    Ixodes brunneus (Acari: Ixodidae) from Two Bird Hosts: A New Michigan Tick

    The tick Ixodes brunneus Koch (Acari: Ixodidae) is reported for the first time in Michigan from two bird hosts at two locations in the lower peninsula. All stages of this tick exclusively feed on birds, and are primarily known from the southern U.S., although abundant records occur from northern states. The role of this species as a vector of pathogens is discussed

    Generation of cell lines to complement Adenovirus vectors using recombination-mediated cassette exchange

    Background Adenovirus serotype 5 (Ad5) has many favourable characteristics for development as a gene therapy vector. However, the utility of current Ad5 vectors is limited by transient transgene expression, toxicity and immunogenicity. The most promising form of vector is the high capacity type, which is deleted for all viral genes. However, these vectors can only be produced to relatively low titres and with the aid of helper virus. Therefore a continuing challenge is the generation of more effective Ad5 vectors that can still be grown to high titres. Our approach is to generate complementing cell lines to support the growth of Ad5 vectors with novel late gene deficiencies. Results We have used LoxP/Cre recombination mediated cassette exchange (RMCE) to generate cell lines expressing Ad5 proteins encoded by the L4 region of the genome, the products of which play a pivotal role in the expression of Ad5 structural proteins. A panel of LoxP parent 293 cell lines was generated, each containing a GFP expression cassette under the control of a tetracycline-regulated promoter inserted at a random genome location; the cassette also contained a LoxP site between the promoter and GFP sequence. Clones displayed a variety of patterns of regulation, stability and level of GFP expression. Clone A1 was identified as a suitable parent for creation of inducible cell lines because of the tight inducibility and stability of its GFP expression. Using LoxP-targeted, Cre recombinase-mediated insertion of an L4 cassette to displace GFP from the regulated promoter in this parent clone, cell line A1-L4 was generated. This cell line expressed L4 100K, 22K and 33K proteins at levels sufficient to complement L4-33K mutant and L4-deleted viruses. Conclusions RMCE provides a method for rapid generation of Ad5 complementing cell lines from a pre-selected parental cell line, chosen for its desirable transgene expression characteristics. Parent cell lines can be selected for high or low gene expression, and for tight regulation, allowing viral protein expression to mirror that found during infection. Cell lines derived from a single parent will allow the growth of different vectors to be assessed without the complication of varying complementing protein expression

    Fixation of Juvenile Cambium from Two Coniferous Species for Ultrastructural Study

    Samples of dormant and of actively growing cambial-zone tissue collected in June and March, respectively, from plantations of young white spruce (Picea glauca [Moench] Voss) and red pine (Pinus resinosa Ait.) near Ottawa, Canada, were fixed in five different solutions at three temperatures. Fixation quality was evaluated by electron microscopy.Not all cellular organelles were preserved equally well by the same fixative in the active and in the dormant conditions, or in both spruce and pine. In general, our best results were obtained with Karnovsky's fixative solution. Somewhat less satisfactory results were obtained with a glutaraldehyde-acrolein mixture followed by straight glutaraldehyde. Poorer results were obtained with a trialdehyde solution while a glutaraldehyde-osmium tetroxide cocktail undoubtedly provided the worst fixation.Different fixative temperatures had a marked effect on fixation quality only when phosphate buffer was used. There was little gained by prolonging the period of fixation beyond a few hours

    Parallel computations and control of adaptive structures

    The equations of motion for structures with adaptive elements for vibration control are presented for parallel computations to be used as a software package for real-time control of flexible space structures. A brief introduction of the state-of-the-art parallel computational capability is also presented. Time marching strategies are developed for an effective use of massive parallel mapping, partitioning, and the necessary arithmetic operations. An example is offered for the simulation of control-structure interaction on a parallel computer and the impact of the approach presented for applications in other disciplines than aerospace industry is assessed

    Phenomenological Constraints on Axion Models of Dynamical Dark Matter

    In two recent papers (arXiv:1106.4546, arXiv:1107.0721), we introduced "dynamical dark matter" (DDM), a new framework for dark-matter physics in which the requirement of stability is replaced by a delicate balancing between lifetimes and cosmological abundances across a vast ensemble of individual dark-matter components whose collective behavior transcends that normally associated with traditional dark-matter candidates. We also presented an explicit model involving axions in large extra spacetime dimensions, and demonstrated that this model has all of the features necessary to constitute a viable realization of the general DDM framework. In this paper, we complete our study by performing a general analysis of all phenomenological constraints which are relevant to this bulk-axion DDM model. Although the analysis in this paper is primarily aimed at our specific DDM model, many of our findings have important implications for bulk axion theories in general. Our analysis can also serve as a prototype for phenomenological studies of theories in which there exist large numbers of interacting and decaying particles.Comment: 48 pages, LaTeX, 13 figures, 1 tabl

    Measurements at low energies of the polarization-transfer coefficient Kyy' for the reaction 3H(p,n)3He at 0 degrees

    Measurements of the transverse polarization coefficient Kyy' for the reaction 3H(p,n)3He are reported for outgoing neutron energies of 1.94, 5.21, and 5.81 MeV. This reaction is important both as a source of polarized neutrons for nuclear physics experiments, and as a test of theoretical descriptions of the nuclear four-body system. Comparison is made to previous measurements, confirming the 3H(p,n)3He reaction can be used as a polarized neutron source with the polarization known to an accuracy of approximately 5%. Comparison to R-matrix theory suggests that the sign of the 3F3 phase-shift parameter is incorrect. Changing the sign of this parameter dramatically improves the agreement between theory and experiment.Comment: 12 pages, RevTeX, 5 eps figures, submitted to Phys. Rev.

    The author has identified the following significant results. Characteristics of LANDSAT MSS imagery present problems in using satellite radiation measurements to estimate the shortwave albedo of an alpine snow cover. Every 15 minute USGS quadrangle contains over 100,000 pixels which poses a computation problem if each pixel is to be evaluated individually. The sampling interval may be sufficiently great to mask some effects of terrain and vegetation on reflectance. Three frames of LANDSAT imagery are needed for complete coverage of the study area, yet less than one third of the area coverage from each frame covers an area of interest. Because of distortions inherent in the imagery, information regarding spacecraft altitude, attitude, and position must be statistically derived with respect to ground control points in the image whose geodetic locations are known. An inspection of shade points indicates that up to one third of the most heavily snow covered areas may saturate in bands 4 through 6. LANDSAT's 9 day repeat cycle is not optimum for snow cover reflectance modeling because the most pronounced changes in albedo occur most nearly following a new snowfall. Such a snowfall, occurring between overpasses, is inadequately represented by extrapolation from the previous overpasses

    Employable knowledge: benchmarking education about standardization in the UK

    For academics and students in the United Kingdom the main source of standards is via British Standards Institution (BSI). The research demonstrates that British, European and International Standards play a key role in many areas of education. In some disciplines their inclusion in the course is mandatory, e.g. in building construction and performing risk assessments of equipment. Where not a requirement, other courses successfully encourage students to understand and apply specific standards and principles from them, to their design and project work, in topic areas such as quality management and user-computer interface design. Assessment practice is a key part of learning and academics have indicated how this fitted into the learning activity, e.g. by expecting students to develop an understanding of standards and reference them in all assessed work. Likewise, students taking part in the survey also suggest that they were actively engaged with one or more standards and that their understanding was measured through an assessed activity. However, students also emphasised the importance of additional support, e.g. introduction to standards in the workplace or as part of work placements, by library staff as an information resource, and by presentations from BSI experts. Employers have also indicated the importance of students’ knowledge, understanding and appreciation of relevant standards in the right context, emphasising their desire for standards to be included more widely in the curriculum. The draft recommendations from this study were subject to International review and comment, the results from this review served to strengthen the recommendations of this work.
