    Projection Operator: A Step Towards Certification of Adaptive Controllers

    One of the major barriers to wider use of adaptive controllers in commercial aviation is the lack of appropriate certification procedures. In order to be certified by the Federal Aviation Administration (FAA), an aircraft controller is expected to meet a set of guidelines on functionality and reliability while not negatively impacting other systems or safety of aircraft operations. Due to their inherent time-variant and non-linear behavior, adaptive controllers cannot be certified via the metrics used for linear conventional controllers, such as gain and phase margin. Projection Operator is a robustness augmentation technique that bounds the output of a non-linear adaptive controller while conforming to the Lyapunov stability rules. It can also be used to limit the control authority of the adaptive component so that the said control authority can be arbitrarily close to that of a linear controller. In this paper we will present the results of applying the Projection Operator to a Model-Reference Adaptive Controller (MRAC), varying the amount of control authority, and comparing controller s performance and stability characteristics with those of a linear controller. We will also show how adjusting Projection Operator parameters can make it easier for the controller to satisfy the certification guidelines by enabling a tradeoff between controller s performance and robustness

    Steering control of an autonomous ground vehicle with application to the DARPA Urban Challenge

    Thesis (S.M.)--Massachusetts Institute of Technology, Dept. of Mechanical Engineering, 2007.Includes bibliographical references (p. 156-157).Fundamental to the design of an Ackerman steered autonomous ground vehicle is the development of a low-level controller that effectively performs trajectory or path tracking. Though ample literature is available on various methods for controlling ground vehicles, little information is presented on the implementation and tuning of such controllers. Moreover, few sources extend ground vehicle control to driving in reverse. This work presents a novel approach to the implementation of the traditional "pure pursuit" style controller in which a dynamic vehicle model is used to map from the path curvature specified by the pure pursuit algorithm to the vehicle's actual steering angle. Additionally, an analytical methodology using a linear model of straight-line path following is used to tune the pure pursuit look-ahead distance. This pure pursuit controller is then contrasted with a simulation-based controller that uses a kinematic model to predict the vehicle's response to a series of different steering inputs; a performance metric is used to select the best command given these predictions. Successful trajectory control results are presented at speeds up to 22 mph. The second focus of this work is the control of a front-wheel steered vehicle driving in reverse. Novel to this work is the presentation of pure pursuit as a stable solution to this problem. Pure pursuit is then contrasted with the mechanism-based controller that was developed by Patwardhan et al. at the University of California Berkeley. In presenting this controller, a new method employing a linear kinematic vehicle model is used to tune the controller parameters. It is then shown that, under specific conditions, the mechanism-based controller and the pure pursuit controller are identical. Both controllers are then compared with the simulation-based controller adapted for driving in reverse.(cont.) Results are presented at speeds up to 6.7 mph. Results for the implementation of these controllers were collected using a 2006 Land Rover LR3 developed for MIT's entry into the 2007 DARPA Urban Challenge.     Investigating Automatic Measurements of Prosodic Accommodation and Its Dynamics in Social Interaction

    Spoken dialogue systems are increasingly being used to facilitate and enhance human communication. While these interactive systems can process the linguistic aspects of human communication, they are not yet capable of processing the complex dynamics involved in social interaction, such as the adaptation on the part of interlocutors. Providing interactive systems with the capacity to process and exhibit this accommodation could however improve their efficiency and make machines more socially-competent interactants. At present, no automatic system is available to process prosodic accommodation, nor do any clear measures exist that quantify its dynamic manifestation. While it can be observed to be a monotonically manifest property, it is our hypotheses that it evolves dynamically with functional social aspects. In this paper, we propose an automatic system for its measurement and the capture of its dynamic manifestation. We investigate the evolution of prosodic accommodation in 41 Japanese dyadic telephone conversations and discuss its manifestation in relation to its functions in social interaction. Overall, our study shows that prosodic accommodation changes dynamically over the course of a conversation and across conversations, and that these dynamics inform about the naturalness of the conversation flow, the speakers’ degree of involvement and their affinity in the conversation

    Implementation and Evaluation of Multiple Adaptive Control Technologies for a Generic Transport Aircraft Simulation

    Presented here is the evaluation of multiple adaptive control technologies for a generic transport aircraft simulation. For this study, seven model reference adaptive control (MRAC) based technologies were considered. Each technology was integrated into an identical dynamic-inversion control architecture and tuned using a methodology based on metrics and specific design requirements. Simulation tests were then performed to evaluate each technology s sensitivity to time-delay, flight condition, model uncertainty, and artificially induced cross-coupling. The resulting robustness and performance characteristics were used to identify potential strengths, weaknesses, and integration challenges of the individual adaptive control technologie

Current treatments are sub-optimal, and the disease is fatal if not treated. Herein, we report our continuing efforts to repurpose the human phosphodiesterase 4 (hPDE4) inhibitor piclamilast to target trypanosomal phosphodiesterase TbrPDEB1. We prepared a range of substituted heterocyclic replacements for the 4-amino-3,5-dichloro-pyridine head group of piclamilast, and found that these compounds exhibited weak inhibitory activity of TbrPDEB1.We acknowledge funding from the National Institutes of Health (R01AI082577)

    Responsiveness of serum C‐reactive protein, interleukin‐17A, and interleukin‐17F levels to ustekinumab in psoriatic arthritis: lessons from two phase III, multicenter, double‐blind, placebo‐controlled trials

    Objective: To evaluate the associations of C‐reactive protein (CRP) and circulating Th17‐associated cytokine levels with psoriatic arthritis (PsA) disease activity and therapeutic response to ustekinumab. Methods: Interleukin‐17A (IL‐17A), IL‐17F, IL‐23, and CRP concentrations were measured in serum samples collected as part of the 2 PSUMMIT phase III studies of ustekinumab in PsA (n = 927). In post hoc analyses, relationships of IL‐17A, IL‐17F, and CRP levels at baseline, week 4, and week 24 with baseline skin and joint disease activity and response to therapy were evaluated using generalized linear models and Pearson's product‐moment correlation tests. Results: Baseline serum levels of IL‐17A and IL‐17F were positively correlated with baseline skin disease scores (r = 0.39–0.62). IL‐23 levels were correlated with skin disease scores to a lesser extent (r = 0.26–0.31). No significant correlations were observed between these cytokine or CRP levels and baseline joint disease activity. There was no significant association of baseline levels of IL‐17A, IL‐17F, IL‐23, or CRP with therapeutic response to ustekinumab in either the skin or joints. Significant reductions from baseline in levels of IL‐17A, IL‐17F, and CRP were seen in patients treated with ustekinumab compared to those treated with placebo. Ustekinumab‐treated patients in whom 75% improvement in the Psoriasis Area and Severity Index score or 20% improvement according to the American College of Rheumatology criteria was achieved after 24 weeks of treatment had greater reductions in CRP level (geometric mean decreases of 51–58% versus 32–33%; P < 0.05), but not IL‐17A or IL‐17F levels, than nonresponders. Conclusion: Baseline serum IL‐23/IL‐17 levels correlated with skin, but not joint, disease activity, suggesting tissue‐specific variation. However, neither baseline Th17‐associated cytokine levels nor CRP level were predictive of therapeutic response to ustekinumab in the skin or joints, despite rapid reductions in their levels following ustekinumab therapy

    Gluino Pair Production at Linear e^+e^- Colliders

    We study the potential of high-energy linear e+e−e^+e^- colliders for the production of gluino pairs within the Minimal Supersymmetric Standard Model (MSSM). In this model, the process e+e−→g~g~e^+e^-\to\tilde{g}\tilde{g} is mediated by quark/squark loops, dominantly of the third generation, where the mixing of left- and right-handed states can become large. Taking into account realistic beam polarization effects, photon and Z0Z^0-boson exchange, and current mass exclusion limits, we scan the MSSM parameter space for various e+e−e^+e^- center-of-mass energies to determine the regions, where gluino production should be visible.Comment: 22 pages, 9 figure

    We generalize a five dimensional black hole solution of low energy effective string theory to arbitrary constant spatial curvature. After interchanging the signature of time and radius we reduce the 5d solution to four dimensions and obtain that way a four dimensional isotropic cosmological space time.     The investigation of lipoxygenases as therapeutic targets in malignant pleural mesothelioma

    Advanced malignant pleural mesothelioma (MPM) has an extremely poor prognosis with limited chemotherapy options, therefore the identification of new therapeutic targets would aid in disease management. Arachidonic acid is metabolised by cyclooxygenase and lipoxygenase enzymes. The lipoxygenase isoenzymes 5-LOX and 12-LOX have been implicated in carcinogenesis. We aimed to examine 5-LOX and 12-LOX protein expression in a large retrospective series of mesothelioma samples. Further to this, the in vitro cytotoxic effects of lipoxygenase pathway inhibitors were investigated in mesothelioma cells. Archival samples from 83 patients with MPM were examined by immunohistochemistry for expression of the 5-LOX and 12-LOX proteins. The MTS assay was used to assess cell viability following 72h treatment with the lipoxygenase pathway inhibitors baicalein, licofelone, MK-886 and zileuton in the MPM cell lines NCI-H2052, NCI-H2452 and MSTO-211H. Positive 12-LOX protein expression was recorded in 69/83 (83%) and positive 5-LOX expression was observed in 56/77 (73%) of MPM tissue samples. Co-expression of 5-LOX with 12-LOX was seen in 46/78 (58%) of MPM samples. Positive expression of 5-LOX, 12-LOX and COX-2 proteins was identified in the NCI-H2052, NCI-H2452 and MSTO-211H MPM cell lines. Baicalein (12-LOX and 15-LOX inhibitor) was effective in 3/3 MPM cell lines at low concentrations with an IC50 range of 9.6ÎŒM to 20.7ÎŒM. We have demonstrated that the 5-LOX and 12-LOX proteins are expressed in a significant proportion of MPM samples (73% and 83% respectively) and may represent novel therapeutic targets in this disease. We have demonstrated that the inhibition of the LOX pathway using baicalein may be effective as a novel treatment for MPM, however further human pharmacokinetic studies are required in order to establish whether the concentration used in vitro is clinically achievable
