Targeting multidrug resistant Mycobacterium tuberculosis htra2 with identical chemical entities of fluoroquinolones

Tuberculosis is a highly communicable and chronic respiratory disease caused by pathogenic bacterium Mycobacterium tuberculosis. The drug - resistant species of Mycobacterium tuberculosis are tough to cure due to its resistant activity toward potential drugs. Available inhibitors of tuberculosis include few antimicrobial fluoroquinolone agents like ciprofloxacin, ofloxacin, and moxifloxacin to treat resistant Mycobacterium strains. Literature study elucidates that macromolecular target namely, HtrA2 of Mycobacterium tuberculosis play a dual role of protease and chaperone. These two activities are dependent on temperature, with low temperatures promoting the chaperone function and high temperatures promoting serine protease activity. Under normal physiological conditions HtrA2 acts as a quality control factor and promotes cell survival. In the present investigation, we screened fluoroquinolone such as ciprofloxacin, moxifloxacin and ofloxacin and their analogues based on better Docking score, absorption, distribution, metabolism and excretion screening and Lipinski′s rule of 5, to find out their efficiency on resistant strain through in silico study. From the results observed, the analogues are suggested to be potent inhibitors of HtrA2 with sufficient scope for further exploration

In this study, various plants like Datura metel, Aegle marmelos, Annona reticulata and Saraca indica were collected and subjected for extraction using various solvents, namely Water, Chloroform and Ethanol. The extracts were done with TLC bioautography for Antioxidant activity and Antibacterial activity, Minimum Inhibitory Concentration, Antibacterial activity, Antioxidant activity (DPPH and FRAP) and Phytochemical analysis. Plants used in this study showed antibacterial and antioxidant activity. These extracts were further utilized for Silver nanoparticle production and were characterized. Silver nanoparticles were utilized for In vitro antibacterial activity, where they did not show any antibacterial activity

Azadirachta indica, generally called as neem, margosa or Indian lilac is an Indian subcontinent native species, it ie well known for its various bioactivity In this study, neem leaves were collected and extracted. The extract was subjected for TLC and antibacterial activity against Pseudomonas aeruginosa. The extract was subjected for green synthesis of silver nanoparticles and it was able to reduce silver to silver nanoparticle of size around 65nm. The silver nanoparticle was also exhibiting antibacterial activity against P.aeruginosa

Measuring health-related quality of life (HRQOL) in HIV-positive individuals— Content analyses of measures based on the international classification of functioning, disability, and health (ICF) and generic and disability core set

© 2015 by Begell House, Inc. Purpose: The purpose of the study is to conduct a content analysis of the items of the Medical Outcome Study-HIV (MOS-HIV), the Multidimentional Quality of Life-HIV (MQOL-HIV), and the HIV Disability Questionnaire (HDQ) by linking these to the ICF and its generic and disability core set. Methods: Four raters individually linked 145 items from the measures based on standardized linking rules. Interrater agreement was determined. For items where there was no agreement among raters, the opinion of a fifth rater, an expert in applying the ICF, was obtained. Results: The items were linked to 74 ICF categories across the three measures. Final interrater agreement was 83.3%. The content of all three measures was highly linkable with ICF. The HDQ had the best representation among the three measures. The HDQ had 27 items linked with the generic and disability core set; of these 16 items were linked with disability codes. Discussion: The HDQ provides a more precise description of the disablement experienced by people living with HIV by addressing the body function, activities, participation, and environmental factors specific to people living with HIV. The HDQ is thus recommended for those who are interested in measuring HIV-specific disabilities more precisely

Preparation, characterization and utilization of coreshell super paramagnetic iron oxide nanoparticles for curcumin delivery.

In this study, super paramagnetic iron oxide nanoparticles (SPIONs) were produced by chemical co-precipitation method, then it was constructed to be a core shell nanoparticle by functionalizing with SDS, loading with curcumin and coating with a biopolymer i.e. chitosan. Each step was analyzed microscopically and spectroscopically. The produced coreshell particles were between 40 and 45nm and these coreshell particles were utilized for drug delivery studies against cervical cancer cell line-HeLa cells. The coreshell SPIONs were found to be releasing curcumin in between 6 and 12 h, which was evidenced by increased apoptotic cells and increased caspase 3 expression in HeLa cells

Metal nanoparticles found its application in most fields because of its different physiochemical properties. Although there are several methods for synthesis of nanoparticles, where green and biological synthesis of nanoparticles reduces the toxicity. These metal nanoparticles are not properly disposed to the environment. Thus, this study was focussed to evaluate the effects of Araucaria heterophylla mediated green synthesised silver nanoparticles on various in vivo models such as earth worm—Eudrilus eugeniae and zebra fish—Danio rerio. Even the biologically synthesised nanoparticles were showing negative impacts on these models. Silver nanoparticles was found to cause epithelial erosion and lipofucshin deposits on Eudrilus eugeniae, where it was causing erosion and structure deformation in guts of Danio rerio
