    Constitutive expression of ftsZ overrides the whi developmental genes to initiate sporulation of Streptomyces coelicolor

    The filamentous soil bacteria Streptomyces undergo a highly complex developmental programme. Before streptomycetes commit themselves to sporulation, distinct morphological checkpoints are passed in the aerial hyphae that are subject to multi-level control by the whi sporulation genes. Here we show that whi-independent expression of FtsZ restores sporulation to the early sporulation mutants whiA, whiB, whiG, whiH, whiI and whiJ. Viability, stress resistance and high-resolution electron microscopy underlined that viable spores were formed. However, spores from sporulation-restored whiA and whiG mutants showed defects in DNA segregation/condensation, while spores from the complemented whiB mutant had increased stress sensitivity, perhaps as a result of changes in the spore sheath. In contrast to the whi mutants, normal sporulation of ssgB null mutants—which fail to properly localise FtsZ—could not be restored by enhancing FtsZ protein levels, forming spore-like bodies that lack spore walls. Our data strongly suggest that the whi genes control a decisive event towards sporulation of streptomycetes, namely the correct timing of developmental ftsZ transcription. The biological significance may be to ensure that sporulation-specific cell division will only start once sufficient aerial mycelium biomass has been generated. Our data shed new light on the longstanding question as to how whi genes control sporulation, which has intrigued scientists for four decades

    Antihyperlipidemic and antiperoxidative effect of Diasulin, a polyherbal formulation in alloxan induced hyperglycemic rats

    BACKGROUND: This study was undertaken to investigation the effect of Diasulin, a poly herbal drug composed of ethanolic extract of ten medicinal plants on blood glucose, plasma insulin, tissue lipid profile, and lipidperoxidation in alloxan induced diabetes. METHODS: Ethanolic extract of Diasulin a, poly herbal drug was administered orally (200 mg/kg body weight) for 30 days. The different doses of Diasulin on blood glucose and plasma insulin in diabetic rats were studied and the levels of lipid peroxides [TBARS, and Hydroperoxide] and tissue lipids [cholesterol, triglyceride, phospholipides and free fatty acids] were also estimated in alloxan induced diabetic rats. The effects were compared with glibenclamide. RESULT: Treatment with Diasulin and glibenclamide resulted in a significant reduction of blood glucose and increase in plasma insulin. Diasulin also resulted in a significant decrease in tissue lipids and lipid peroxide formation. The effect produced by Diasulin was comparable with that of glibenclamide. CONCLUSION: The decreased lipid peroxides and tissue lipids clearly showed the antihyperlipidemic and antiperoxidative effect of Diasulin apart from its antidiabetic effect

    Internally coupled ears in living mammals.

    It is generally held that the right and left middle ears of mammals are acoustically isolated from each other, such that mammals must rely on neural computation to derive sound localisation cues. There are, however, some unusual species in which the middle ear cavities intercommunicate, in which case each ear might be able to act as a pressure-difference receiver. This could improve sound localisation at lower frequencies. The platypus Ornithorhynchus is apparently unique among mammals in that its tympanic cavities are widely open to the pharynx, a morphology resembling that of some non-mammalian tetrapods. The right and left middle ear cavities of certain talpid and golden moles are connected through air passages within the basicranium; one experimental study on Talpa has shown that the middle ears are indeed acoustically coupled by these means. Having a basisphenoid component to the middle ear cavity walls could be an important prerequisite for the development of this form of interaural communication. Little is known about the hearing abilities of platypus, talpid and golden moles, but their audition may well be limited to relatively low frequencies. If so, these mammals could, in principle, benefit from the sound localisation cues available to them through internally coupled ears.     Background Preschoolers regularly display disruptive behaviors in child care settings because they have not yet developed the social skills necessary to interact prosocially with others. Disruptive behaviors interfere with daily routines and can lead to conflict with peers and educators. We investigated the impact of a social skills training program led by childcare educators on children’s social behaviors and tested whether the impact varied according to the child’s sex and family socio-economic status. Methods Nineteen public Child Care Centers (CCC, n = 361 children) located in low socio-economic neighborhoods of Montreal, Canada, were randomized into one of two conditions: 1) intervention (n = 10 CCC; 185 children) or 2) wait list control (n = 9 CCC; 176 children). Educators rated children’s behaviors (i.e., disruptive and prosocial behaviors) before and after the intervention. Hierarchical linear mixed models were used to account for the nested structure of the data. Results At pre-intervention, no differences in disruptive and prosocial behaviors were observed between the experimental conditions. At post-intervention, we found a significant sex by intervention interaction (β intervention by sex = − 1.19, p = 0.04) indicating that girls in the intervention condition exhibited lower levels of disruptive behaviors compared to girls in the control condition (f2 effect size = − 0.15). There was no effect of the intervention for boys. Conclusions Girls may benefit more than boys from social skills training offered in the child care context. Studies with larger sample sizes and greater intervention intensity are needed to confirm the results

    Heterologous expression of tylosin polyketide synthase and production of a hybrid bioactive macrolide in Streptomyces venezuelae

    Tylosin polyketide synthase (Tyl PKS) was heterologously expressed in an engineered strain of Streptomyces venezuelae bearing a deletion of pikromycin PKS gene cluster using two compatible low-copy plasmids, each under the control of a pikAI promoter. The mutant strain produced 0.5 mg/l of the 16-membered ring macrolactone, tylactone, after a 4-day culture, which is a considerably reduced culture period to reach the maximum production level compared to other Streptomyces hosts. To improve the production level of tylactone, several precursors for ethylmalonyl-CoA were fed to the growing medium, leading to a 2.8-fold improvement (1.4 mg/ml); however, switching the pikAI promoter to an actI promoter had no observable effect. In addition, a small amount of desosamine-glycosylated tylactone was detected from the extract of the mutant strain, revealing that the native glycosyltransferase DesVII displayed relaxed substrate specificity in accepting the 16-membered ring macrolactone to produce the glycosylated tylactone. These results demonstrate a successful attempt for a heterologous expression of Tyl PKS in S. venezuelae and introduce S. venezuelae as a rapid heterologous expression system for the production of secondary metabolites.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/45861/1/253_2006_Article_318.pd

    Daily Deviations in Anger, Guilt, and Sympathy: A Developmental Diary Study of Aggression

    With a diary study of 4- and 8-year-olds, we tested the association between daily deviations in anger and aggressive behavior, and whether this link was moderated by feelings of guilt and sympathy. Caregivers reported their children’s anger and aggression for 10 consecutive days (470 records; N = 80, 53 % girls). To calculate daily anger deviations from average anger levels, we subtracted each child’s average anger score (i.e., across 10 days) from his/her daily anger scores. Children reported their guilty feelings in response to vignettes depicting intentional harm, as well as their dispositional sympathy levels. Multilevel modeling indicated that within-child spikes in daily anger were associated with more aggression, above and beyond between-child differences in average anger levels. However, this association was weaker for children who reported higher levels of guilt. Sympathy did not moderate the anger-aggression link.     Erosion rates and sediment flux within the Potomac River basin quantified over millennial timescales using beryllium isotopes

    Beryllium isotopes measured in detrital river sediment are often used to estimate rates of landscape change at a basin scale, but results from different beryllium isotope systems have rarely been compared. Here, we report measurements of in situ and meteoric 10Be (10Bei and 10Bem, respectively) along with measurements of reactive and mineral phases of 9Be (9Bereac and 9Bemin, respectively) to infer long-term rates of landscape change in the Potomac River basin, North America. Using these data, we directly compare results from the two different 10Be isotope systems and contextualize modern sediment flux from the Potomac River basin to Chesapeake Bay. Sixty-two measurements of 10Bei in river sand show that the Potomac River basin is eroding on average at 29.6 ± 14.1 Mg km–2 yr–1 (11 ± 5.2 m m.y.–1 assuming a rock density of 2700 kg m–3)—a rate consistent with other estimates in the mid-Atlantic region. 10Bei erosion rates correlate with basin latitude, suggesting that periglacial weathering increased with proximity to the former Laurentide Ice Sheet margin. Considering the 10Bei-derived erosion rate as a sediment flux over millennia, rates of sediment delivery from the Potomac River to Chesapeake Bay are up to ∼5× lower than contemporary sediment yields implying modern land-use practices have accelerated erosion and sediment transport over background rates. However, 10Bei erosion rate data suggest that regulatory benchmark levels used to manage sediment export from the Potomac River basin to Chesapeake Bay are set appropriately to reduce sedimentation and restore the Bay’s ecological health. The mean of 56 10Bem/9Bereac-derived denudation rates (40.0 ± 21.7 Mg km–2 yr–1) is higher than, but statistically indistinguishable from, the mean 10Bei erosion rate (29.6 ± 14.1 Mg km–2 yr–1; p = 0.003). However, when considered basin by basin, 10Bem/9Bereac-determined denudation rates are only weakly correlated (R2 = 0.208; p 1, suggesting that 10Bem is being retained and sediment is being stored within the Potomac River basin. The Appalachian Plateau is the only physiographic province where sediment export dominates, likely as the result of ongoing relief growth in catchments draining the Appalachian Mountain divide. 10Bem concentrations measured in the 150 k.y. Hybla Valley sediment core, taken from the lower Potomac River basin, suggest that 10Bem and sediment are preferentially stored in the catchment when vegetation proxies for climate suggest warmer conditions prevailed. 10Bem and sediment are exported when vegetation proxies for climate suggest conditions are colder, perhaps a reflection of periglacial activity or changes in storm frequency and/or magnitude over glacial-interglacial cycles.
