    Ion conduction through potassium channels is a fundamental process of life. On the basis of crystallographic data, it was originally proposed that potassium ions and water molecules are transported through the selectivity filter in an alternating arrangement, suggesting a "water-mediated" knock-on mechanism. Later on, this view was challenged by results from molecular dynamics simulations that revealed a "direct" knock-on mechanism where ions are in direct contact. Using solid-state nuclear magnetic resonance techniques tailored to characterize the interaction between water molecules and the ion channel, we show here that the selectivity filter of a potassium channel is free of water under physiological conditions. Our results are fully consistent with the direct knock-on mechanism of ion conduction but contradict the previously proposed water-mediated knock-on mechanism

    Exploring Entrepreneurship Education Effectiveness at British Universities – An Application of the World Café Method

    Universities have made significant investments in entrepreneurship programs for decades, but the scope, relevance and usefulness of entrepreneurship education are still questioned. This study aims to explore the meaning of effectiveness as it relates to entrepreneurship education in a grounded and holistic sense, recognizing both the range of stakeholders involved in the design, delivery and experience of entrepreneurship education and the underlying complexity of the issue at hand. Two World Café events, a method designed to elicit grounded knowledge, were organized to seek insights from a diverse range of stakeholders. Results confirm and illustrate the complex nature of effectiveness in entrepreneurship education. The purpose of specific educational initiatives, diverse audiences’ expectations and contextual factors must be considered in any meaningful attempt at identifying effectiveness. Findings also revealed a consensus that effectiveness relates to creating a transformational process, which leads to a shift in attitudes towards entrepreneurship. This shift prepares students for careers that go beyond the launch of a new venture. The role of time lags in assessing effectiveness was also identified. We suggest an agenda for future research and practical implications

    Epizootiologists recurrently encounter symbionts and pathobionts in the haemolymph (blood equivalent) of shellfish. One such group is the dinoflagellate genus , which contains several species that cause debilitating disease in decapod crustaceans. The shore crab acts as a mobile reservoir of microparasites, including sp., thereby posing a risk to other co-located commercially important species, e.g. velvet crabs ( ). Despite the widespread prevalence and documented seasonality of infection dynamics, there is a knowledge gap regarding host-pathogen antibiosis, namely, how avoids the host's immune defences. Herein, we interrogated the haemolymph of -positive and -negative crabs for extracellular vesicle (EV) profiles (a proxy for cellular communication), alongside proteomic signatures for post-translational citrullination/deimination performed by arginine deiminases, which can infer a pathologic state. Circulating EV numbers in parasitized crab haemolymph were reduced significantly, accompanied by smaller EV modal size profiles (albeit non-significantly) when compared to -negative controls. Differences were observed for citrullinated/deiminated target proteins in the haemolymph between the parasitized and control crabs, with fewer hits identified overall in the former. Three deiminated proteins specific to parasitized crab haemolymph were actin, Down syndrome cell adhesion molecule (DSCAM), and nitric oxide synthase - factors that contribute to innate immunity. We report, for the first time, sp. could interfere with EV biogenesis, and that protein deimination is a putative mechanism of immune-modulation in crustacean- interactions

    The adjoint problem in the presence of a deformed surface: the example of the Rosensweig instability on magnetic fluids

    The Rosensweig instability is the phenomenon that above a certain threshold of a vertical magnetic field peaks appear on the free surface of a horizontal layer of magnetic fluid. In contrast to almost all classical hydrodynamical systems, the nonlinearities of the Rosensweig instability are entirely triggered by the properties of a deformed and a priori unknown surface. The resulting problems in defining an adjoint operator for such nonlinearities are illustrated. The implications concerning amplitude equations for pattern forming systems with a deformed surface are discussed.Comment: 11 pages, 1 figur

    The aerological flights at Boston are part of the general research program of the Meteorological Division of Massachusetts Institute of Technology, which program since 1929 has been directed especially toward the study of American air masses and fronts. Recently, some results of these studies were published by Willett, who based his investigations on a series continuous over three years of mornìng and evening weather maps, analyzed at the Institute, together with upper air soundings from the United States Weather Bureau stations at Dallas, Omaha, Chicago, Groesbeck, Atlanta and from the United States Navy at Seattle, Anacostia, Pensacola and San Diego. These upper air data facilitated the determination of the properties of the air masses and so proved of inestimable value for the study. But the use of the data also showed that improvement both in the number of stations and in the quality of observations was highly desirable. Ascents in the northeastern part of the United States were lacking. Knowledge of the vertical structure of air masses reaching this region, however, is of special interest in forecasting for this densely populated district. For these reasons and since the direct comparison of actual local weather developments with upper air conditions is also con- sidered to be very valuable, the Institute started its own airplane station at Boston. In addition to "regular" ascents at the time of the morning surface observations, special flights were made when particularly interesting weather situations prevailed. On a number of days series of ascents were carried out to obtain cross sections through fronts passing over Boston. Other special flights were made to obtain information on atmospheric turbulence. For this same purpose and also in order to study the diurnal changes of temperature in the lowest 5,000 feet, several series are planned of a number of comparatively low altitude flights at short intervals throughout the day

    Maturing Satellite Kinematics into a Competitive Probe of the Galaxy-Halo Connection

    The kinematics of satellite galaxies moving in a dark matter halo are a direct probe of the underlying gravitational potential. Thus, the phase-space distributions of satellites represent a powerful tool to determine the galaxy-halo connection from observations. By stacking the signal of a large number of satellite galaxies this potential can be unlocked even for haloes hosting a few satellites on average. In this work, we test the impact of various modelling assumptions on constraints derived from analysing satellite phase-space distributions in the non-linear, 1-halo regime. We discuss their potential to explain the discrepancy between average halo masses derived from satellite kinematics and gravitational lensing previously reported. Furthermore, we develop an updated, more robust analysis to extract constraints on the galaxy-halo relation from satellite properties in spectroscopic galaxy surveys such as the SDSS. We test the accuracy of this approach using a large number of realistic mock catalogues. Furthermore, we find that constraints derived from such an analysis are complementary and competitive with respect to the commonly used galaxy clustering and galaxy-galaxy lensing observables.Comment: 24 pages, 15 figures; resubmitted to MNRAS after first referee repor

    Updated Results on the Galaxy-Halo Connection from Satellite Kinematics in SDSS

    We present new results on the relationship between central galaxies and dark matter haloes inferred from observations of satellite kinematics in the Sloan Digital Sky Survey (SDSS) DR7. We employ an updated analysis framework that includes detailed mock catalogues to model observational effects in SDSS. Our results constrain the colour-dependent conditional luminosity function (CLF) of dark matter haloes, as well as the radial profile of satellite galaxies. Confirming previous results, we find that red central galaxies live in more massive haloes than blue galaxies at fixed luminosity. Additionally, our results suggest that satellite galaxies have a radial profile less centrally concentrated than dark matter but not as cored as resolved subhaloes in dark matter-only simulations. Compared to previous works using satellite kinematics by More et al., we find much more competitive constraints on the galaxy-halo connection, on par with those derived from a combination of galaxy clustering and galaxy-galaxy lensing. We compare our results on the galaxy-halo connection to other studies using galaxy clustering and group catalogues, showing very good agreement between these different techniques.     Via Hexagons to Squares in Ferrofluids: Experiments on Hysteretic Surface Transformations under Variation of the Normal Magnetic Field

    We report on different surface patterns on magnetic liquids following the Rosensweig instability. We compare the bifurcation from the flat surface to a hexagonal array of spikes with the transition to squares at higher fields. From a radioscopic mapping of the surface topography we extract amplitudes and wavelengths. For the hexagon--square transition, which is complex because of coexisting domains, we tailor a set of order parameters like peak--to--peak distance, circularity, angular correlation function and pattern specific amplitudes from Fourier space. These measures enable us to quantify the smooth hysteretic transition. Voronoi diagrams indicate a pinning of the domains. Thus the smoothness of the transition is roughness on a small scale.Comment: 17 pages, 14 figure

    COLOR III: a multicentre randomised clinical trial comparing transanal TME versus laparoscopic TME for mid and low rectal cancer

    Total mesorectal excision (TME) is an essential component of surgical management of rectal cancer. Both open and laparoscopic TME have been proven to be oncologically safe. However, it remains a challenge to achieve complete TME with clear circumferential resections margin (CRM) with the conventional transabdominal approach, particularly in mid and low rectal tumours. Transanal TME (TaTME) was developed to improve oncological and functional outcomes of patients with mid and low rectal cancer.An international, multicentre, superiority, randomised trial was designed to compare TaTME and conventional laparoscopic TME as the surgical treatment of mid and low rectal carcinomas. The primary endpoint is involved CRM. Secondary endpoints include completeness of mesorectum, residual mesorectum, morbidity and mortality, local recurrence, disease-free and overall survival, percentage of sphincter-saving procedures, functional outcome and quality of life. A Quality Assurance Protocol including centralised MRI review, histopathology re-evaluation, standardisation of surgical techniques, and monitoring and assessment of surgical quality will be conducted.The difference in involvement of CRM between the two treatment strategies is thought to be in favour of the TaTME. TaTME is therefore expected to be superior to laparoscopic TME in terms of oncological outcomes in case of mid and low rectal carcinomas
