Morphological study of the iris musculature in diurnal and nocturnal raptors
In literature it is established that the iris musculature consists of striate muscle fibers in birds while in mammals it consists of smooth muscles. Some authors report the presence of smooth muscle tissue also in the iris of some species of birds. In the present study we report on the iris muscle tissues (type of tissue, direction and mean diameter of muscle fibers or cells) in five species of Accipitriformes (diurnal raptors) and four species of Strigiformes (nocturnal raptors) because they show different way of life depending of their predatory behavior. This morphological study was carried out from raptors died or euthanized at the Wild Life Rescue Centre of Sea and Water birds in Livorno (Italy). From histological examination of iris serial radial sections we find both striated and smooth musculature even if with marked differences among analyzed species, not directly correlated with diurnal or nocturnal lifestyle. Striated fibers are always present, mainly with cross direction, throughout the iris stroma, while the histological differences concern the smooth cells. Indeed, harrier and sparrow hawk (Accipitriformes) and great horned owl and little owl (Strigiformes) show a compact layer of cross smooth muscle cells throughout the iris stroma. In the other species analyzed smooth muscle cells are slightly detectable as scattered or not detectable. Since the cross smooth muscle tissue allows to maintain a myotic state for extended periods of time, our results might be correlated more to the predatory behavior than the taxonomic order
Impact of soluble tumor necrosis factor-related apoptosis-inducing ligand released by engineered adipose mesenchymal stromal cells on white blood cells
Background aims: The proapoptotic protein tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is physiologically expressed by immune cells and performs regulatory functions in infections, autoimmune diseases and cancer, where it acts as a tumor suppressor. Adipose-derived mesenchymal stromal cells (AD-MSCs) also may play immunomodulatory roles in both primary and acquired immune responses. We have previously demonstrated the efficacy of an anticancer gene therapy based on AD-MSC engineered to secrete a soluble TRAIL variant (sTRAIL) against pancreatic cancer. However, the impact of AD-MSC sTRAIL on leukocyte subsets has been not yet considered also to predict a possible immunotoxicity profile in the clinical translation of this cell-based anticancer strategy. Methods: Monocytes, polymorphonuclear cells and T lymphocytes were freshly isolated from the peripheral blood of healthy donors. Immunophenotype and functional (DR4 and DR5) and decoy (DcR1 and DcR2) TRAIL receptors were tested by flow cytometry. The viability of white blood cells treated with sTRAIL released by gene-modified AD-MSC or co-cultured with AD-MSC sTRAIL was then evaluated by both metabolic assays and flow cytometry. In addition, cytokine profile in co-cultures was analyzed by multiplex enzyme-linked immunosorbent assay. Results: Monocytes and polymorphonuclear cells showed high positivity for DR5 and DcR2, respectively, whereas T cells revealed negligible expression of all TRAIL receptors. Irrespective of TRAIL receptors' presence on the cell membrane, white blood cells were refractory to the proapoptotic effect displayed by sTRAIL secreted by gene-modified AD-MSC, and direct cell-to-cell contact with AD-MSC sTRAIL had negligible impact on T-cell and monocyte viability. Cytokine crosstalk involving interleukin 10, tumor necrosis factor alpha, and interferon gamma secreted by T lymphocytes and vascular endothelial growth factor A and interleukin 6 released by AD-MSC was highlighted in T-cell and AD-MSC sTRAIL co-cultures. The interactive effect of herbivory, nutrient enrichment and mucilage on shallow rocky macroalgal communities
This paper focuses on the interactive short and long-term effect of three different stressors on a macroalgal assemblage. Three stressors are considered: herbivory, nutrients and mucilage. The experiment was conducted in Tavolara Punta Coda Cavallo Marine Protected Area (Mediterranean Sea) during a bloom of the benthic mucilage-producing microalga Chrysophaeum taylorii (Pelagophyceae); this microalga is recently spreading in the Mediterranean Sea. On a rocky substratum, 36 plots 20 × 20 cm in size were prepared. Factorial combinations of three experimental treatments were applied in triplicate, including three grazing levels crossed with two nutrient enrichment and two mucilage removal treatments. Significant differences were observed among treatments 8 weeks later, at the end of summer. In particular, dark filamentous algae were more abundant in all enriched plots, especially where mucilage and macroalgae had been removed; a higher percent cover of crustose coralline algae was instead observed where nutrients had been increased and no grazing pressure acted. Furthermore, the abundance of Dictyota spp. and Laurencia spp. was significantly higher in enriched mucilage-free plots where the grazing pressure was null or low. However, the effects of the treatments on the overall assemblage of the macroalgal community were not long persistent (36 weeks later). These results illustrate the capacity of a shallow-water macroalgal community to quickly recover from the simultaneous impacts of herbivory, nutrient enrichment, and mucilage
The evaluation of the fibromyalgia patients.
Fibromyalgia (FM) is a rheumatic disease characterized by musculoskeletal pain, chronic diffuse tension and/or stiffness in joints and muscles, easy fatigue, sleep and emotional disturbances, and pressure pain sensitivity in at least 11 of 18 tender points. At present, there are no instrumental tests or specific diagnostic markers for FM; in fact, many of the existing indicators are significant for research purposes only. Many differential diagnoses may be excluded by an extensive clinical examination and patient history. Considering overlap of FM with other medical conditions, the treating physicians should be vigilant: chest-X-rays and abdominal ultrasonography are the first steps of general evaluation for all the patients with suspected FM. Functional neuroimaging methods have revealed a large number of supraspinal effects in FM, a disorder mediated by mechanisms that are essentially unknown. Many treatments are used in FM patients, but evaluating their therapeutic effects in FM is difficult because the syndrome is so multifaceted. An integrated assessment of the Good Environmental Status of Mediterranean Marine Protected Areas

targets to achieve Good Environmental Status (GES) of marine ecosystems by implementing the Marine Strategy
Framework Directive (MSFD) across member states. We combined an extensive dataset across five Mediterranean
ecoregions including 26 Marine Protected Areas (MPAs), their reference unprotected areas, and a no-trawl case
study. Our aim was to assess if MPAs reach GES, if their effects are local or can be detected at ecoregion level or
up to a Mediterranean scale, and which are the ecosystem components driving GES achievement. This was
undertaken by using the analytical tool NEAT (Nested Environmental status Assessment Tool), which allows an
integrated assessment of the status of marine systems. We adopted an ecosystem approach by integrating data from several ecosystem components: the seagrass Posidonia oceanica, macroalgae, sea urchins and fish. Thresholds to define the GES were set by dedicated workshops and literature review.
In the Western Mediterranean, most MPAs are in good/high status, with P. oceanica and fish driving this result
within MPAs. However, GES is achieved only at a local level, and the Mediterranean Sea, as a whole, results in a
moderate environmental status. Macroalgal forests are overall in bad condition, confirming their status at risk.
The results are significantly affected by the assumption that discrete observations over small spatial scales are
representative of the total extension investigated. This calls for large-scale, dedicated assessments to realistically
detect environmental status changes under different conditions.
Understanding MPAs effectiveness in reaching GES is crucial to assess their role as sentinel observatories of
marine systems. MPAs and trawling bans can locally contribute to the attainment of GES and to the fulfillment of
the MSFD objectives. Building confidence in setting thresholds between GES and non-GES, investing in long-term
monitoring, increasing the spatial extent of sampling areas, rethinking and broadening the scope of complementary tools of protection (e.g., Natura 2000 Sites), are indicated as solutions to ameliorate the status of the
