    The flavonoid dioclein reduces the production of pro-inflammatory mediators in vitro by inhibiting PDE4 activity and scavenging reactive oxygen species

    Reactive oxygen species, cytokines and chemokines produced at inflammatory sites are pivotal events in the progression of many diseases. Flavonoids are well-known for their antioxidant and anti-inflammatory activities. Here, we investigated the effects of the flavonoid dioclein on the production of mediators of inflammation in vitro and possible underlying mechanisms. Murine macrophages were pretreated with dioclein, rolipram, a PDE4 (cyclic nucleotide phosphosdiesterase type 4) inhibitor, or butylated hydroxytoluene (BHT), an antioxidant, and then activated with LPS or LPS/IFN-gamma. The concentration of TNF-alpha, IL-6, CXCL1/KC, CCL2/JE, and nitric oxide (NO) was determined on culture supernatants. To evaluate potential mechanisms of action, dioclein was tested for inhibition of PDE4 activity and for antioxidant properties by chemiluminescence assays. Dioclein was efficient in reducing the production of cytokines, chemokines and NO in a concentration-dependent manner (from 5 to 50 mu M). Dioclein was more effective than BHT and rolipram, while having similar inhibitory effects to the combination of BHT plus rolipram. Dioclein inhibited PDE4 activity with an approximate IC(50) of 16.8 +/- 1.4 mu M and strongly reduced the concentration of reactive oxygen species in cell and cell-free systems, being more effective than the standard antioxidant BHT. The flavonoid dioclein possesses significant antioxidant and PDE4 inhibitory activity, showing that the substance may have substantial advantages over mechanisms of action already described for many flavonoids. Such effects account for the anti-inflammatory effects of dioclein, mainly by reducing the concentration of mediators of inflammation, such as cytokines, chemokines and reactive oxygen species by macrophages.     O objetivo deste trabalho foi avaliar a influência da temperatura de armazenamento e do uso do filme de cloreto de polivinila (PVC) sobre a perda de matéria fresca e água, incidência de danos causados por frio e metabolismo pós-colheita dos carboidratos, em raízes tuberosas de mandioquinha-salsa (Arracacia xanthorrhiza). O filme de PVC reduziu a perda de matéria fresca e manteve o teor de água das raízes, durante o armazenamento por 60 dias a 5 e 10ºC. Os danos causados por frio foram inibidos nas raízes embaladas em filme de PCV, em ambas as temperaturas de armazenamento. As baixas temperaturas induziram o acúmulo de açúcares solúveis e a degradação de amido e, para as raízes armazenadas sem PVC, o aumento do conteúdo dos açúcares solúveis foi transiente e a taxa de degradação de amido foi superior à das raízes armazenadas com PVC.<br>The objective of this work was to evaluate the influence of the storage temperature and stretch polyvinylchloride (PVC) film on the loss of fresh weight and water, on the development of chilling injury symptoms, and on the postharvest metabolism of carbohydrates, in arracacha tuber roots (Arracacia xanthorrhiza). The PVC film reduced the fresh weight loss and kept water content in the roots during 60-day storage period at 5 and 10ºC. PVC film in both storage temperatures inhibited the development of external and internal chilling injury symptoms. The low temperatures induced the increase of soluble sugar content and decrease of starch concentration, where the increase in soluble sugar was transient in roots stored without PVC film, and the rate of starch degradation was higher compared to the roots stored with PVC

    Recent studies on coffee (Coffea arabica L.) cultivation in agroforestry systems in Southern Brazil have shown the potential of partial shading to improve management of this crop. The objective of this work was to evaluate microclimatic conditions and their effects on coffee production of plants shaded with pigeon pea (Cajanus cajan) in comparison to unshaded ones, from May 2001 to August 2002 in Londrina, State of Paraná, Brazil. The appraised microclimatic characteristics were: global radiation, photosynthetic and radiation balance; air, leaf and soil temperatures; and soil humidity. Shading caused significant reduction in incident global solar radiation, photosynthetically active radiation and net radiation, and attenuated maximum leaf, air and soil temperatures, during the day. Shade also reduced the rate of cooling of night air and leaf temperatures, especially during nights with radiative frost. Soil moisture at 0-10 cm depth was higher under shade. The shaded coffee plants produced larger cherries due to slower maturation, resulting in larger bean size. Nevertheless, plants under shade emitted less plagiotropic branches, with smaller number of nodes per branch, and fewer nodes with fruits, resulting in a large reduction in coffee production. These results show the need to find an optimal tree density and management that do not compromise coffee production and protect against extreme temperatures.<br>Recentes estudos sobre cafeeiros (Coffea arabica L.) cultivados em sistemas agroflorestais no Sul do Brasil têm mostrado o potencial do sombreamento parcial no manejo desta cultura. O objectivo deste trabalho foi avaliar as condições do microclima e seus efeitos na produção de café sombreado com guandu (Cajanus cajan), em comparação ao cultivado a pleno sol, no período de maio de 2001 a agosto de 2002 em Londrina, PR. As características microclimáticas avaliadas foram: radiação global, fotossintética e saldo de radiação; temperaturas do ar, da folha e do solo; e umidade do solo. O sombreamento causou significativa redução na radiação solar global incidente, na radiação fotossinteticamente ativa e na radiação líquida, e atenuou as temperaturas máximas foliar, do ar e do solo, durante o dia. O sombreamento também evitou o declínio acentuado, durante a noite, das temperaturas do ar e da folha, especialmente durante noites com geadas de radiação. Os valores de umidade do solo, na camada 0-10 cm de profundidade, foram maiores nos cafeeiros sombreados. As plantas sombreadas produzem frutos maiores em virtude da maturação mais lenta, entretanto, emitem menos ramos plagiotrópicos, com menor número de nós por ramo e poucos nós com frutos, o que resulta na redução da produção de café. Estes resultados evidenciam a necessidade de se definir uma densidade ótima de sombreamento, que não comprometa a produção e proteja contra as temperaturas extremas
