes (QQE) have been both investigated for their QS interfering capabilities. Here, we first simulated the combination effect of QQE and QSI in blocking bacterial QS. The effect was next validated by experiments using AiiA as QQE and G1 as QSI on Pseudomonas aeruginosa LasR/I and RhlR/I QS circuits. Combination of QQE and QSI almost completely blocked the P. aeruginosa las and rhl QS systems. Our findings provide a potential chemical biology application strategy for bacterial QS disruption

    Characteristics and causes analysis of Nandianzi landslide in Lingtai County, Gansu Province

    On October 3, 2021, a large landslide occurred at the loess-mudstone interface in Nandianzi, Lingtai, its successful early warning measures preventing casualties. In order to investigate the occurrence mechanism behind the Nandianzi landslide, a basic investigation was conducted, covering the topography, lithology, hydrogeological conditions, and human engineering activities related to the landslide. Based on the characteristics and differences of the crack development of the landslide, the landslide mass was divided into five blocks. The characteristics of each block were thoroughly analyzed through qualitative and quantitative analysis. The specific sliding situation of different parts of the landslide was analyzed, and further evidence was provided for the objective rationality of landslide classification and zoning, as well as the analysis of landslide mechanisms. Ultimately, it is concluded that the main causes of landslide disasters are as follows: (1) Large-scale excavation and earthwork activities at the lower and middle parts of the slope and the toe, leading to slope steepening and reduced resistance to sliding; (2) Formation of slope depressions, causing inadequate drainage and softening of the rock layer contact surface, thereby diminishing slope stability; and (3) Prolonged heavy rainfall that leads to instability and causes significant loss. While the Nandianzi landslide in Lintai county represents a successfully averted disaster, it serves as a noteworthy case study and a cautionary example for scientifically and standardizedly approaching urban construction and rural revitalization in China. This study holds significance value for monitoring, early warning,risk assessmen, and engineering treatment in comparable regions

    PFKFB3 Control of Cancer Growth by Responding to Circadian Clock Outputs

    Circadian clock dysregulation promotes cancer growth. Here we show that PFKFB3, the gene that encodes for inducible 6-phosphofructo-2-kinase as an essential supporting enzyme of cancer cell survival through stimulating glycolysis, mediates circadian control of carcinogenesis. In patients with tongue cancers, PFKFB3 expression in both cancers and its surrounding tissues was increased significantly compared with that in the control, and was accompanied with dys-regulated expression of core circadian genes. In the in vitro systems, SCC9 tongue cancer cells displayed rhythmic expression of PFKFB3 and CLOCK that was distinct from control KC cells. Furthermore, PFKFB3 expression in SCC9 cells was stimulated by CLOCK through binding and enhancing the transcription activity of PFKFB3 promoter. Inhibition of PFKFB3 at zeitgeber time 7 (ZT7), but not at ZT19 caused significant decreases in lactate production and in cell proliferation. Consistently, PFKFB3 inhibition in mice at circadian time (CT) 7, but not CT19 significantly reduced the growth of implanted neoplasms.     Not performing an OGTT results in underdiagnosis, inadequate risk assessment and probable cost increases of (pre)diabetes in Han Chinese over 40 years: a population-based prospective cohort study

    Objective: To explore the influence by not performing an oral glucose tolerance test (OGTT) in Han Chinese over 40 years. Design: Overall, 6682 participants were included in the prospective cohort study and were followed up for 3 years. Methods: Fasting plasma glucose (FPG), 2-h post-load plasma glucose (2h-PG), FPG and 2h-PG (OGTT), and HbA1c testing using World Health Organization (WHO) or American Diabetes Association (ADA) criteria were employed for strategy analysis. Results: The prevalence of diabetes is 12.4% (95% CI: 11.6–13.3), while the prevalence of prediabetes is 34.1% (95% CI: 32.9–35.3) and 56.5% (95% CI: 55.2–57.8) using WHO and ADA criteria, respectively. 2h-PG determined more diabetes individuals than FPG and HbA1c. The testing cost per true positive case of OGTT is close to FPG and less than 2h-PG or HbA1c. FPG, 2h-PG and HbA1c strategies would increase costs from complications for false-positive (FP) or false-negative (FN) results compared with OGTT. Moreover, the least individuals identified as normal by OGTT at baseline developed (pre)diabetes, and the most prediabetes individuals identified by HbA1c or FPG using ADA criteria developed diabetes. Conclusions: The prevalence of isolated impaired glucose tolerance and isolated 2-h post-load diabetes were high, and the majority of individuals with (pre)diabetes were undetected in Chinese Han population. Not performing an OGTT results in underdiagnosis, inadequate developing risk assessment and probable cost increases of (pre)diabetes in Han Chinese over 40 years and great consideration should be given to OGTT in detecting (pre)diabetes in this population. Further population-based prospective cohort study of longer-term effects is necessary to investigate the risk assessment and cost of (pre)diabetes

    Ginsenoside Rb1 Prevents H2O2-Induced HUVEC Senescence by Stimulating Sirtuin-1 Pathway

    We have previously reported that Ginsenoside Rb1 may effectively prevent HUVECs from senescence, however, the detailed mechanism has not demonstrated up to now. Recent studies have shown that sirtuin-1 (Sirt1) plays an important role in the development of endothelial senescence. The purpose of this study was to explore whether Sirt1 is involved in the action of Ginsenoside Rb1 regarding protection against H2O2-induced HUVEC Senescence.Senescence induced by hydrogen peroxide (H2O2) in human umbilical vein endothelial cells (HUVECs) was examined by analyzing plasminogen activator inhibitor-1 (PAI-1) expression, cell morphology, and senescence-associated beta-galactosidase (SA-β-gal) activity. The results revealed that 42% of control-treated HUVECs were SA-β-gal positive after treatment by 60 µmol/L H2O2, however, this particular effect of H2O2 was decreased more than 2-fold (19%) in the HUVECs when pretreated with Rb1 (20 µmol/L) for 30 min. Additionally, Rb1 decreased eNOS acetylation, as well as promoted more NO production that was accompanied by an increase in Sirt1 expression. Furthermore, upon knocking down Sirt1, the effect of Rb1 on HUVEC senescence was blunted.The present study indicated that Ginsenoside Rb1 acts through stimulating Sirt1 in order to protect against endothelial senescence and dysfunction. As such, Sirt1 appears to be of particular importance in maintaining endothelial functions and delaying vascular aging
