    Translation of findings from laboratory studies of food and alcohol intake into behavior change interventions : the experimental medicine approach.

    Objectives: Laboratory studies have contributed important information about the determinants of food and alcohol intake, and they have prompted the development of behavior change interventions that have been evaluated in randomized controlled trials conducted in the field. In this article we apply a recent experimental medicine (EM) framework to this body of research. Method: A conceptual review and focused discussion of the relevant literature is presented. Results: We illustrate how it is possible to translate findings from studies of food and alcohol intake in the laboratory into interventions that are effective for changing behavior in the real world. We go on to demonstrate how systematic failures can occur at different stages within the EM framework, and how these failures ultimately result in interventions that are ineffective for changing behavior. We also consider methodological issues that may constrain the external validity of findings from laboratory studies including demand effects, participant characteristics, and the timing and dose of behavioral interventions. Throughout, we make recommendations to improve the translation of findings from laboratory studies into behavior change interventions that are effective in the field. Conclusions: Consideration of the EM framework will help to ensure that promising candidate interventions for eating and drinking that are identified in laboratory studies can fulfill their translational promise

    Corrections to the Central Limit Theorem for Heavy-Tailed Probability Densities

    Classical Edgeworth expansions provide asymptotic correction terms to the Central Limit Theorem (CLT) up to an order that depends on the number of moments available. In this paper, we provide subsequent correction terms beyond those given by a standard Edgeworth expansion in the general case of regularly varying distributions with diverging moments (beyond the second). The subsequent terms can be expressed in a simple closed form in terms of certain special functions (Dawson's integral and parabolic cylinder functions), and there are qualitative differences depending on whether the number of moments available is even, odd or not an integer, and whether the distributions are symmetric or not. If the increments have an even number of moments, then additional logarithmic corrections must also be incorporated in the expansion parameter. An interesting feature of our correction terms for the CLT is that they become dominant outside the central region and blend naturally with known large-deviation asymptotics when these are applied formally to the spatial scales of the CLT

    Vortex microavalanches in superconducting Pb thin films

    Local magnetization measurements on 100 nm type-II superconducting Pb thin films show that flux penetration changes qualitatively with temperature. Small flux jumps at the lowest temperatures gradually increase in size, then disappear near T = 0.7Tc. Comparison with other experiments suggests that the avalanches correspond to dendritic flux protrusions. Reproducibility of the first flux jumps in a decreasing magnetic field indicates a role for defect structure in determining avalanches. We also find a temperature-independent final magnetization after flux jumps, analogous to the angle of repose of a sandpile.Comment: 6 pages, 5 figure

    Arbuscular mycorrhizal fungal‐induced tolerance is determined by fungal identity and pathogen density

    Societal Impact Statement Plant-parasitic nematodes are a major concern for global food security, and many existing control options are being phased out due to adverse impacts on the environment. Here, we show that although application of arbuscular mycorrhizal fungi (AMF) increases host tolerance to these parasites, these benefits decrease as the parasite burden increases, limiting long-term benefits. This effect was consistent between experiments in the glasshouse and in the field environment, demonstrating the relevance of research into usable technologies. Our findings have potential to aid decision making regarding application of AMF inocula for optimum results in agricultural systems. Summary Plant-parasitic nematodes are a leading global threat to crop production and food security aims. Control strategies based on nematicides and fertilisers are increasingly undesirable due to economic and environmental impacts. Arbuscular mycorrhizal fungi (AMF) may induce host tolerance against pests such as the potato cyst nematode (PCN). Here, we determined the impact of PCN density on the tolerance induced by AMF-host interactions. Additionally, we evaluated the effects of five AMF inocula on PCN fitness though glasshouse and field trials. Greater PCN densities reduce the increased tolerance that AMF may confer on their hosts. This may be due to reduced mycorrhizal colonisation of hosts under higher PCN infection and potentially a threshold at which the presence of PCN severely impacts fungal growth. When tested in the field, the outcomes of AMF inoculation on crop yields were still positive. Inoculation of soil in the field also increased PCN multiplication, suggesting that AMF-induced tolerance may become reduced in the near future when the threshold PCN density is reached. Addition of AMF to agricultural soils may provide a short-term benefit yet lead to a long-term detriment by increasing PCN populations. The effects observed were driven by only one out of the five introduced AMF species, indicating that the remaining species were redundant for this application. This raises important considerations for future application of AMF inocula in agricultural systems and aids our understanding of how widely used ‘beneficial’ soil amendments impact the agricultural ecosystem

    Interleukin-7 deficiency in rheumatoid arthritis: consequences for therapy-induced lymphopenia

    We previously demonstrated prolonged, profound CD4+ T-lymphopenia in rheumatoid arthritis (RA) patients following lymphocyte-depleting therapy. Poor reconstitution could result either from reduced de novo T-cell production through the thymus or from poor peripheral expansion of residual T-cells. Interleukin-7 (IL-7) is known to stimulate the thymus to produce new T-cells and to allow circulating mature T-cells to expand, thereby playing a critical role in T-cell homeostasis. In the present study we demonstrated reduced levels of circulating IL-7 in a cross-section of RA patients. IL-7 production by bone marrow stromal cell cultures was also compromised in RA. To investigate whether such an IL-7 deficiency could account for the prolonged lymphopenia observed in RA following therapeutic lymphodepletion, we compared RA patients and patients with solid cancers treated with high-dose chemotherapy and autologous progenitor cell rescue. Chemotherapy rendered all patients similarly lymphopenic, but this was sustained in RA patients at 12 months, as compared with the reconstitution that occurred in cancer patients by 3–4 months. Both cohorts produced naïve T-cells containing T-cell receptor excision circles. The main distinguishing feature between the groups was a failure to expand peripheral T-cells in RA, particularly memory cells during the first 3 months after treatment. Most importantly, there was no increase in serum IL-7 levels in RA, as compared with a fourfold rise in non-RA control individuals at the time of lymphopenia. Our data therefore suggest that RA patients are relatively IL-7 deficient and that this deficiency is likely to be an important contributing factor to poor early T-cell reconstitution in RA following therapeutic lymphodepletion. Furthermore, in RA patients with stable, well controlled disease, IL-7 levels were positively correlated with the T-cell receptor excision circle content of CD4+ T-cells, demonstrating a direct effect of IL-7 on thymic activity in this cohort

    The Frequency Dependence of Critical-velocity Behavior in Oscillatory Flow of Superfluid Helium-4 Through a 2-micrometer by 2-micrometer Aperture in a Thin Foil

    The critical-velocity behavior of oscillatory superfluid Helium-4 flow through a 2-micrometer by 2-micrometer aperture in a 0.1-micrometer-thick foil has been studied from 0.36 K to 2.10 K at frequencies from less than 50 Hz up to above 1880 Hz. The pressure remained less than 0.5 bar. In early runs during which the frequency remained below 400 Hz, the critical velocity was a nearly-linearly decreasing function of increasing temperature throughout the region of temperature studied. In runs at the lowest frequencies, isolated 2 Pi phase slips could be observed at the onset of dissipation. In runs with frequencies higher than 400 Hz, downward curvature was observed in the decrease of critical velocity with increasing temperature. In addition, above 500 Hz an alteration in supercritical behavior was seen at the lower temperatures, involving the appearance of large energy-loss events. These irregular events typically lasted a few tens of half-cycles of oscillation and could involve hundreds of times more energy loss than would have occurred in a single complete 2 Pi phase slip at maximum flow. The temperatures at which this altered behavior was observed rose with frequency, from ~ 0.6 K and below, at 500 Hz, to ~ 1.0 K and below, at 1880 Hz.Comment: 35 pages, 13 figures, prequel to cond-mat/050203

    Robust exponential decay of correlations for singular-flows

    We construct open sets of Ck (k bigger or equal to 2) vector fields with singularities that have robust exponential decay of correlations with respect to the unique physical measure.     On-disk coronal rain

    Small and elongated, cool and dense blob-like structures are being reported with high resolution telescopes in physically different regions throughout the solar atmosphere. Their detection and the understanding of their formation, morphology and thermodynamical characteristics can provide important information on their hosting environment, especially concerning the magnetic field, whose understanding constitutes a major problem in solar physics. An example of such blobs is coronal rain, a phenomenon of thermal non- equilibrium observed in active region loops, which consists of cool and dense chromospheric blobs falling along loop-like paths from coronal heights. So far, only off-limb coronal rain has been observed and few reports on the phenomenon exist. In the present work, several datasets of on-disk H{\alpha} observations with the CRisp Imaging SpectroPolarimeter (CRISP) at the Swedish 1-m Solar Telescope (SST) are analyzed. A special family of on-disk blobs is selected for each dataset and a statistical analysis is carried out on their dynamics, morphology and temperatures. All characteristics present distributions which are very similar to reported coronal rain statistics. We discuss possible interpretations considering other similar blob-like structures reported so far and show that a coronal rain interpretation is the most likely one. Their chromospheric nature and the projection effects (which eliminate all direct possibility of height estimation) on one side, and their small sizes, fast dynamics, and especially, their faint character (offering low contrast with the background intensity) on the other side, are found as the main causes for the absence until now of the detection of this on-disk coronal rain counterpart.Comment: 18 pages, 10 figures.     Effects of coastal urbanization on salt-marsh faunal assemblages in the northern Gulf of Mexico

We used a landscape ecology approach to quantify the composition and configuration of salt-marsh habitats and developed surfaces at multiple spatial scales surrounding three small, first-order salt-marsh tidal creeks arrayed along a gradient of urbanization in two river-dominated estuaries. From May 3 to June 4, 2010, nekton and macroinfauna were collected weekly at all six sites. Due to the greater abundance of grass shrimp Palaemonetes spp., brown shrimp Farfantepenaeus aztecus, blue crab Callinectes sapidus, Gulf Menhaden Brevoortia patronus, and Spot Leiostomus xanthurus, tidal creeks in intact natural (IN) salt-marsh landscapes supported a nekton assemblage that was significantly different from those in partially urbanized (PU) or completely urbanized (CU) salt-marsh landscapes. However, PU landscapes still supported an abundant nekton assemblage. In addition, the results illustrated a linkage between life history traits and landscape characteristics. Resident and transient nekton species that have specific habitat requirements are more likely to be impacted in urbanized landscapes than more mobile species that are able to exploit multiple habitats. Patterns were less clear for macroinfaunal assemblages, although they were comparatively less abundant in CU salt-marsh landscapes than in either IN or PU landscapes. The low abundance or absence of several macroinfaunal taxa in CU landscapes may be viewed as an additional indicator of poor habitat quality for nekton. The observed patterns also suggested that benthic sediments in the CU salt-marsh landscapes were altered in comparison with IN or PU landscapes. The amount of developed shoreline and various metrics related to salt marsh fragmentation were important drivers of observed patterns in nekton and macroinfaunal assemblages
