Analysis of β-globin chromatin micro-environment using a novel 3C variant, 4Cv
Copyright: Š 2010 Pink et al.Higher order chromatin folding is critical to a number of developmental processes, including the regulation of gene expression. Recently developed biochemical techniques such as RNA TRAP and chromosome conformation capture (3C) have provided us with the tools to probe chromosomal structures. These techniques have been applied to the β-globin locus, revealing a complex pattern of interactions with regions along the chromosome that the gene resides on. However, biochemical and microscopy data on the nature of β-globin interactions with other chromosomes is contradictory. Therefore we developed a novel 4C variant, Complete-genome 3C by vectorette amplification (4Cv), which allows an unbiased and quantitative method to examine chromosomal structure. We have used 4Cv to study the microenvironment of the β-globin locus in mice and show that a significant proportion of the interactions of β-globin are inter-chromosomal. Furthermore, our data show that in the liver, where the gene is active, β-globin is more likely to interact with other chromosomes, compared to the brain where the gene is silent and is more likely to interact with other regions along the same chromosome. Our data suggest that transcriptional activation of the β-globin locus leads to a change in nuclear position relative to the chromosome territory.Ryan Pink is supported by a grant from Action Medical Research; Daniel Caley is supported by a grant from The Dunhill Medical Trust; David Carter is supported by a grant from the British Society for Haematology
Vibrational Stability of NLC Linac and Final Focus Components
Vertical vibration of linac components (accelerating structures, girders and
quadrupoles) in the NLC has been studied experimentally and analytically.
Effects such as structural resonances and vibration caused by cooling water
both in accelerating structures and quadrupoles have been considered.
Experimental data has been compared with analytical predictions and simulations
using ANSYS. A design, incorporating the proper decoupling of structure
vibrations from the linac quadrupoles, is being pursued.Comment: 3 pages, 8 figures presented at the LINAC 2002 conference, Gyeongju

Modeling and Analysis of Power Processing Systems (MAPPS). Volume 2: Appendices
The computer programs and derivations generated in support of the modeling and design optimization program are presented. Programs for the buck regulator, boost regulator, and buck-boost regulator are described. The computer program for the design optimization calculations is presented. Constraints for the boost and buck-boost converter were derived. Derivations of state-space equations and transfer functions are presented. Computer lists for the converters are presented, and the input parameters justified
Modeling and Analysis of Power Processing Systems (MAPPS). Volume 1: Technical report
Computer aided design and analysis techniques were applied to power processing equipment. Topics covered include: (1) discrete time domain analysis of switching regulators for performance analysis; (2) design optimization of power converters using augmented Lagrangian penalty function technique; (3) investigation of current-injected multiloop controlled switching regulators; and (4) application of optimization for Navy VSTOL energy power system. The generation of the mathematical models and the development and application of computer aided design techniques to solve the different mathematical models are discussed. Recommendations are made for future work that would enhance the application of the computer aided design techniques for power processing systems
Separability of Black Holes in String Theory
We analyze the origin of separability for rotating black holes in string
theory, considering both massless and massive geodesic equations as well as the
corresponding wave equations. We construct a conformal Killing-Stackel tensor
for a general class of black holes with four independent charges, then identify
two-charge configurations where enhancement to an exact Killing-Stackel tensor
is possible. We show that further enhancement to a conserved Killing-Yano
tensor is possible only for the special case of Kerr-Newman black holes. We
construct natural null congruences for all these black holes and use the
results to show that only the Kerr-Newman black holes are algebraically special
in the sense of Petrov. Modifying the asymptotic behavior by the subtraction
procedure that induces an exact SL(2)^2 also preserves only the conformal
Killing-Stackel tensor. Similarly, we find that a rotating Kaluza-Klein black
hole possesses a conformal Killing-Stackel tensor but has no further
enhancements.Comment: 27 page
Finite-temperature Mott transitions in multi-orbital Hubbard model
We investigate the Mott transitions in the multi-orbital Hubbard model at
half-filling by means of the self-energy functional approach. The phase
diagrams are obtained at finite temperatures for the Hubbard model with up to
four-fold degenerate bands. We discuss how the first-order Mott transition
points and as well as the critical temperature depend
on the orbital degeneracy. It is elucidated that enhanced orbital fluctuations
play a key role to control the Mott transitions in the multi-orbital Hubbard
model.Comment: 8 pages, 7 figure
