The Use of the grassmeter as a simplified method to estimate dry matter yield on annual self-reseeding medics and clovers
BACKGROUND. The grassmeter non-destructive
method was used to estimate dry matter yield (DMY)
of 24 annual self-reseeding legumes. The possibility
was evaluated of using the grassmeter as an alternative
to conventional destructive quadrats for large
field plot experiments.
METHODS. The linear regressions between sward
height measured by the grassmeter and DMY were
analysed. Three types of regression equations were
calculated: (i) ‘general’, a single equation including
the three-year set of height/DMY pairs of data; (ii)
‘specific’, one regression equation for each species;
(iii) ‘preliminary’, calculated from the data collected
in the spring and autumn cuts of the first year.
RESULTS. The ‘general’ calibration gave a satisfactory
estimate of DMY (R2=0.55). The ‘specific’ calibration
was particularly accurate for Trifolium
brachycalicynum “Osilo” (R2=0.88), T. squarrosum
“Chilivani” (R2=0.81) and Medicago polymorpha
“Circle Valley” (R2=0.81), but not for M. rugosa
“Sapo” (R2=0.22) and “Paraponto” (R2=0.26). The
‘preliminary’ calibration, which was based only on the
destructive assessment of DMY at the first cut, provided
reliable estimates of DMY until the sward conditions,
namely cover rate and weeds, were comparable
to those of the calibration period. The coefficient
of variation of the DMY estimate based on the
grassmeter ‘general’ calibration was about 10% lower
than that of the DMY measured with conventional
quadrats.
CONCLUSIONS. Results suggest that the grassmeter
could effectively replace destructive measurements
for estimating the DMY of annual self-reseeding
legumes, thus reducing labour requirements and
number of samples to be processed, or increasing,
when needed, the number of accessions to compare
and hence the inference of the experiment
Mediche e trifogli annuali autoriseminanti per usi foraggeri e non convenzionali: I. Adattamento e persistenza
The objective of this research was to test the adaptation and persistence of 24 pasture legume accessions, chosen
among Australian varieties, Italian commerciaI materials and Sardinian ecotypes, in view of forage and not conventional
uses. During three years, the evolution of sward height, the specific soil covering rate and the percentage of weeds were
estimated. Edaphic and clima tic factors selected the commerciaI materials, that yielded high dry matter productions only at the
first year.
The medics established well and reached high productive levels in the first year, but among them only Medicago arabica
"IAS", M. polymorpha cv. "Circle Valley" and M. truncatula cv. "Paraggio" persisted during the three-years. The early
flowering and the high dry matter yield indicate that M. truncatula cv. "Paraggio" is suitable to be integrated into rotational
cereal systems and with M. polymorpha cv. "Circle Valley" could be used as a cover crop in vineyards or in mixtures with other
legumes at different flowering earliness to improve the pasture composition. M. arabica "IAS", due to its long cycle and
high productivity, may be only used as a forage crop. Trifolium brachycalycinum "Funtana Bona", with its high competitive
ability against weeds, its favourable seasonal production pattern, could be used for artificial or natural pasture improvement.
T. yanninicum cv. "Larissa", with its shorter cycle and Iow produetivity, proved to be suitable as a cover crop in the vineyards.
T. squarrosum "Chilìvani", a late clover, can be considered as an alternative to annual medi es and subterranean clovers
for pasture improvement on sub-alcaline soils.
The results indicate once more that local ecotypes were on average more persistent and adapted than commercial varieties.
Obiettivo della ricerca è stato quello di valutare la capacità di adattamento e la persistenza di 24 accessioni di leguminose
annuali scelte fra varietà australiane, materiali commerciali nazionali e collezioni sarde, in vista di possibili impieghi produttivi ed
extra produttivi. Per un triennio, su terreni subalcalini della Sardegna nord occidentale, sono state valutate la produzione
e l'evoluzione del ricoprimento specifico del terreno, dell'infestazione e dell'altezza della copertura vegetale.
I risultati hanno evidenziato una marcata selettività dei fattori pedoclimatici soprattutto nei confronti della persistenza
dei materiali commerciali. Le mediche hanno raggiunto buoni livelli produttivi al primo anno, ma soltanto Medicago
arabica "IAS", M. polymorpha cv. "Cirde Valley" e M. truncatula cv. "Paraggio" hanno mostrato buona persistenza.
La precocità e la elevata produzione di M. truncatula cv. "Paraggio", ne suggeriscono l'impiego in rotazione con cereali
autunno vernini, insieme a M. polymorpha cv. "Circle Valley" come "cover crop" in vigneti o in miscugli oligofiti con componenti
a diversa precocità per la costituzione di pascoli artificiali. M. arabica "IAS", per il ciclo lungo e la elevata produzione è risultata
invece più indicata per destinazioni strettamente foraggere. Trifolium brachycalycinum "Funtana Bona", in virt๠della
competitività con le specie spontanee, la regolarità di produzione e la buona distribuzione stagionale, sembrerebbe adatto
per la costituzione di pascoli artificiali o l'infittimento di pascoli naturali; T. yanninicum cv. "Larissa" invece, per il suo
ciclo più breve e la minore produttività, è apparso più adatto per l'inerbimento dei vigneti. T. squarrosum "Chilivani", per la
lunghezza del ciclo produttivo, si è rivelato una valida alternativa alle mediche annuali ed ai trifogli sotterranei nel miglioramento
dei pascoli su terreni sub-alcalini.
I risultati indicano una generale superiorità dei materiali locali rispetto alla gran parte dei materiali commerciali

The results of a three year trial run in Sardinia within a larger experiment on rainfed forage systems for dairy sheep are
reported. The agronomic, zootechnic and economic validity of an improved pasture (sod seeding of Lolium rigidum ecotype
«Nurra» and Trifolium brachycalycinum «Clare») as an alternative to the grazing of short term forage crops (oats) were
evaluated. The forage production and consumption of the improved pasture were higher than those of the oats in two years out
of three and on average in the three years. The seasonal distribution of the forage production was better in the improved
pasture for the quicker establishment in Autumn. The sowing date of the oats was delayed in the second year because of the
abundant and frequent rainfall in the early Autumn. The subclover showed a slow establishment and was less competitive than
annual ryegrass in the first year. Its self-reseeding was affected by the damage of Bruchidius trifolii. Annual ryegrass gradually
established a thick and persistent sward through a seed yield over than 50000 seeds m-2. The milk yield of ewes grazing the
improved pasture resulted 20% higher than that of the ewes grazing oats. The average annual costs ofthe oats were about two
times higher and the agronomie and zootechnic results lower than those of the improved pasture. Taking into account the
environmental aspects (soil protection from erosion in the Autumn) and the management aspects, it was concluded that the
self-reseeding improved pasture is an effective alternative to short term forage crops in the dairy sheep forage farming systems
of the Mediterranean environment.
Vengono riportati i risultati di una prova triennale svolta in Sardegna nell'ambito di un confronto tra sistemi foraggeri
asciutti per ovini da latte. E’ stata valutata la validità in termini agronomici, zootecnici ed economici di un pascolo migliorato con
la semina su sodo di un miscuglio di Lolium rigidum ecotipo «Nurra» e Trifolium brachycalycinum «Clare» come possibile soluzione
alternativa al pascolamento degli erbai di cereali autunno vernini. La disponibilità foraggera e i consumi alimentari rilevati
sul pascolo migliorato sono risultati superiori a quelli dell'erbaio in due annate su tre e nella media del triennio. La distribuzione
stagionale della produzione è risultata più favorevole nel pascolo migliorato per il più rapido reinsediamento in autunno. L'epoca
di semina dell'erbaio nella seconda annata è stata ritardata per le abbondanti e frequenti precipitazioni autunnali che hanno
impedito la preparazione del letto di semina. Il trifoglio brachicalicino ha mostrato un più lento insediamento e minore
competitività rispetto al loglio rigido, e la risemina naturale è stata fortemente limitata dall'attacco di un coleottero (Bruchidius
trifolii). Il loglio rigido ha invece progressivamente costituito un cotico fitto e persistente, attraverso produzioni di seme sempre
superiori a 50000 semi m-2. Le produzioni di latte ottenute sul pascolo migliorato sono risultate mediamente superiori del 20%
rispetto a quelle ottenute con il pascolamento dell'avena. I costi colturali medi annui dell’erbaio sono risultati circa doppi rispetto
a quelli del pascolo migliorato, e i risultati agronomici e zootecnici nettamente inferiori. E’ stato concluso che il pascolo migliorato
con autori seminanti costituisce un'efficace alternativa al pascolamento degli erbai di cereali nei sistemi foraggeri per ovini da
latte in ambiente mediterraneo, in linea con l'esigenza di tutela ambientale (protezione del suolo dall'erosione in autunno)
Interactome-Seq: A Protocol for Domainome Library Construction, Validation and Selection by Phage Display and Next Generation Sequencing
Folding reporters are proteins with easily identifiable phenotypes, such as antibiotic resistance, whose folding and function is compromised when
fused to poorly folding proteins or random open reading frames. We have developed a strategy where, by using TEM-1 \u3b2-lactamase (the enzyme
conferring ampicillin resistance) on a genomic scale, we can select collections of correctly folded protein domains from the coding portion of the
DNA of any intronless genome. The protein fragments obtained by this approach, the so called "domainome", will be well expressed and soluble,
making them suitable for structural/functional studies.
By cloning and displaying the "domainome" directly in a phage display system, we have showed that it is possible to select specific protein
domains with the desired binding properties (e.g., to other proteins or to antibodies), thus providing essential experimental information for gene
annotation or antigen identification.
The identification of the most enriched clones in a selected polyclonal population can be achieved by using novel next-generation sequencing
technologies (NGS). For these reasons, we introduce deep sequencing analysis of the library itself and the selection outputs to provide complete
information on diversity, abundance and precise mapping of each of the selected fragment. The protocols presented here show the key steps for
library construction, characterization, and validation
Effetti di tipologie di suolo e colture foraggere sulle perdite per ruscellamento di azoto, fosforo e potassio in differenti areali italiani
Le colture foraggere svolgono un ruolo importante in molti areali italiani. Per una corretta pianificazione del territorio agricolo è necessario approfondire la conoscenza non solo delle caratteristiche produttive di queste colture, ma anche dei loro rapporti con l'ambiente.
Questo lavoro prende in considerazione le perdite di azoto, fosforo e potassio dovute al ruscellamento in colture foraggere a diverso livello di intensificazione (dal pascolo alla coltura di erba medica, dal mais al doppio ciclo di mais e loiessa) in tre ambienti italiani: la Pianura Padana nord-occidentale, l'Appennino Toscano e la pianura sarda, con suoli varianti da franco-sabbioso a franco-argilloso.
Il monitoraggio quanti-qualitativo del ruscellamento è stato fatto per periodi variabili da due a sei anni. La pendenza era dello 0,5% per gli appezzamenti in Piemonte e Sardegna e del 10% in Toscana.
Per quanto riguarda l'azoto i rilasci sono risultati più bassi nei terreni più pianeggianti, anche per i ridotti volumi di ruscellamento registrati, non superando mai 15 kg di N ha -1 anno -1. Nei terreni in pendenza si sono invece registrati valori più elevati, con un massimo annuale di circa 30 kg ha -1 anno -1 di azoto, in relazione anche all'elevato ruscellamento ed erosione di un evento eccezionale. Per il fosforo solo in pochi casi si sono raggiunte perdite di 5 kg ha -1 anno -1, mentre nella maggior parte dei casi non sono stati superati 2 kg ha -1 anno -1. In Sardegna i rilasci di tale elemento sono da considerarsi pressoché trascurabili. Le perdite di potassio sono risultate minime in Sardegna e massime in Piemonte, dove si sono registrati valori dell' ordine di 10 kg ha -1 anno -1.
Ove era possibile il confronto, si è verificato che le colture prative riducono il rilascio di elementi nutritivi rispetto alla coltura del mais e che la qualità delle acque di superficie appare legata piuttosto alle tecniche colturali che alla tipologia di suolo.
Fodder crops play an important role in many Italian environments. The knowledge of the main productive characteristics of these crops is as important as their relationships with the environment, expecially for a proper territorial management.
This paper compares nitrogen, phosphorous and potassium contents in runoff of some forage crops of different intensity (pasture, lucerne, silage maize, Italian ryegrass/maize double cropping) in different ltalian environments (north-western plain of Piemonte, Apennines hills of Tuscany and Mediterranean plain of Sardegna) on different typology of soils.
Runoff data have been collected for periods ranging between two and six year, from plots of different extension and slope (0,5% in the plains, 10% in Apennine hills).
Nitrogen losses, for the small amount of runoff, have been qui te low from ilat fields, being always less than 15 kg ha -1 year-1. Losses from slope fields have been higher, with a maximum of 30 kg ha -1 year-1, due to very high level of runoff and erosion in a conspicuous episode. Phosphorous losses only in a few cases were higher than 5 kg ha -I year -I, while mostly they have b>!en less than 2 kg ha -1 year-1.
In the Mediterranean plain such losses have been quite insignificant. Minimum potassium 10sses were recorded in the Mediterranean plain, while in north-western plain they reached about 10 kg-1 year-1.
Maize was, on average, the crop with highest nutrient losses, while quality of the runoff water was more related to agricultural practices than to soil types
Characterization and Comparison of Ocular Surface Microbiome in Newborns
The ocular microbiome is of fundamental importance for immune eye homeostasis, and its alteration would lead to an impairment of ocular functionality. Little evidence is reported on the composition of the ocular microbiota of term infants and on the impact of antibiotic prophylaxis. Methods: A total of 20 conjunctival swabs were collected from newborns at birth and after antibiotic treatment. Samples were subjected to 16S rRNA sequencing via system MiSeq Illumina. The data were processed with the MicrobAT software and statistical analysis were performed using two-way ANOVA. Results: Antibiotic prophylaxis with gentamicin altered the composition of the microbiota. In detail, a 1.5- and 2.01-fold reduction was recorded for Cutibacterium acnes (C. acnes) and Massilia timonae (M. timonae), respectively, whereas an increase in Staphylococcus spp. of 6.5 times occurred after antibiotic exposure. Conclusions: Antibiotic prophylaxis altered the ocular microbiota whose understanding could avoid adverse effects on eye health
Modellizzazione della lisciviazione dei nitrati: calibrazione e validazione del modello LEACHN in diversi suoli e colture foraggere
The LEACHN model calibration and validation was realized in three Italian enviroments (Piedmont, Tuscany and
Sardinia), using nitrate losses by drainage from 4 soil types ranging from sandy-loam to day-loam. A set of large drainage
lysimeters were used to obtain leaching data. In each location one lysimeter was cultivated with lucerne, while the others
were cropped with widespread crops in the area (maize for silage in Piedmont and Sardinia and permanent pasture in
Tuscany). Measured leaching losses ranged from l to 68 kg ha-1 year-1. The calibration parameters were humus mineralization rate and nitrification rate. The calibration was realized on a not nitrogen fixing crop, while the validation was
executed on lucerne, even if there is not a specific N-fixation subroutine in LEACHN. After calibration, the prediction of
nitrate losses cumulated over the whole period (two years) or over each month resulted acceptable for all crops, while the single
event prediction was totally inadequate. The LEACHN model seems to be applicable for the nitrate leaching prediction on
a territorial scale.
In tre areali italiani (pianura piemontese, conca interappenninica toscana, pianura irrigua sarda) e su quattro diversi tipi
di terreno si è calibrato e validato il modello LEACHN relativamente alla previsione della lisciviazione dei nitrati. Si è operato
con lisimetri a drenaggio di grandi dimensioni. In tutte le situazioni un lisimetro era coltivato a erba medica, mentre altri con
colture di ampia diffusione nell'arcale: mais da trinciato integrale in Piemonte, doppio ciclo colturale di loiessa e mais da
granella in Sardegna e pascolo in Toscana e Sardegna. Le perdite per lisciviazione misurate sono risultate comprese tra l e 68
kg ha-1 anno-1 di NO-3-N. Come parametri di calibrazione si sono usati il tasso di mineralizzazione dell'humus e il tasso di nitrificazione dello ione ammonio, calibrati sulla coltura non azotofissatrice. Sull'erba medica, per la quale non è prevista alcuna routine per l'azotofissazione, è stata invece condotta la validazione. Dopo la calibrazione, la simulazione della lisciviazione cumulata e dei totali mensili lisciviati è risultata accettatile, anche su erba medica, mentre quella dei singoli eventi è stata del tutto insufficiente. Il modello nel complesso è apparso applicabile per la predizione delle perdite di nitrati anche su scala territoriale
Caratterizzazione della dinamica produttiva di pascoli naturali italiani
This work studies herbage production and its seasonal distribution in indigenous pastures, and analyses the relationship
between the environmental factors (soil, clima, vegetation) and the productivity of these resources.
The investigations have been carried on during the period 1983-90 by the joint activity of 10 different University Istitutions
in 23 different environments distributed along the Italian peninsula and the main islands.
For each environment, pasture production has been measured with the Corrall and Fenlon method, analysing the more
important vegetational and ecological conditions; altogether the total yearly production and the seasonal pattern of herbage
production have been detected on 104 pastures.
The total herbage yield is not significantly influenced by the latitudinal gradient, and the overall regional (alps, central
Appenine, south Apennine and islands) production is about 2.3 t ha-1 year-1 The wide range (0.5-6.3 t ha-1 year-1) of
herbage production, on small or medium scale, seems to be due to evident changeof environmental or management factors.
Five types of seasonal distribution of herbage growth are evidenced with multivariate analysis methods, based on the
growing season and the amplitude of the growth. With mean temperature above 12°C and total rainfall below 800 mm,
herbage distribution shows a standstill during summer period and an evident regrowth in autumn. On the contrary, for
the 4 other distribution types, the winter standstill become important, and the types are distinct by summer growth amplitude
and by the growing season lenght. With cluster analysis method, for each type of herbage distribution, have been
pointed out under-types characterized by interannual herbage production variation.
Among the environmental factors, vegetation characheristics, expressed as Pasture Value following Daget and Poissonet
seems to be strictly correlated with total production. The comparative poor role played by the soil and climatic factor,
may be due to the strong past and present antropic influence, related with management and utilization techniques.
Il presente lavoro ha come scopo l'approfondimento delle conoscenze sulla produzione e sulla distribuzione stagionale
della crescita dell'erba dei pascoli naturali, nonché l'analisi delle interazioni tra i fattori ambientali, pedo-climatici e vegetazionali,
e la risposta produttiva di queste risorse.
La ricerca è stata condotta nel periodo 1983-90 da 10 diverse Istituzioni Universitarie, in 23 ambienti differenti, distribuiti
lungo tutta la penisola e le isole maggiori.
Per ogni ambiente, con il metodo di rilievo di Corrall e Fenlon, è stata saggiata la risposta produttiva di pascoli rappresentativi
delle principali situazioni vegetazionali e di giacitura; complessivamente sono state rilevate la produzione totale
annua e la curva di produttività media pluriennale di 104 pascoli.
Riguardo la produzione annua complessiva si è osservato che essa non presenta variazioni significative lungo il gradiente
latitudinale, collocandosi tra le diverse regioni (alpina, centro appenninica, suq, appenninica e insulare) attorno a 2.3
t ha-1 anno-1. La fitomassa raccolta è soggetta invece a variazioni sensibili (0.5-6.3 t ha-1 anno-1) riconducibili a fattori
ambientali e gestionali che si esprimono su piccola e media scala.
Con metodi di analisi multivariata si sono individuate 5 tipologie distributive della crescita dell'erba, in rapporto alla
stagione vegetativa e alle variazioni dell'intensità di crescita nel corso della stagione stessa. Con temperature medie e precipitazioni
annue rispettivamente maggiori di 12°C e minori di 800 mm, risulta evidente la stasi vegetativa nel trimestre
estivo e la ripresa vegetativa autunnale. Nel caso opposto la stasi è invernale e le 4 tipologie afferenti a questo modello,
sono distinguibili dall'entità della crescita nei mesi estivi e dalla durata della stagione vegetativa. Per ogni tipologia produttiva,
sono state evidenziate, tramite l'analisi cluster, sotto-tipologie distinte per la variabilità produttiva interannuale.
Tra i fattori ambientali, la vegetazione, espressa attraverso l'indice del valore pastorale di Daget e Poissonet, presenta
una buona capacità predittiva nei confronti del livello produttivo dei pascoli. Il contributo comparativamente modesto offerto
dai fattori pedoclimatici sembra attribuibile alla forte influenza antropica, pregressa e attuale, attraverso le cure colturali
e l'utilizzazione
Gestione e miglioramento di pascoli italiani
Experimental results of trials on pastures management and improvement, conducted by 10 University Institutes within
the study team «Characterization of italian pastures» are reported.
The trials carried out in different soil and climate conditions, ranging from the Alps to the southern Appennines and
to the Islands, in different years, provide some conclusive and generaI considerations.
With periodical cuts, the forage availability period becomes longer, while using one cut at flowering yield results are higher.
As far as management techniques are concerned, fertilization with the three main nutrients affects positively quanti-qualitative yield, although in real situations the application of one or two elements may be sufficient and more economically
convenient, and in relation to the botanical composition.
Undersowing determines positive effects both on botanical composition and on pasture utilization. Positive results, furthermore, are obtained by mechanical scrub clearing.
Vengono riferiti i risultati sperimentali delle prove sulla gestione e sul miglioramento dei pascoli condotte da dieci Istituti
Universitari costituenti il gruppo di studio «Caratterizzazione di pascoli italiani», Dalle prove condotte in ambienti pedoclimatici
diversi, dalle alpi all'appennino meridionale ed alle isole, e in annate differenti, sono emerse come generalizzabili
alcune considerazioni conclusive.
Con sfalci periodici si allunga il periodo di disponibilità di foraggio verde, mentre con quello unico alla fioritura la
produzione risulta più elevata.
Per gli interventi tecnici, la concimazione con i tre macroelementi influisce favorevolmente sull'entità e sulla qualità
della produzione, anche se, nelle situazioni concrete, può essere sufficiente, e più vantaggioso dal lato economico, la somministrazione di uno o due elementi, anche in relazione alla facies floristica.
La trasemina determina effetti favorevoli sia sulla composizione floristica sia nella migliore utilizzazione del pascolo.
Risultati positivi, infine, si conseguono con il decespugliamento meccanico
