Chemoenzymatische Synthesemethoden - Zugang zur duftenden Welt der Chemie und darüber hinauseine
Die Chiralität ist ein wesentlicher Bestandteil des Lebens und mit chiralen Verbindungenkönnen biologische Systeme spezifi sch adressiert werden, um gezielte Wirkungen zu erzielen.Daher werden chiralen Verbindungen nicht nur in der Pharmazie, sondern beispielsweiseauch in der Agrar- und Lebensmittelchemie eine große Bedeutung zugeschrieben. Um denBedarf an chiralen Verbindungen zu decken sind stereoselektive Synthesemethoden gefragt.Enzyme als Biokatalysatoren sind aufgrund ihrer bekannten Chemo-, Stereo- und Regioselektivitätsehr gut geeignete Werkzeuge um chirale Verbindungen unter milden Reaktionsbedingungenherzustellen.In dieser Arbeit wurden chemoenzymatische Methoden zur stereoselektiven Synthese vonγ-Butyrolactonen und α-arylierten Michael-Addukten etabliert. Die Enreduktase wurde alsSchlüsselenzym mit weiteren Oxidoreduktasen kombiniert. Somit konnte ausgehend vonaktivierten Alkenen Verbindungen mit zwei defi nierten Stereozentren gebildet und die Grenzender Enzyme aufgezeigt werde
Enantioselective, Catalytic One‐Pot Synthesis of γ‐Butyrolactone‐Based Fragrances
Herein the preparative (1 g scale), stereoselective syntheses of various alkyl‐substituted γ‐butyrolactone fragrances 1 is described. The α,β‐unsaturated γ‐keto esters 2 as starting materials were synthesized by a Horner‐Wadsworth‐Emmons reaction and are further reduced by an ene reductase and alcohol dehydrogenase in a one‐pot enzyme cascade to nine desired γ‐butyrolactones 1, among them whisky (1 c) and cognac lactone (1 d). The products 1 were obtained in moderate to good yields and very good diastereoselectivities. Furthermore, the position of a nBu‐substituent was permutated to study the effect on the enzyme cascade
Solvatochromic Fluorescent 2‑Substituted 3‑Ethynyl Quinoxalines: Four-Component Synthesis, Photophysical Properties, and Electronic Structure

3-ethynylquinoxalines can be rapidly synthesized
in generally excellent yields by a consecutive four-component synthesis
starting from electron-rich π-nucleophiles, oxalyl chloride,
terminal alkynes, and 1,2-diaminoarenes. The title compounds are highly
fluorescent with a pronounced emission solvatochromism. The photophysical
properties and electronic structure were additionally corroborated
by computations on the DFT level of theory
