    Forma Quadrata Britanniae : the case of Londinium

    In a previous research on the urban form of Alnwick, in Northumberland, we set out to integrate the well-known urban analysis of M. R. G. Conzen with the method of 'reading' of the Muratorian school (Cataldi, 2013). In particular, we had tried to apply the theory of the Forma Quadrata (Cataldi, 2014) to the entire territory of Great Britain, assuming that the origin of the 'square' geographical system, oriented secundum coelum, had been placed by the Romans in Dover (Portus Dubris), from where in 43 A.D. the occupation of the Island had begun. The verification of this hypothesis , with particular reference to the territory of Roman London, was carried out on the Map Ordnance Survey of Roman Britain at a scale of 1: 625,000, which topographically documents the location of the Roman centers and military fortresses within the entire roman road network, still largely functioning. We are pleased to present a preview of this work, which we are submitting in particular to the attention of the British colleagues, who are more interested in the knowledge of the permanent substrate structures, which have conditioned, and still condition, the territorial structure of Great Britain

    Assay of riboflavin in sample wines by capillary zone electrophoresis and laser-induced fluorescence detection

    To routinely assay the concentration of riboflavin (RF) in wines, a rapid and sensitive method was developed and evaluated. The method is based on a simple sample preparation, capillary zone electrophoretic separation and laser-induced fluorescence detection (CZE-LIF). Sample-preparation required only dilution and filtration. Under optimized conditions, the limit of detection of riboflavin was 0.5 mug/L, using a hydrodynamic sample introduction of 10 s at 54 mbar. The method was fully validated: the recovery of RF in wines was >95%. The concentrations of RF within the three sample types of Italian wines investigated here ranged from 69 to 151 mug/L with a mean value(+/-SD) of 112 +/- 25 mug/L, from 74 to 193 mug/L with a mean value of 115 +/- 45,ug/L, and from 156 to 292 mug/L with a mean value of 226 +/- 40 mug/L, for white, rose and red wines, respectively. Such an accurate and highly sensitive CZE-LIF method represents a powerful improvement over previous methods in terms of sensitivity, simplicity, and efficiency. It is well suited to satisfy the demands for accurate and sensitive detection with minimal sample preparation and cleanup

    ARMONIA Project: Transfrontier Strategy in the Management of Earthquakes

    The ARMONIA project between Italy and Austria, represents a cross-border strategy for prevention and management of seismic risk through real-time production of shaking and damage scenarios using integration and processing of data from seismic and accelerometer monitoring networks. Partners are developing an innovative seismic monitoring system to have an immediate evaluation of the damages distribution and to plan interventions immediately after the occurrence of a destructive earthquake. The first year of ARMONIA project was focused on the reinforcement and improvement of cross border existing free-field accelerometric network and implementation of a dense monitoring network of "significant" buildings through the creation of a dense monitoring network of sensible target buildings in near fault high risk areas along the cross-border territory between Italy and Austria. New instrumentation has been tested and new sites have been selected by following common protocols. This allows to enhance the effectiveness in a transnational prospective, in order to obtain the required quality of the data together with the maximum distribution of the instruments in the territory and provides high level of efficiency adding important information in case of emergency

    Ibuprofen and Lipoic Acid Diamide as Co-Drug with Neuroprotective Activity: Pharmacological Properties and Effects in β-Amyloid (1–40) Infused Alzheimer's Disease Rat Model

    Both oxidative stress and inflammation are elevated in brains of Alzheimer's disease patients, but their pathogenic significance still remains unclear. Current evidence support the hypothesis that non-steroidal anti-inflammatory drugs (NSAIDs) and antioxidant therapy might protect against the development of Alzheimer's disease, and ibuprofen has the strongest epidemiological support. In the present work our attention was focused on (R)-α-lipoic acid considered as a potential neuroprotective agent in Alzheimer's disease therapy. In particular, we investigated a new co-drug (1) obtained by joining (R)-α-lipoic acid and ibuprofen via a diamide bond, for evaluating its potential to antagonize the deleterious structural and cognitive effects of β-amyloid (1–40) in an infused Alzheimer's disease rat model. Our results indicated that infusion of β-amyloid (1–40) impairs memory performance through a progressive cognitive deterioration; however, ibuprofen and co-drug 1 seemed to protect against behavioural detriment induced by simultaneous administration of β-amyloid (1–40) protein. The obtained data were supported by the histochemical findings of the present study: β-amyloid protein was less expressed in 1-treated than in ibuprofen and (R)-α-lipoic acid alone-treated cerebral cortex. Taken together, the present findings suggest that co-drug 1 treatment may protect against the cognitive dysfunction induced by intracerebroventricular infusion of β-amyloid (1–40) in rats. Thus, co-drug 1 could prove useful as a tool for controlling Alzheimer's disease-induced cerebral amyloid deposits and behavioural deterioration

    Identification of two proteins that interact with the Erp virulence factor from Mycobacterium tuberculosis by using the bacterial two-hybrid system

    <p>Abstract</p> <p>Background</p> <p>The exported repetitive protein (<it>erp</it>) gene encodes a secreted 36-kDa protein with a central domain containing several proline-glycine-leucine-threonine-serine (PGLTS) repeats. It has been demonstrated that <it>erp </it>is a virulence-associated factor since the disruption of this gene impairs the growth of <it>Mycobacterium bovis </it>and <it>Mycobacterium tuberculosis </it>in mice.</p> <p>Results</p> <p>In order to elucidate the function of Erp we searched for Erp-binding proteins from <it>M. tuberculosis </it>by using a bacterial two-hybrid system. Our results indicate that Erp interacts specifically with two putative membrane proteins, Rv1417 and Rv2617c. Further analysis revealed that the latter two interact with each other, indicating that Rv1417, Rv2617c and Erp are connected through multiple interactions. While Rv1417 is disseminated in several <it>Actinomycetales </it>genera, orthologues of Rv2617c are exclusively present in members of the <it>M. tuberculosis </it>complex (MTC). The central and amino-terminal regions of Erp were determined to be involved in the interaction with Rv1417 and Rv2627c. Erp forms from <it>Mycobacterium smegmatis </it>and <it>Mycobacterium leprae </it>were not able to interact with Rv2617c in two-hybrid assays. Immunolocalization experiments showed that Rv1417 and Rv2617c are found on the cell membrane and Erp on the bacterial cell wall. Finally, comparative genomics and expression studies revealed a possible role of Rv1417 in riboflavin metabolism.</p> <p>Conclusion</p> <p>We identified interactive partners of Erp, an <it>M. tuberculosis </it>protein involved in virulence, which will be the focus of future investigation to decipher the function of the Erp family protein.</p

    Study of the role of Mce3R on the transcription of mce genes of Mycobacterium tuberculosis

    <p>Abstract</p> <p>Background</p> <p><it>mce3 </it>is one of the four virulence-related <it>mce </it>operons of <it>Mycobacterium tuberculosis</it>. In a previous work we showed that the overexpression of Mce3R in <it>Mycobacterium smegmatis </it>and <it>M. tuberculosis </it>abolishes the expression of <it>lacZ </it>fused to the <it>mce3 </it>promoter, indicating that Mce3R represses <it>mce3 </it>transcription.</p> <p>Results</p> <p>We obtained a knockout mutant strain of <it>M. tuberculosis </it>H37Rv by inserting a hygromycin cassette into the <it>mce3R </it>gene. The mutation results in a significant increase in the expression of <it>mce3 </it>genes either <it>in vitro </it>or in a murine cell macrophages line as it was determined using promoter-<it>lacZ </it>fusions in <it>M. tuberculosis</it>. The abundance of <it>mce1</it>, <it>mce2 </it>and <it>mce4 </it>mRNAs was not affected by this mutation as it was demonstrated by quantitative RT-PCR. The <it>mce3R </it>promoter activity in the presence of Mce3R was significantly reduced compared with that in the absence of the regulator, during the <it>in vitro </it>culture of <it>M. tuberculosis</it>.</p> <p>Conclusion</p> <p>Mce3R repress the transcription of <it>mce3 </it>operon and self regulates its own expression but does not affect the transcription of <it>mce1</it>, <it>mce2 </it>and <it>mce4 </it>operons of <it>M. tuberculosis</it>.</p
