    Stimulatory activity of four green freshwater sponges on aquatic mycotal communities

    The influence of the four species of green sponges (Ephydatia muelleri, Heteromeyenia stepanowii, Spongilla fluviatilis, and Spongilla lacustris) on the occurrence of aquatic mycotal species in the water of three four water bodies of different trophy was investigated in this study. Seeds and snake exuviae were used as baits. For the measurement of the primary and extracellular production by symbiotic algae of green sponges and assimilation of those products by mycota, radioactive carbon (14C) was used. A total of 75 mycotal species were found to be growing on the baits. The fewest mycota were noted in the containers in water from oligotrophic Lake Hańcza; the most in the containers with eutrophic water from River Supraśl. More mycota were found to grow in the containers with green sponges (Sp) than in the controls (Co) in water from all water bodies. The mean ratio of Sp/Co in green sponges ranged from 2.30 (E. muelleri) to 4.80 (H. stepanowii); in brown colonies (without symbiotic algae) it was 0.90. Mean value of 14C fixation (primary production) in symbiotic algae of Spogilla fluviatilis was 5.67 mg C g-1 dry weight sponge per hour. The effect of green sponges on the abundance of aquatic mycotal species is caused by dissolved organic matter produced during photosynthesis by symbiotic zoochlorellae, a symbionts of green sponges and excreted into the water environment (S. fluviatilis excreted mean 12.8% of carbon fixation). Those excreted organic substances serve as nutrients for aquatic mycota. The mean value of extracellular products assimilated by mycota was 4.96 mg C g-1 dry weight mycelium per hour.Key words: Sponges, symbiotic zoochlorellae, aquatic mycota, interactions, hydrochemistry

    Amount and qualities of carotenoids in fillets of fish species fed with natural feed in some fisheries of West African Coast

    Using column (CC), thin- layer (TLC) and high- performance liquid chromatography (HPLC), carotenoid content was examined in the fillets (muscles with skin) of 16 fish species from the fisheries of West African Coast. 15 carotenoids, including 6 ketocarotenoids (4’- hydroxyechinenone, canthaxanthin, phoenicopterone, phoenicoxanthin, astaxanthin, 7,8- didehydroastaxanthin) were found, with a predominance of 3,4- dihydro- α- carotene, zeaxanthin, phoenicopterone, canthaxanthin, phoenicoxanthin and astaxanthin. Total carotenoid content in the examined material ranged from 0.086 (Merluccius merluccius) to 0.352 μg g-1 wet mass (Macrurus aequalis).     Zoosporic aquatic fungi growing on avian excrements in various types of water bodies

    AbstractCoprophilic fungi in the water from six different water bodies were studied between 1995–1997. For hydrochemical analysis samples of water were collected every three months while for estimation of fungus content every month. Excrements of 3 phytophagous (greylag goose, roman-nosed goose, mute swan), 4 carnivorous (grey heron, white stork, marsh harrier, cormorant) and 3 omnivorous bird species (musk-duck, mallard, black-headed gull) were used as bait.Thirty-six fungus species growing on avian excrements in the water of all examined water bodies were found, 6 of these species belonged to Chytridiomycetes and 30 to Oomycetes. The largest number of fungus species was detected on excrements of musk duck, mallard and mute swan, the smallest one on excrements of roman-nosed goose, marsh harrier and cormorant. Such fungus species as Achlya americana, Aphanomyces laevis, Dictyuchus monosporus, Pythium debaryanum, Pythium rostratum, Saprolegnia asterophora, Saprolegnia ferax and Saprolegnia parasitica were detected on excrements of all birds examined.     Carotenoids and retinyl esters are the source of vitamin A in the human body and its natural derivatives takes part in the regulation of cell replication and differentiation in the human endometrium, may induce the leiomyoma growth and has a role in differentiation of endometrial adenocarcinoma. The aim of the study was to demonstrate the presence of carotenoids in tissues from the normal uterus and from various tumors of the uterine corpus, as well as to compare the total content, major carotenoids and % of carotenoids belonging to the provitamin A group between the tissues examined. Using three independent methods of chromatography (CC, TLC, HPLC) we analysed 140 human samples. We identified 13 carotenoids belonging to the eg. provitamin A group and epoxy carotenoids. In all the samples beta-carotene, beta-cryptoxanthin, lutein, neoxanthin, violaxanthin and mutatoxanthin were isolated. In normal tissues, the mean carotenoid content was the highest in the follicular phase endometrium (9.9 microg/g), while the highest percentage of carotenoids belonging to provitamin A group was found in the luteal phase (18.2%). In the pathological group, the highest mean values were demonstrated for epithelial lesions (8.0 microg/g), and within this group - in endometrioid adenocarcinoma (10.8 microg/g). In both groups, violaxanthin, beta-cryptoxanthin, lutein epoxide and mutatoxanthin were the predominant carotenoids. We have demonstrated that all uterine tissues show a concentration of beta-carotene and beta-cryptoxanthin, being the source of vitamin A. The highest total values of carotenoids obtained in the group of endometrioid adenocarcinoma seem to confirm certain enzymatic defects in carotenoid metabolism in the course of the neoplastic process or some metabolic modifications.     Can vitamin A modify the activity of docetaxel in MCF-7 breast cancer cells?

    Docetaxel is one of the most effective chemotherapeutic agents in the treatment of breast cancer. On the other hand, the vitamin A family compounds play the essential roles in many biological processes in mammary gland. The aim of our study was to investigate the effect of all-trans retinol, carotenoids (beta-carotene, lycopene) and retinoids (9-cis, 13-cis and all-trans retinoic acid) on the activity of docetaxel and to compare these effects with the estradiol and tamoxifen actions on human ER(+) MCF-7 breast cancer cell line. The evaluation was based on [3H] thymidine incorporation and the proliferative activity of PCNA and Ki 67 positive cells. In our study, the incorporation of [3H] thymidine into cancer cells was inhibited to 50% by 0.2, 0.5 and 1 microM of docetaxel in the 24-hour culture and addition of estradiol (0.001 microM) didn't influence the results. However, addition of tamoxifen caused a statistically significant decrease of the percentage of the proliferating cells in the culture medium with 0.2 and 0.5 microM of docetaxel (38.99 +/- 2.84%,

    P. 510-518The aquatic fungi cultured from eggs, alevins and broodfish of brown trout Salmo trutta belonged to the genus Saprolegnia and were identified as S. diclina, S. australis, S. ferax, S. furcata, S. hypogyna, S. unispora and S. parasitica. The species obtained from infected eggs and alevins were different to those from infected fish. Several Saprolegnia species were isolated from eggs and alevins, whereas all the isolates obtained from broodfish were the pathogenic S. parasitica.S

    Endometrioid carcinoma represents approximately 10% of cases of the malignant ovarian epithelial tumors. According to literature, the vitamin A (carotenoids and retinoids) plays an essential role in cell proliferation, differentiation and apoptosis in both normal and neoplastic ovarian tissues. Apart from that, the retinoids alter a cytotoxic effect of chemiotherapeutics, i.e. docetaxel, on ovarian cancer cell lines. Retinoids act on cancer cells throughout different mechanism than taxanes, so they may be the potential candidates for the new treatment strategies of ovarian cancer. The aim of the study was to determine the effects of vitamin A family compounds (retinol, beta-carotene, lycopene, all-trans -, 9-cis - and 13-cis retinoic acid) on the growth and proliferation of CRL-11731 endometrioid ovary cancer cell line and on docetaxel and estradiol activity in this culture. The assay was based on [3H] thymidine incorporation and the proliferative activity of PCNA- and Ki 67-positive cells. The apoptotic index and expression of the Bcl-2 and p53 antigens in CRL-11731 cells were also studied. Among vitamin A family compounds retinol and carotenoids, but not retinoids, inhibited the growth of cancer cells in dose dependent manner. Only the concentration of 100 muM of docetaxel inhibited incorporation [3H] thymidine into CRL-11731 cancer cells. Retinol (33.4%+/-8.5), carotenoids (beta-carotene 20 muM 4.7%+/-2.9, 50 muM 2.2%+/-0.9; lycopene 10 muM 7.6%+/-0.8, 20 muM 5.2%+/-2.5, 50 muM 2.9%+/-1.2), and 13-cis retinoic acid (19.7%+/-2.2) combined with docetaxel (100 muM) significantly decreased the percentage of proliferating cells (

    The effect of doxorubicin and retinoids on proliferation, necrosis and apoptosis in MCF-7 breast cancer cells.

    Doxorubicin (Adriamycin) is the most active drug in the treatment of breast cancer. The aim of this study was to investigate the interaction of doxorubicin and retinoids in the inhibition of proliferation of hormone sensitive (ER+) human breast cancer cell line MCF-7 and to find out whether this combination can result in the enhancement of its therapeutic effect. As a comparison we also used estradiol and tamoxifen. We also made an attempt to elucidate the effect of these compounds on the stimulation of the apoptotic pathway in breast cancer cells. Cell proliferation in a 24-hour culture was assessed by [3H] thymidine incorporation into cancer cells and by immunocytochemical analysis of cellular cycle-related PCNA and Ki-67 antigens expression, after the incubation of the cell culture with 10, 20 and 50 nM doxorubicin (DOX), 2 nM estradiol (E2), 10 microM tamoxifen (TAM) and 1 nM, 0.01, 0.1, 1 and 10 microM of all-trans retinoid acid (ATRA). The assessment of cell viability and analysis of apoptotic and necrotic cells were performed after the 72-hour incubation of the culture with the examined substances and following apoptosis induction using acridine orange and ethidine bromide. Of the doxorubicin concentrations used in the study, 20 nM inhibited thymidine incorporation to 84.83 +/- 10.00% (control=100%). In the same culture conditions, 2 nM E2 stimulated cancer cells to 157.09 +/- 8.84%. Concentrations of 10 microM TAM and 10 microM ATRA inhibited the proliferation to 63.16 +/- 7.85% and 52.19 +/- 3.21%, respectively. A statistically significant reduction of these values was observed when 20 nM DOX was added to medium with E2 - 39.24 +/- 7.6%, TAM - 48.34 +/- 2.05% and ATRA - 21.98 +/- 1.69%, respectively; the percentage of PCNA- and Ki-67-positive cells was also reduced. Despite high antiproliferative efficacy of 20 nM DOX and 10 microM ATRA combination, the percentage of apoptotic cells was only 25 +/- 0.81%, being similar to that obtained in the culture with 20 nM DOX. The concentrations of 10, 20 and 50 nM DOX that were used to inhibit the proliferation of MCF-7 cell line were not particulary effective. The inhibitory effect was obtained when 20 nM of DOX and E2, TAM or ATRA were used simultaneously. The use of E2 caused a two-fold decrease in the percentage of proliferating cells. It was also shown that the effectiveness of DOX in combination with ATRA is significantly higher than that of DOX combined with TAM, which might suggest a valuable approach to the treatment of breast cancer
