    A new method for automatic Multiple Partial Discharge Classification

    A new wavelet based feature parameter have been developed to represent the characteristics of PD activities, i.e. the wavelet decomposition energy of PD pulses measured from non-conventional ultra wide bandwidth PD sensors such as capacitive couplers (CC) or high frequency current transformers (HFCT). The generated feature vectors can contain different dimensions depending on the length of recorded pulses. These high dimensional feature vectors can then be processed using Principal Component Analysis (PCA) to map the data into a three dimensional space whilst the first three most significant components representing the feature vector are preserved. In the three dimensional mapped space, an automatic Density-Based Spatial Clustering of Applications with Noise (DBSCAN) algorithm is then applied to classify the data cluster(s) produced by the PCA. As the procedure is undertaken in a three dimensional space, the obtained clustering results can be easily assessed. The classified PD sub-data sets are then reconstructed in the time domain as phase-resolved patterns to facilitate PD source type identification. The proposed approach has been successfully applied to PD data measured from electrical machines and power cables where measurements were undertaken in different laboratories

    The Time of Flight System of the AMS-02 Space Experiment

    The Time-of-Flight (TOF) system of the AMS detector gives the fast trigger to the read out electronics and measures velocity, direction and charge of the crossing particles. The new version of the detector (called AMS-02) will be installed on the International Space Station on March 2004. The fringing field of the AMS-02 superconducting magnet is 1.0Ă·2.51.0\div2.5 kG where the photomultiplers (PM) are installed. In order to be able to operate with this residual field, a new type of PM was chosen and the mechanical design was constrained by requiring to minimize the angle between the magnetic field vector and the PM axis. Due to strong field and to the curved light guides, the time resolution will be 150Ă·180150\div180 ps, while the new electronics will allow for a better charge measurement.Comment: 5 pages, 4 figures.     The complexity of soil biological sustainability

    Additions of organic amendments to soil not only compensate for decreased soil C, but also contribute to energy requirements for conserving biological activity levels. The soil microbial biomass displays some highly conserved, and possibly unique, characteristics that do not permit a classic interpretation of microbial metabolic parameter data. The resilience of soil microbial biomass and the role of soil organic matter in sustaining microbial biomass under practically zero C inputs were assessed in two long term incubation experiments using soils from the Broadbalk experiment at Rothamsted (UK). Soils with low organic C contents, showed the greatest decline in biomass C during the first 30 d of incubation. The ATP concentration of this rapidly declining microbial biomass did not change during the prolonged incubation period, confirming this peculiar character of the soil microbial biomass. Specific respiration rate did not depend upon substrate availability, being higher in soils that had received the lowest C inputs. Qualitative and quantitative changes observed in humic fractions suggest that humified soil organic matter is a much more dynamic soil fraction than is normally considered and provides a utilizable energy reserve for soil microorganisms. Carbon levels can be successfully restored in soils through practices such as incorporation of crop residues, re\u2010vegetation and application of manures, biosolids and composts. Some amendments, such as olive mill waste compost, promote incorporation of altered lignin structures, N\u2010containing compounds and carbohydrates into humic acids. The mineral\u2010bound fraction of humic C also increases, after their addition, and contributes to the accumulation of the most inert soil C pools

    Critical involvement of calcium-dependent cytosolic phospholipase A2\u3b1 in aortic valve interstitial cell calcification

    The involvement of calcium-dependent cytosolic phospholipase A2\u3b1 (cPLA2\u3b1) in aortic valve calcification is not exhaustively elucidated. Here, cPLA2\u3b1 expression in aortic valve interstitial cell (AVIC) pro-calcific cultures simulating either metastatic or dystrophic calcification was estimated by qPCR, Western blotting, and counting of cPLA2\u3b1-immunoreactive cells, with parallel ultrastructural examination of AVIC calcific degeneration. These evaluations also involved pro-calcific AVIC cultures treated with cPLA2\u3b1 inhibitor dexamethasone. cPLA2\u3b1 over-expression resulted for both types of pro-calcific AVIC cultures. Compared to controls, enzyme content was found to increase by up to 300% and 186% in metastatic and dystrophic calcification-like cultures, respectively. Increases in mRNA amounts were also observed, although they were not as striking as those in enzyme content. Moreover, cPLA2\u3b1 increases were time-dependent and strictly associated with mineralization progression. Conversely, drastically lower levels of enzyme content resulted for the pro-calcific AVIC cultures supplemented with dexamethasone. In particular, cPLA2\u3b1 amounts were found to decrease by almost 88% and 48% in metastatic and dystrophic calcification-like cultures, respectively, with mRNA amounts showing a similar trend. Interestingly, these drastic decreases in cPLA2\u3b1 amounts were paralleled by drastic decreases in mineralization degrees, as revealed ultrastructurally. In conclusion, cPLA2\u3b1 may be regarded as a crucial co-factor contributing to AVIC mineralization in vitro, thus being an attractive potential target for designing novel therapeutic strategies aimed to counteract onset or progression of calcific aortic valve diseases

    An assay system to evaluate riboflavin/UV-A corneal phototherapy efficacy in a porcine corneal organ culture model

    The purpose of this study is to investigate the response of a porcine corneal organ cultures to the riboflavin/UV-A phototherapy in the injury healing of induced lesions. A porcine corneal organ culture model has been established. Corneal alterations in the stroma were valuated setting an assay system, based on an automated image analysis method able to quantify the damaged (brightness values), within of the 24 regions of interest (ROIs) in which the corneal section has been divided and integrating the data analysis with a multi-aspect approach. Three group of corneas have been analyzed: (healthy, injured and injured-and-treated group). Our study revealed a significant effect of the riboflavin/UV-A phototherapy in the injury healing of porcine corneas after induced lesions. The injured corneas had significant differences of brightness values in comparison to treated (p< 0.00) and healthy (p<0.001) corneas whereas the treated and healthy corneas showed not significant difference (p = 0.995). Riboflavin/UV-A phototherapy shows a significant effect in the restoring the brightness values of damaged corneas to the values of healthy corneas, suggesting treatment restores the injury healing of corneas after lesions. Our assay system may be compared to clinical diagnostic method such as the OCT imaging for in vivo damaged ocular structures investigations

    How resistant are levodopa-resistant axial symptoms? Response of freezing, posture, and voice to increasing levodopa intestinal infusion rates in Parkinson disease

    Background and purpose: Treatment of freezing of gait (FoG) and other Parkinson disease (PD) axial symptoms is challenging. Systematic assessments of axial symptoms at progressively increasing levodopa doses are lacking. We sought to analyze the resistance to high levodopa doses of FoG, posture, speech, and altered gait features presenting in daily-ON therapeutic condition. Methods: We performed a pre-/postinterventional study including patients treated with levodopa/carbidopa intestinal gel infusion (LCIG) with disabling FoG in daily-ON condition. Patients were evaluated at their usual LCIG infusion rate (T1), and 1 h after 1.5× (T2) and 2× (T3) increase of the LCIG infusion rate by quantitative outcome measures. The number of FoG episodes (primary outcome), posture, speech, and gait features were objectively quantified during a standardized test by a blinded rater. Changes in motor symptoms, dyskinesia, and plasma levodopa concentrations were also analyzed. Results: We evaluated 16 patients with a mean age of 69 ± 9.4 years and treated with LCIG for a mean of 2.2&nbsp;± 2.1 years. FoG improved in 83.3% of patients by increasing the levodopa doses. The number of FoG episodes significantly decreased (mean&nbsp;=&nbsp;2.3 at T1, 1.7 at T2, 1.2 at T3; p&nbsp;= 0.013). Posture and speech features did not show significant changes, whereas stride length (p&nbsp;= 0.049), turn duration (p&nbsp;= 0.001), and turn velocity (p&nbsp;= 0.024) significantly improved on doubling the levodopa infusion rate. Conclusions: In a short-term evaluation, the increase of LCIG dose can improve "dopa-resistant" FoG and gait issues in most advanced PD patients with overall good control of motor symptoms in the absence of clinically significant dyskinesia

    An assay for secologanin in plant tissues based on enzymatic conversion into strictosidine

    The secoiridoid glucoside secologanin is the terpenoid building block in the biosynthesis of terpenoid indole alkaloids. A method for its determination in plant tissues and in cell suspension cultures has been developed. This assay is based on the condensation of secologanin with tryptamine, yielding strictosidine, in a reaction catalysed by the enzyme strictosidine synthase (STR; E.C. 4.3.3.2). Subsequently, the formation of strictosidine is quantified by high performance liquid chromatography (HPLC). STR was isolated from transgenic Nicotiana tabacum cells expressing a cDNA-derived gene coding for STR from Catharanthus roseus. The high specificity of STR for secologanin, in combination with a sensitive and selective HPLC system, allows a simple extraction of secologanin from plant tissue. The detection limit of this method is 15 ng secologanin. Using this assay, secologanin contents were determined in tissues of various plant species; Lonicera xylosteum hairy roots were found to contain 1% of secologanin on a dry weight basis. # 1998 John Wiley & Sons, Ltd.info:eu-repo/semantics/publishedVersio
