Inhibitory Effect of Helicteres gardneriana Ethanol Extract on Acute Inflammation
The anti-inflammatory effect of an ethanol extract of Helicteres gardneriana (Nees) Castiglioni was assayed in experimental models of pleurisy and microcirculation in situ. Treatment of animals with 500 mg/kg body weight reduced the exudate volume (35% reduction) induced by intrapleural injection of carrageenan and the migration of polymorphonuclear cells into the inflamed pleural cavity of rats (40%). Additionally, rolling and adhesion of leukocytes and the number of leukocytes that migrated toward the perivascular space in response to the carrageenan injection were decreased by the extract (500 mg/kg). These data demonstrate the anti-inflammatory effect of the ethanol extract of Helicteres gardneriana and imply that inhibition of leukocyte-endothelial interactions is important in the extract's mechanism of action
Effect of crude extracts of leaves of Smallanthus sonchifolius (yacon) on glycemia in diabetic rats
O presente estudo investigou o efeito do extrato bruto das folhas de yacon, obtido por extração a frio (Y C) ou a quente (Y H), ou por extração hidro-alcoólica (Y E), na glicemia de ratos diabéticos (induzidos por estreptozocina, 50 mg/kg, i.p.) e não diabéticos. Os animais foram tratados por via oral com os extratos (400 mg/kg) ou água por um período de 3, 7, 10 e 14 dias. Após o tratamento, os animais foram anestesiados e amostras de sangue foram coletadas para determinações dos níveis de glicose e creatinina plasmática e da atividade de enzimas (AST, ALT, ALP). Também foi avaliado o consumo de ração, água, volume de urina, peso corporal dos animais e a DL50 do Y E. O tratamento dos animais com Y E, por 14 dias, reduziu a glicemia de ratos diabéticos e não diabéticos. Adicionalmente, o Y E restaurou a atividade das enzimas que estavam alteradas, e melhorou o ganho de peso dos animais diabéticos. Estes resultados demonstraram que a efetividade do extrato de yacon está relacionada ao método de preparação do extrato e ao tempo de tratamento dos animais. Liver Fatty Acid Composition and Inflammation in Mice Fed with High-Carbohydrate Diet or High-Fat Diet
Both high-carbohydrate diet (HCD) and high-fat diet (HFD) modulate liver fat accumulation and inflammation, however, there is a lack of data on the potential contribution of carbohydrates and lipids separately. For this reason, the changes in liver fatty acid (FA) composition in male Swiss mice fed with HCD or HFD were compared, at the time points 0 (before starting the diets), and after 7, 14, 28 or 56 days. Activities of stearoyl-CoA desaturase-1 (SCD-1), ∆-6 desaturase (D6D), elongases and de novo lipogenesis (DNL) were estimated. Liver mRNA expression of acetyl-CoA carboxylase 1 (ACC1) was evaluated as an additional indicator of the de novo lipogenesis. Myeloperoxidase activity, nitric oxide (NO) production, and mRNA expressions of F4/80, type I collagen, interleukin (IL)-6, IL-1β, IL-10, and tumor necrosis factor-α (TNF-α) were measured as indication of the liver inflammatory state. The HCD group had more intense lipid deposition, particularly of saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs). This group also showed higher DNL, SCD-1, and D6D activities associated with increased NO concentration, as well as myeloperoxidase activity. Livers from the HFD group showed higher elongase activity, stored more polyunsaturated fatty acids (PUFAs) and had a lower omega-6/omega-3 fatty acid (n-6/n-3) ratio. In conclusion, liver lipid accumulation, fatty acids (FA) composition and inflammation were modulated by the dietary composition of lipids and carbohydrates. The HCD group had more potent lipogenic and inflammatory effects in comparison with HFD

Toxicity studies of Cordia salicifolia extract
ABSTRACT. This study was carried out to determine the acute toxicity of the whole Cordia salicifolia extract (LD 50) after oral and intraperitoneal administration in mice, and its effect on certain biochemical parameters in the plasma of rats after 90 days of administration. The oral LD 50 value of the extract was higher than 2000 mg/kg while the LD 50 by intraperitoneal injections was about 920 mg/kg. A daily oral administration of extracts at 20, 100, 200 and 400 mg/kg doses for 90 days did not cause significant changes in the body weight gain, organs weight or biochemical assays and hematology in the animals. The results showed that the administration of the extract for a prolonged period did not produce toxic effects in the animals. Key words: Cordia salicifolia, porangaba, medicinal plants, toxicity. RESUMO. Estudo da toxidade do extrato de Cordia salicifolia. Este estudo foi realizado para determinar a toxicidade aguda do extrato total de Cordia salicifolia (DL50) após administração oral e intraperitoneal em camundongos, assim como os efeitos do extrato sobre alguns parâmetros bioquímicos no plasma de ratos após um tratamento prolongado (90 dias). A DL 50 do extrato administrado por v.o. foi maior que 2000 mg/Kg, enquanto a DL 50 por via i.p. foi aproximadamente 920 mg/Kg. A administração oral diária do extrato nas doses de 20, 100, 200 e 400 mg/kg por um período de 90 dias não causou modificações no ganho de peso corporal, no peso dos órgãos, nos parâmetros hematológicos e bioquímicos dos animais. Estes resultados indicam que a administração do extrato por um período mais prolongado não provocou efeitos de toxicidade nos animais.
