    Este estudo apresenta os principais resultados da anĂĄlise de como habitantes de Alto ParaĂ­so de GoiĂĄs estĂŁo buscando alternativas para o desenvolvimento sustentĂĄvel do municĂ­pio, por meio da implantação, pela Embrapa, de projeto relativo ao resgate do cafĂ©. A mineração e as atividades agropecuĂĄrias foram exercidas na regiĂŁo atĂ© os anos 60, quando a atividade principal passou a ser o turismo, a partir da criação do Parque Nacional da Chapada dos Veadeiros e da inauguração de BrasĂ­lia. Em 2000, o fluxo de turistas decaiu devido a problemas relativos Ă  saĂșde pĂșblica, acarretando estagnação da economia local. Nos Ășltimos anos, produtores familiares despertaram para a existĂȘncia, ali, de um cafĂ© que pode ser comercializado em nichos de mercado de grĂŁos especiais: orgĂąnicos e de origem definida, e buscaram, na Embrapa, o desenvolvimento de projeto de pesquisa. O estudo na regiĂŁo revelou que por meio do desvelamento de valores - histĂłria, cultura e tradiçÔes - Ă© possĂ­vel estabelecer estratĂ©gia mais eficiente de busca de mercado, a partir da experiĂȘncia revelada no trabalho concreto e na cultura dos produtores. E que o desenvolvimento rural deve ser buscado por meio do desenvolvimento de atividades da nova ruralidade e da aplicação de abordagem territorial de desenvolvimento

    Host immunity in the protective response to nasal immunization with a pneumococcal antigen associated to live and heat-killed Lactobacillus casei

    Background: At present, available pneumococcal vaccines have failed to eradicate infections caused by S. pneumoniae. Search for effective vaccine continues and some serotype independent pneumococcal proteins are considered as candidates for the design of new vaccines, especially a mucosal vaccine, since pneumococci enter the body through mucosal surfaces. Selection of the appropriate adjuvant is important for mucosal vaccines, and lactic acid bacteria (LAB) with immunostimulant properties are promissory candidates. In this work, we assessed the adjuvant effect of a probiotic strain, Lactobacillus casei (L. casei), when nasally administered with a pneumococcal antigen (pneumococcal protective protein A: PppA) for the prevention of pneumococcal infection. Adjuvanticity of both live (LcV) and heat-killed (LcM) was evaluated and humoral and cellular antigen-specific immune response was assessed in mucosal and systemic compartments. The potential mechanisms induced by nasal immunization were discussed.Results: Nasal immunization of young mice with PppA+LcV and PppA+LcM induced anti-PppA IgA and IgG antibodies in mucosal and systemic compartments and levels of these specific antibodies remained high even at day 45 after the 3rd Immunization (3rd I). These results were correlated with IL-4 induction by the mixture of antigen plus LcV and LcM. Also, PppA+Lc (V and M) induced stimulation of Th1 and Th17 cells involved in the defence against pneumococci. The protection against pneumococcal respiratory challenge at day 30 after the 3rd I showed that PppA+LcV and PppA+LcM immunizations significantly reduced pathogen counts in nasal lavages while prventing their passage into lung and blood. Survival of mice immunized with the co-application of PppA plus LcV and LcM was significantly higher than in mice immunized with PppA alone and control mice when intraperitoneal challenge was performed. No significant differences between the treatments involving LcV and LcM were found.Conclusions: Live and heat-killed L. casei enhanced the antigen-specific immune response when administered nasally with a pneumococcal antigen. Considering the potential risk associated with live bacteria, the design of a nasal vaccine based on pneumococcal antigens and heat-killed L. casei emerges as a safe and effective strategy for the prevention of pneumococcal infections and opens new possibilities of application of dead LAB as adjuvants in vaccine formulations against other pathogens.Fil: Vintiñi, Elisa Ofelia.     Saude mental na estrategia saude da familia : revisao da literatura brasileira = Mental health in the Family Health Strategy : a review of Brazilian literature

    The Family Health Strategy establishes the principles of the Brazilian Primary Health Care and shares important goals with the Psychiatric Reform. The principles of territory-centered care and longitudinal care should enhance innovative actions of mental health promotion, prevention and rehabilitation. The aim of this review was to analyze the main themes approached by the Brazilian scientific literature concerning mental health in the Family Health Strategy. We read the titles of 267 articles published between 1999 and 2009. We followed specific criteria to select 38 articles for thematic analysis. The main themes were the demands in mental health, the perceptions and practices of health personnel and the role of the psychologist in Primary Care. The publications identified several problems: stereotypical views about mental disorders, the dominance of the hospitalization rationale, and the absence of clinical reports, strategies, qualified support to families and integrated health actions. The qualitative meta-analysis indicated questions that may strengthen the debate on the topic, the reflection on further research and on professional practice in the interface between Mental Health and Family Health

    Leishmanicidal Metabolites from Cochliobolus sp., an Endophytic Fungus Isolated from Piptadenia adiantoides (Fabaceae)

    Protozoan parasites belonging to genera Leishmania and Trypanosoma are the etiological agents of severe neglected tropical diseases (NTDs) that cause enormous social and economic impact in many countries of tropical and sub-tropical areas of the world. In our screening program for new drug leads from natural sources, we found that the crude extract of the endophytic fungus Cochliobolus sp. (UFMGCB-555) could kill 90% of the amastigote-like forms of Leishmania amazonensis and inhibit by 100% Ellman's reagent reduction in the trypanothione reductase (TryR) assay, when tested at 20 ”g mL−1. UFMGCB-555 was isolated from the plant Piptadenia adiantoides J.F. Macbr (Fabaceae) and identified based on the sequence of the internally transcribed spacer (ITS) regions of its ribosomal DNA. The chromatographic fractionation of the extract was guided by the TryR assay and resulted in the isolation of cochlioquinone A and isocochlioquinone A. Both compounds were active in the assay with L. amazonensis, disclosing EC50 values (effective concentrations required to kill 50% of the parasite) of 1.7 ”M (95% confidence interval = 1.6 to 1.9 ”M) and 4.1 ”M (95% confidence interval = 3.6 to 4.7 ”M), respectively. These compounds were not active against three human cancer cell lines (MCF-7, TK-10, and UACC-62), indicating some degree of selectivity towards the parasites. These results suggest that cochlioquinones are attractive lead compounds that deserve further investigation aiming at developing new drugs to treat leishmaniasis. The findings also reinforce the role of endophytic fungi as an important source of compounds with potential to enter the pipeline for drug development against NTDs
