    Sex differences in self-report anxiety and sleep quality during COVID-19 stay-at-home orders

    Background: COVID-19 and home isolation has impacted quality of life, but the perceived impact on anxiety and sleep remains equivocal. The purpose of this study was to assess the impact of COVID-19 and stay-at-home orders on self-report anxiety and sleep quality, with a focus on sex differences. We hypothesized that the COVID-19 pandemic would be associated with increased anxiety and decreased sleep quality, with stronger associations in women. Methods: One hundred three participants (61 female, 38 ± 1 years) reported perceived changes in anxiety and sleep quality due to stay-at-home orders during the COVID-19 pandemic and were administered the Spielberger State-Trait Anxiety Inventory (STAI), Pittsburgh Sleep Quality Index (PSQI), and Insomnia Severity Index (ISI). Chi-square and T test analyses were utilized to assess sex differences in reported anxiety and sleep. Analysis of covariance was used to compare the associations between reported impact of COVID-19 and anxiety/sleep parameters. Results: Women (80.3%) reported higher prevalence of increased general anxiety due to COVID-19 when compared to men (50%; p = 0.001) and elevated STAI state anxiety compared to men (43 ± 1 vs. 38 ± 1 a.u., p = 0.007). Despite these differences in anxiety, the perceived impact of COVID-19 on PSQI was not different between sexes. However, when stratified by perceived changes in anxiety due to COVID-19, participants with higher anxiety responses to COVID-19 had higher ISI compared to those with no perceived changes in anxiety (9 ± 1 vs. 5 ± 1 a.u., p = 0.003). Additionally, participants who reported reduced sleep quality due to COVID-19 reported higher state anxiety (45 ± 1 a.u.) compared to those that perceived no change (36 ± 2 a.u., p = 0.002) or increased (36 ± 2 a.u., p \u3c 0.001) sleep quality. Conclusion: COVID-19 and state-ordered home isolation was associated with higher anxiety and reduced sleep quality, with a stronger association in women with respect to anxiety

    Adenomatous Polyposis Coli is Present Near the Minimal Level Required for Accurate Graded Responses to the Wingless Morphogen

    The mechanisms by which the Wingless (Wg) morphogen modulates the activity of the transcriptional activator Armadillo (Arm) to elicit precise, concentration-dependent cellular responses remain uncertain. Arm is targeted for proteolysis by the Axin/Adenomatous polyposis coli (Apc1 and Apc2)/Zeste-white 3 destruction complex, and Wg-dependent inactivation of destruction complex activity is crucial to trigger Arm signaling. In the prevailing model for Wg transduction, only Axin levels limit destruction complex activity, whereas Apc is present in vast excess. To test this model, we reduced Apc activity to different degrees, and analyzed the effects on three concentration-dependent responses to Arm signaling that specify distinct retinal photoreceptor fates. We find that both Apc1 and Apc2 negatively regulate Arm activity in photoreceptors, but that the relative contribution of Apc1 is much greater than that of Apc2. Unexpectedly, a less than twofold reduction in total Apc activity, achieved by loss of Apc2, decreases the effective threshold at which Wg elicits a cellular response, thereby resulting in ectopic responses that are spatially restricted to regions with low Wg concentration. We conclude that Apc activity is not present in vast excess, but instead is near the minimal level required for accurate graded responses to the Wg morphogen

    Preliminary evidence of increased striatal dopamine in a nonhuman primate model of maternal immune activation.

    Women exposed to a variety of viral and bacterial infections during pregnancy have an increased risk of giving birth to a child with autism, schizophrenia or other neurodevelopmental disorders. Preclinical maternal immune activation (MIA) models are powerful translational tools to investigate mechanisms underlying epidemiological links between infection during pregnancy and offspring neurodevelopmental disorders. Our previous studies documenting the emergence of aberrant behavior in rhesus monkey offspring born to MIA-treated dams extends the rodent MIA model into a species more closely related to humans. Here we present novel neuroimaging data from these animals to further explore the translational potential of the nonhuman primate MIA model. Nine male MIA-treated offspring and 4 controls from our original cohort underwent in vivo positron emission tomography (PET) scanning at approximately 3.5-years of age using [18F] fluoro-l-m-tyrosine (FMT) to measure presynaptic dopamine levels in the striatum, which are consistently elevated in individuals with schizophrenia. Analysis of [18F]FMT signal in the striatum of these nonhuman primates showed that MIA animals had significantly higher [18F]FMT index of influx compared to control animals. In spite of the modest sample size, this group difference reflects a large effect size (Cohen's d = 0.998). Nonhuman primates born to MIA-treated dams exhibited increased striatal dopamine in late adolescence-a hallmark molecular biomarker of schizophrenia. These results validate the MIA model in a species more closely related to humans and open up new avenues for understanding the neurodevelopmental biology of schizophrenia and other neurodevelopmental disorders associated with prenatal immune challenge

    On lunar exospheric column densities and solar wind access beyond the terminator from ROSAT soft X-ray observations of solar wind charge exchange

    We analyze the Röntgen satellite (ROSAT) position sensitive proportional counter soft X-ray image of the Moon taken on 29 June 1990 by examining the radial profile of the surface brightness in three wedges: two 19° wedges (one north and one south) 13–32° off the terminator toward the dark side and one wedge 38° wide centered on the antisolar direction. The radial profiles of both the north and the south wedges show significant limb brightening that is absent in the 38° wide antisolar wedge. An analysis of the soft X-ray intensity increase associated with the limb brightening shows that its magnitude is consistent with that expected due to solar wind charge exchange (SWCX) with the tenuous lunar atmosphere based on lunar exospheric models and hybrid simulation results of solar wind access beyond the terminator. Soft X-ray imaging thus can independently infer the total lunar limb column density including all species, a property that before now has not been measured, and provide a large-scale picture of the solar wind-lunar interaction. Because the SWCX signal appears to be dominated by exospheric species arising from solar wind implantation, this technique can also determine how the exosphere varies with solar wind conditions. Now, along with Mars, Venus, and Earth, the Moon represents another solar system body at which SWCX has been observed

First published January 31, 2003 10.1152/japplphysiol. 01109.2002The effects of resistance training on arterial blood pressure and muscle sympathetic nerve activity (MSNA) at rest have not been established. Although endurance training is commonly recommended to lower arterial blood pressure, it is not known whether similar adaptations occur with resistance training. Therefore, we tested the hypothesis that whole body resistance training reduces arterial blood pressure at rest, with concomitant reductions in MSNA. Twelve young [21 Ϯ 0.3 (SE) yr] subjects underwent a program of whole body resistance training 3 days/wk for 8 wk. Resting arterial blood pressure (n ϭ 12; automated sphygmomanometer) and MSNA (n ϭ 8; peroneal nerve microneurography) were measured during a 5-min period of supine rest before and after exercise training. Thirteen additional young (21 Ϯ 0.8 yr) subjects served as controls. Resistance training significantly increased one-repetition maximum values in all trained muscle groups (P Ͻ 0.001), and it significantly decreased systolic (130 Ϯ 3 to 121 Ϯ 2 mmHg; P ϭ 0.01), diastolic (69 Ϯ 3 to 61 Ϯ 2 mmHg; P ϭ 0.04), and mean (89 Ϯ 2 to 81 Ϯ 2 mmHg; P ϭ 0.01) arterial blood pressures at rest. Resistance training did not affect MSNA or heart rate. Arterial blood pressures and MSNA were unchanged, but heart rate increased after 8 wk of relative inactivity for subjects in the control group (61 Ϯ 2 to 67 Ϯ 3 beats/min; P ϭ 0.01). These results indicate that whole body resistance exercise training might decrease the risk for development of cardiovascular disease by lowering arterial blood pressure but that reductions of pressure are not coupled to resistance exercise-induced decreases of sympathetic tone

    Statins enhance the efficacy of HER2-targeting radioligand therapy in drug-resistant gastric cancers

    Human epidermal growth factor receptor 2 (HER2) is overexpressed in various cancer types. HER2-targeting trastuzumab plus chemotherapy is used as first-line therapy for HER2-positive recurrent or primary metastatic gastric cancer, but intrinsic and acquired trastuzumab resistance inevitably develop over time. To overcome gastric cancer resistance to HER2-targeted therapies, we have conjugated trastuzumab with a beta-emitting therapeutic isotope, lutetium-177, to deliver radiation locally to gastric tumors with minimal toxicity. Because trastuzumab-based targeted radioligand therapy (RLT) requires only the extramembrane domain binding of membrane-bound HER2 receptors, HER2-targeting RLT can bypass any resistance mechanisms that occur downstream of HER2 binding.     Musculoskeletal Outcomes From Chronic High-Speed, High-Impulse Resistance Exercise

    While bones and muscles adapt to mechanical loading, it appears that very specific types of stimuli must be applied to achieve osteogenesis. Our study assessed musculoskeletal outcomes to 30 training sessions on an Inertial Exercise Trainer (Newnan, GA). Subjects (n = 13) performed workouts with their left leg, while their right served as an untreated control. Work-outs entailed three 60-s sets each of knee extension, hip extension and calf press exercises, separated by 90-s rests. Before and after the 30 training sessions, subjects underwent strength tests (knee and ankle extensors of both legs), DEXA scans (hip, knee and ankles of both legs), and blood draws. After 30 training sessions 2 × 2 ANOVAs showed left leg peak torques rose significantly. 2 × 2 ANCOVAs, with bone scan area as a covariate, showed significant left leg calcaneal bone mineral content ( + 29 %) and density ( + 33 %) increases after 30 training sessions. A significant decline in C-terminal telopeptides of type I collagen, a blood marker of bone resorption, also occurred after 30 training sessions. The Inertial Exercise Trainer’s large volume of training session repetitions elicited high peak force, peak acceleration and impulses that likely provided a mechanical loading stimulus that evoked calcaneal accretion

    TERMS Photometry of Known Transiting Exoplanets

    The Transit Ephemeris Refinement and Monitoring Survey (TERMS) conducts radial velocity and photometric monitoring of known exoplanets in order to refine planetary orbits and predictions of possible transit times. This effort is primarily directed towards planets not known to transit, but a small sample of our targets consist of known transiting systems. Here we present precision photometry for 6 WASP planets acquired during their transit windows. We perform a Markov Chain Monte Carlo (MCMC) analysis for each planet and combine these data with previous measurements to redetermine the period and ephemerides for these planets. These observations provide recent mid-transit times which are useful for scheduling future observations. Our results improve the ephemerides of WASP-4b, WASP-5b and WASP-6b and reduce the uncertainties on the mid-transit time for WASP-29b.
