    The Oak Ridge strains of wild type N. crassa grow well with high molecular wight xylan from oat spelts (Sigma X-0376) as the sole carbon source. Xylan, a substituted ß-1,4 linked polymer of xylose, induces high levels of xylanase, xylosidase and ß-galactosidase activities in both culture medium and mycelium. To assay for xylanase activity, chromogenic and fluorogenic substrates have been prepared by procedures based on those of Biely et al. 1985. Anal. Biochem. 144:142-146; Biely et al. 1985. Anal. Batches of xylan that have a tan color and granular texture should be dissolved in boiling water, precipitated with two volumes of absolute ethanol, and dried as described below before use. These techniques may also be used to prepare chromogenic and fluorogenic substrates from dextrans, glucans or other high molecular weight polysaccharides to assay endoglycosidases for which suitable assay reagents are not readily available

In contrast with their contribution to society, young carers have huge associated costs related to poor outcomes and the numbers that end up not in employment, education or training (NEET). Missing school due to caring responsibilities is likely to have an effect on future education and job prospects. Understanding the impact of the caring role on the school experience of young carers will enable school nurses to provide appropriate support for young carers, improving their school experience and subsequent outcomes

    Aspirated capacitor measurements of air conductivity and ion mobility spectra

    Measurements of ions in atmospheric air are used to investigate atmospheric electricity and particulate pollution. Commonly studied ion parameters are (1) air conductivity, related to the total ion number concentration, and (2) the ion mobility spectrum, which varies with atmospheric composition. The physical principles of air ion instrumentation are long-established. A recent development is the computerised aspirated capacitor, which measures ions from (a) the current of charged particles at a sensing electrode, and (b) the rate of charge exchange with an electrode at a known initial potential, relaxing to a lower potential. As the voltage decays, only ions of higher and higher mobility are collected by the central electrode and contribute to the further decay of the voltage. This enables extension of the classical theory to calculate ion mobility spectra by inverting voltage decay time series. In indoor air, ion mobility spectra determined from both the novel voltage decay inversion, and an established voltage switching technique, were compared and shown to be of similar shape. Air conductivities calculated by integration were: 5.3 +- 2.5 fS/m and 2.7 +- 1.1 fS/m respectively, with conductivity determined to be 3 fS/m by direct measurement at a constant voltage. Applications of the new Relaxation Potential Inversion Method (RPIM) include air ion mobility spectrum retrieval from historical data, and computation of ion mobility spectra in planetary atmospheres.Comment: To be published in Review of Scientific Instrument

    Increasing amounts of variable renewable energy sources will cause fundamental and structural changes to thermal power plant operating regimes. Maintaining key reserve requirements will lead to an increase in power plant start-ups and cycling operations for some units. An enhanced unit commitment model with energy storage and flexible CO2 capture is formulated. High-resolution on-/offshore wind data for the U.K., and probabilistic wind power forecast, model wind imbalances at operational timescales. The strategic use of flexible CO2 capture and energy storage helps maintain reserve levels, decreasing power plant cycling operations and wind curtailment. A temporally explicit variability assessment of net demand illustrates the generation flexibility requirements and the nonlinear impacts of increasing wind capacity on power plant operating regimes

    The correspondence between the 't Hooft limit of N=4 super Yang-Mills theory and tree-level IIB superstring theory on AdS(5)xS(5) in a Ramond-Ramond background at values of lambda=g^2 N ranging from infinity to zero is examined in the context of unitarity. A squaring relation for the imaginary part of the holographic scattering of identical string fields in the two-particle channels is found, and a mismatch between weak and strong 't Hooft coupling is pointed out within the correspondence.     <p><b>Background and Purpose:</b> The relationship between baseline and recurrent vascular events may be important in the targeting of secondary prevention strategies. We examined the relationship between initial event and various types of further vascular outcomes and associated effects of blood pressure (BP)–lowering.</p> <p><b>Methods:</b> Subsidiary analyses of the Perindopril Protection Against Recurrent Stroke Study (PROGRESS) trial, a randomized, placebo-controlled trial that established the benefits of BP–lowering in 6105 patients (mean age 64 years, 30% female) with cerebrovascular disease, randomly assigned to either active treatment (perindopril for all, plus indapamide in those with neither an indication for, nor a contraindication to, a diuretic) or placebo(s).</p> <p><b>Results:</b> Stroke subtypes and coronary events were associated with 1.5- to 6.6-fold greater risk of recurrence of the same event (hazard ratios, 1.51 to 6.64; P=0.1 for large artery infarction, P<0.0001 for other events). However, 46% to 92% of further vascular outcomes were not of the same type. Active treatment produced comparable reductions in the risk of vascular outcomes among patients with a broad range of vascular events at entry (relative risk reduction, 25%; P<0.0001 for ischemic stroke; 42%, P=0.0006 for hemorrhagic stroke; 17%, P=0.3 for coronary events; P homogeneity=0.4).</p> <p><b>Conclusions:</b> Patients with previous vascular events are at high risk of recurrences of the same event. However, because they are also at risk of other vascular outcomes, a broad range of secondary prevention strategies is necessary for their treatment. BP–lowering is likely to be one of the most effective and generalizable strategies across a variety of major vascular events including stroke and myocardial infarction.</p&gt

    All cubic couplings in type IIB supergravity on AdS5×S5AdS_5\times S^5 that involve two scalar fields sIs^I that are mixtures of the five form field strength on S5S^5 and the trace of the graviton on S5S^5 are derived by using the covariant equations of motion and the quadratic action for type IIB supergravity on AdS5×S5AdS_5\times S^5. All corresponding three-point functions in SYM4_4 are calculated in the supergravity approximation. It is pointed out that the scalars sIs^I correspond not to the chiral primary operators in the N=4{\cal N}=4 SYM but rather to a proper extension of the operators.Comment: Latex, 24p, misprints are correcte
