    Transport proteins determine drug sensitivity and resistance in a protozoan parasite, Trypanosoma brucei

    Drug resistance in pathogenic protozoa is very often caused by changes to the ‘transportome’ of the parasites. In Trypanosoma brucei, several transporters have been implicated in uptake of the main classes of drugs, diamidines and melaminophenyl arsenicals. The resistance mechanism had been thought to be due to loss of a transporter known to carry both types of agents: the aminopurine transporter P2, encoded by the gene TbAT1. However, although loss of P2 activity is well-documented as the cause of resistance to the veterinary diamidine diminazene aceturate (Berenil®), cross-resistance between the human-use arsenical melarsoprol and the diamidine pentamidine (MPXR) is the result of loss of a separate High Affinity Pentamidine Transporter (HAPT1). A genome-wide RNAi library screen for resistance to pentamidine, published in 2012, gave the key to the genetic identity of HAPT1 by linking the phenomenon to a locus that contains the closely related T. brucei aquaglyceroporin genes TbAQP2 and TbAQP3. Further analysis determined that knockdown of only one pore, TbAQP2, produced the MPXR phenotype. TbAQP2 is an unconventional aquaglyceroporin with unique residues in the “selectivity region” of the pore, and it was found that in several MPXR lab strains the WT gene was either absent or replaced by a chimeric protein, recombined with parts of TbAQP3. Importantly, wild-type AQP2 was also absent in field isolates of T. b. gambiense, correlating with the outcome of melarsoprol treatment. Expression of a wild-type copy of TbAQP2 in even the most resistant strain completely reversed MPXR and re-introduced HAPT1 function and transport kinetics. Expression of TbAQP2 in Leishmania mexicana introduced a pentamidine transport activity indistinguishable from HAPT1. Although TbAQP2 has been shown to function as a classical aquaglyceroporin it is now clear that it is also a high affinity drug transporter, HAPT1. We discuss here a possible structural rationale for this remarkable ability

    Projects for sewage outfall siting for pollution control in the lower Chesapeake Bay wetlands are reported. A dye-buoy/photogrammetry and remote sensing technique was employed to gather circulation data used in outfall siting. This technique is greatly favored over alternate methods because it is inexpensive, produces results quickly, and reveals Lagrangian current paths which are preferred in making siting decisions. Wetlands data were obtained by interpretation of color and color infrared photographic imagery from several altitudes. Historical sequences of photographs are shown that were used to document wetlands changes. Sequential infrared photography of inlet basins was employed to determine tidal prisms, which were input to mathematical models to be used by state agencies in pollution control. A direct and crucial link between remote sensing and management decisions was demonstrated in the various projects

    Remote sensing was used in the resolution of estuarine problems facing federal and Virginia governmental agencies. A prototype Elizabeth River Surface Circulation Atlas was produced from photogrammetry to aid in oil spill cleanup and source identification. Aerial photo analysis twice led to selection of alternative plans for dredging and spoil disposal which minimized marsh damage. Marsh loss due to a mud wave from a highway dyke was measured on sequential aerial photographs. An historical aerial photographic sequence gave basis to a potential Commonwealth of Virginia legal claim to accreting and migrating coastal islands

    Role of Schizosaccharomyces pombe RecQ homolog recombination and checkpoint genes in UV Damage tolerance

    The cellular responses to DNA damage are complex and include direct DNA repair pathways that remove the damage and indirect damage responses which allow cells to survive DNA damage that has not been, or cannot be, removed. We have identified the gene mutated in the rad12.502 strain as a Schizosaccharomyces pombe recQ homolog. The same gene (designated rqh1) is also mutated in the hus2.22 mutant. We show that Rqh1 is involved in a DNA damage survival mechanism which prevents cell death when UV-induced DNA damage cannot be removed. This pathway also requires the correct functioning of the recombination machinery and the six checkpoint tad gene products plus the Cds1 kinase. Our data suggest that Rqh1 operates during S phase as part of a mechanism which prevents DNA damage causing cell lethality. This process may involve the bypass of DNA damage sites by the replication fork. Finally, in contrast with the reported literature, we do not find that rqh1 (rad12) mutant cells are defective in UV dimer endonuclease activity

    The regional employment returns from wave and tidal energy: A Welsh analysis

    The paper examines the expected regional employment returns connected to the development of tidal stream and wave-based electricity generation in a UK region - Wales. New employment provides economic development opportunities and the paper demonstrates that there is a need to develop the evidence base on these regional employment impacts. It addresses how far a region which is adjacent to significant marine resources is likely to benefit from a change in the energy generation mix which could feature more wave and tidal stream technologies

    We have reviewed the Comment of Geyer et al. [arXiv:0708.1548] concerning our recent work [Phys. Rev. A 75, 060102 (R) (2007)], and while we disagree with their criticisms, we acknowledge them for giving us the opportunity to add interesting addition material and a more detailed description of our experiment. We describe further our calculation and explain why a more sophisticated model is not warranted. We also present detailed experiments on the effects of electrostatic forces in our measurements and show that the contribution due to work function differences is small and that the residual electrostatic force is dominated by trapped charges and external fields. Finally, we estimate the effect of double layer interactions. These additional calculations and measurements support our original conclusion that the experimental results are consistent with the Lifshitz theory

    Priority sectors in city regions? Some issues from a study of the Cardiff Capital Region

    The City Region is becoming the spatial focus for economic development policy across many parts of the European continent. But these functional regions have taken on a new impetus in the UK with the introduction of ‘city deals’ aimed at improving network and coordination of actors in local authorities. One of the goals of city regions is to improve industrial policy particularly lacking since the abolition of many of the Regional Development Agencies across the UK. However, city regions in developing policy appear to be following in an unquestioning manner the industrial priorities of earlier institutions, and nowhere is this more obvious than in the case of the identification of priority industry development sectors. Too often the selection of industries and clusters for special support has been undertaken in an unquestioning manner. In this paper we focus on the case of the Cardiff Capital Region. We review approaches to identify priority sectors in this case and the problems associated with this policy approach
