    Arylsulfatase - producing bacteria in marine sediments

    A total of 313 strains of bacteria which hydrolysed tripotassium phenolphthalein disulfate (PDS) were isolated from the sediments of three biotopes, namely, Vellar estuary, backwater and mangrove during the period of investigation. They were identified to the generic level. The following genera were encountered, namely, Vibrio, Bacillus, Alcaligenes, Micrococcus, Pseudomonas, Cytophaga-Flavobacterium, Aeromonas, Corynebacterium and members of Enterobacteriaceae. Vibrio and Bacillus were found to be the dominant groups representing 29.26% and 41.80% respectively of the total isolates. Because of the importance of the Vibrio group in marine environment these isolates were further identified to the species level and it included V. parahaemolyticus, V. alginolyticus, V. consticola, V. anguillarum and V. fischeri. These observations suggest that different groups of arylsulfatase – producing bacteria probably occur in marine sediments

    Cultural conditions of arylsulfatase activity in Escherichia coli

    Arylsulfatase activity and growth were estimated in Escherichia coli, isolated from marine sediment. Maximum activity was observed at pH 6.6 whereas the maximum growth was at pH 5.6. 2x10ˉ³ M is the optimum substrate concentration for the highest level of enzyme activity/synthesis as well as for its growth. In general higher substrate concentration tended to inhibit enzyme activity and also the growth of the bacterium. Maximum growth and highest enzyme activity occurred at 29°C and above this temperature decreased both of them. Besides these, glucose, sodium sulfate, sodium chloride, sodium dihydrogen phosphate, sodium acetate and ammonium chloride at higher concentrations were inhibiting the enzyme activity and growth. Above 0.2% of glucose, 3% of sodium chloride, 10x10ˉ³ M concentrations of sodium sulfate, sodium dihydrogen phosphate, sodium acetate and ammonium chloride inhibited the activity and growth also. These observations indicate that, to generalize a compound as inhibitor or activator it is difficult since this depends not only on its concentration but also on the source of the enzyme when more than one type is encountered in nature

    Hypothermia for encephalopathy in low and middle-income countries (HELIX): Study protocol for a randomised controlled trial

    BACKGROUND: Therapeutic hypothermia reduces death and disability after moderate or severe neonatal encephalopathy in high-income countries and is used as standard therapy in these settings. However, the safety and efficacy of cooling therapy in low- and middle-income countries (LMICs), where 99% of the disease burden occurs, remains unclear. We will examine whether whole body cooling reduces death or neurodisability at 18-22 months after neonatal encephalopathy, in LMICs. METHODS: We will randomly allocate 408 term or near-term babies (aged ≤ 6 h) with moderate or severe neonatal encephalopathy admitted to public sector neonatal units in LMIC countries (India, Bangladesh or Sri Lanka), to either usual care alone or whole-body cooling with usual care. Babies allocated to the cooling arm will have core body temperature maintained at 33.5 °C using a servo-controlled cooling device for 72 h, followed by re-warming at 0.5 °C per hour. All babies will have detailed infection screening at the time of recruitment and 3 Telsa cerebral magnetic resonance imaging and spectroscopy at 1-2 weeks after birth. Our primary endpoint is death or moderate or severe disability at the age of 18 months. DISCUSSION: Upon completion, HELIX will be the largest cooling trial in neonatal encephalopathy and will provide a definitive answer regarding the safety and efficacy of cooling therapy for neonatal encephalopathy in LMICs. The trial will also provide important data about the influence of co-existent perinatal infection on the efficacy of hypothermic neuroprotection. TRIAL REGISTRATION: ClinicalTrials.gov, NCT02387385.     Prostaglandin I2 Signaling Drives Th17 Differentiation and Exacerbates Experimental Autoimmune Encephalomyelitis

    BACKGROUND: Prostaglandin I(2) (PGI(2)), a lipid mediator currently used in treatment of human disease, is a critical regulator of adaptive immune responses. Although PGI(2) signaling suppressed Th1 and Th2 immune responses, the role of PGI(2) in Th17 differentiation is not known. METHODOLOGY/PRINCIPAL FINDINGS: In mouse CD4(+)CD62L(+) naïve T cell culture, the PGI(2) analogs iloprost and cicaprost increased IL-17A and IL-22 protein production and Th17 differentiation in vitro. This effect was augmented by IL-23 and was dependent on PGI(2) receptor IP signaling. In mouse bone marrow-derived CD11c(+) dendritic cells (BMDCs), PGI(2) analogs increased the ratio of IL-23/IL-12, which is correlated with increased ability of BMDCs to stimulate naïve T cells for IL-17A production. Moreover, IP knockout mice had delayed onset of a Th17-associated neurological disease, experimental autoimmune encephalomyelitis (EAE), and reduced infiltration of IL-17A-expressing mononuclear cells in the spinal cords compared to wild type mice. These results suggest that PGI(2) promotes in vivo Th17 responses. CONCLUSION: The preferential stimulation of Th17 differentiation by IP signaling may have important clinical implications as PGI(2) and its analogs are commonly used to treat human pulmonary hypertension

    The global retinoblastoma outcome study : a prospective, cluster-based analysis of 4064 patients from 149 countries

    DATA SHARING : The study data will become available online once all analyses are complete.BACKGROUND : Retinoblastoma is the most common intraocular cancer worldwide. There is some evidence to suggest that major differences exist in treatment outcomes for children with retinoblastoma from different regions, but these differences have not been assessed on a global scale. We aimed to report 3-year outcomes for children with retinoblastoma globally and to investigate factors associated with survival. METHODS : We did a prospective cluster-based analysis of treatment-naive patients with retinoblastoma who were diagnosed between Jan 1, 2017, and Dec 31, 2017, then treated and followed up for 3 years. Patients were recruited from 260 specialised treatment centres worldwide. Data were obtained from participating centres on primary and additional treatments, duration of follow-up, metastasis, eye globe salvage, and survival outcome. We analysed time to death and time to enucleation with Cox regression models. FINDINGS : The cohort included 4064 children from 149 countries. The median age at diagnosis was 23·2 months (IQR 11·0–36·5). Extraocular tumour spread (cT4 of the cTNMH classification) at diagnosis was reported in five (0·8%) of 636 children from high-income countries, 55 (5·4%) of 1027 children from upper-middle-income countries, 342 (19·7%) of 1738 children from lower-middle-income countries, and 196 (42·9%) of 457 children from low-income countries. Enucleation surgery was available for all children and intravenous chemotherapy was available for 4014 (98·8%) of 4064 children. The 3-year survival rate was 99·5% (95% CI 98·8–100·0) for children from high-income countries, 91·2% (89·5–93·0) for children from upper-middle-income countries, 80·3% (78·3–82·3) for children from lower-middle-income countries, and 57·3% (52·1-63·0) for children from low-income countries. On analysis, independent factors for worse survival were residence in low-income countries compared to high-income countries (hazard ratio 16·67; 95% CI 4·76–50·00), cT4 advanced tumour compared to cT1 (8·98; 4·44–18·18), and older age at diagnosis in children up to 3 years (1·38 per year; 1·23–1·56). For children aged 3–7 years, the mortality risk decreased slightly (p=0·0104 for the change in slope). INTERPRETATION : This study, estimated to include approximately half of all new retinoblastoma cases worldwide in 2017, shows profound inequity in survival of children depending on the national income level of their country of residence. In high-income countries, death from retinoblastoma is rare, whereas in low-income countries estimated 3-year survival is just over 50%.     Friedmann solutions and higher-dimensional 5D Kaluza–Klein solutions using mathematical packages such as Sagemath and Cadabra are calculated. A modified Friedmann equation powered by loop quantum gravity in higher dimensions is calculated in this work. Loop quantization in extra-dimensional space is predicted. Modified equation of state for non-interacting dark matter and dark energy are calculated. It has been predicted that the higher curvature due to phantom density would be a local kind of quantized curvature. The modified Friedmann solutions with Kaluza–Klein interpretation are found. To achieve a conformal exit, the non-interacting solutions are discussed in this work. The obtained results are compared with the ΛCDM and quintessence models.     Seasonal occurence of phosphate solubilizing bacteria in the vellar estuary, Porto Novo

    218-221Relative distribution of limnotolerant (freshwater tolerant) and halotolerant (saline water tolerant) phosphate solubilizing bacteria were estimated from 3 stations in Vellar estuary using different media. In addition, nutrients such as nitrite, nitrate silicate and phosphate were also analysed. Vertical distribution of phosphate dissolving bacteria at each station showed higher concentration in the sediments than in the corresponding overlying water. The ratio between the marine and freshwater phosphate solubilizing bacteria altered due to variation in salinity. In general, clayey sediment harboured more phosphate solubilizing bacteria than the sandy ones. Influences of abiotic factors like organic carbon, total phosphorus, total nitrogen and salinity on the occurrence and distribution of phosphate solubilizing bacteria were discussed

    Seasonal distribution of arylsulfatase in the marine environment at Porto Novo, East Coast of India

    192-195Arylsulfatase activity was recorded in all the sediment samples collected from different biotopes (estuarine, backwaters and mangrove). The season as well as the nature of biotope influenced the enzyme activity. Highest activity was recorded in mangrove biotope, followed by backwaters and estuary (10.39, 4.93 and 4.71 µg phenolphthalein g-1 .h-1 respectively). Irrespective of variations in biotope, higher activities, in general, were found during summer and premonsoon periods. Organic carbon, phosphate and sulfate influenced the enzyme activity. Organic carbon (0.75%) In particular was the chief influencing factor for maximum arylsulfatase activity (10.39 µg phenolphthalein g-1 .h-1) in sediments. Eventhough phosphate did not affect much of the activity, sulfate concentration (5.06 mg.g-1) inhibited the activity under natural conditions also
