The genome of polymorphonuclear neutrophils maintains normal coding sequences
Genetic studies often use genomic DNA from whole blood cells, of which the majority are the polymorphonuclear myeloid cells. Those cells undergo dramatic change of nuclear morphology following cellular differentiation. It remains elusive if the nuclear morphological change accompanies sequence alternations from the intact genome. If such event exists, it will cause a serious problem in using such type of genomic DNA for genetic study as the sequences will not represent the intact genome in the host individuals. Using exome sequencing, we compared the coding regions between neutrophil, which is the major type of polymorphonuclear cells, and CD4+ T cell, which has an intact genome, from the same individual. The results show that exon sequences between the two cell types are essentially the same. The minor differences represented by the missed exons and base changes between the two cell types were validated to be mainly caused by experimental errors. Our study concludes that genomic DNA from whole blood cells can be safely used for genetic studies
Effects of resveratrol supplementation on methotrexate chemotherapy‐induced bone loss
Intensive cancer chemotherapy is known to cause bone defects, which currently lack treatments. This study investigated the effects of polyphenol resveratrol (RES) in preventing bone defects in rats caused by methotrexate (MTX), a commonly used antimetabolite in childhood oncology. Young rats received five daily MTX injections at 0.75 mg/kg/day. RES was orally gavaged daily for seven days prior to, and during, five‐day MTX administration. MTX reduced growth plate thickness, primary spongiosa height, trabecular bone volume, increased marrow adipocyte density, and increased mRNA expression of the osteogenic, adipogenic, and osteoclastogenic factors in the tibial bone. RES at 10 mg/kg was found not to affect bone health in normal rats, but to aggravate the bone damage in MTX‐treated rats. However, RES supplementation at 1 mg/kg preserved the growth plate, primary spongiosa, bone volume, and lowered the adipocyte density. It maintained expression of genes involved in osteogenesis and decreased expression of adipogenic and osteoclastogenic factors. RES suppressed osteoclast formation ex vivo of bone marrow cells from the treated rats. These data suggest that MTX can enhance osteoclast and adipocyte formation and cause bone loss, and that RES supplementation at 1 mg/kg may potentially prevent these bone defects
Plasmoid ejection and secondary current sheet generation from magnetic reconnection in laser-plasma interaction
Reconnection of the self-generated magnetic fields in laser-plasma
interaction was first investigated experimentally by Nilson {\it et al.} [Phys.
Rev. Lett. 97, 255001 (2006)] by shining two laser pulses a distance apart on a
solid target layer. An elongated current sheet (CS) was observed in the plasma
between the two laser spots. In order to more closely model magnetotail
reconnection, here two side-by-side thin target layers, instead of a single
one, are used. It is found that at one end of the elongated CS a fan-like
electron outflow region including three well-collimated electron jets appears.
The ( MeV) tail of the jet energy distribution exhibits a power-law
scaling. The enhanced electron acceleration is attributed to the intense
inductive electric field in the narrow electron dominated reconnection region,
as well as additional acceleration as they are trapped inside the rapidly
moving plasmoid formed in and ejected from the CS. The ejection also induces a
secondary CS
Strong quantum fluctuation of vortices in the new superconductor

and the irreversibility line has been determined. A
big separation between and has been found showing the
existence of a quantum vortex liquid state induced by quantum fluctuation of
vortices in the new superconductor . Further investigation on the
magnetic relaxation shows that both the quantum tunneling and the thermally
activated flux creep weakly depends on temperature. But when the melting field
is approached, a drastic rising of the relaxation rate is observed.
This may imply that the melting of the vortex matter at a finite temperature is

Emerging and reemerging helminthiases and the public health of China.
Despite great strides in their control throughout the People's Republic of China, helminth infections remain an important public health problem. The Institute of Parasitic Diseases of the Chinese Academy of Preventive Medicine, under the guidance of the Chinese Ministry of Health, completed a nationwide survey of more than 1 million people that showed the high prevalence and intensity of intestinal nematode infections; prevalence can sometimes exceed 50% in the Yangtze River valley provinces. Schistosoma japonicum is also a major cause of illness in this region. Attempts to control Chinese helminthic diseases with conventional anthelminthic drugs have been partially thwarted by high posttreatment rates of reinfection. Recently, several new human trematode pathogens have been identified. Novel approaches to chemoprophylaxis and vaccination may alleviate the public health problem caused by Chinese helminths. However, recombinant helminth vaccine development will depend on first cataloguing the extensive genetic diversity of Chinese helminths and candidate vaccine antigens. Evidence from biogeography, genetics, and systematics suggests that the genetic diversification of Chinese helminths and their vectors is an ongoing evolutionary process that began 12 million years ago near the convergence of major Asian river systems. New fusion transcripts identified in normal karyotype acute myeloid leukemia
Genetic aberrations contribute to acute myeloid leukemia (AML). However, half of AML cases do not contain the well-known aberrations detectable mostly by cytogenetic analysis, and these cases are classified as normal karyotype AML. Different outcomes of normal karyotype AML suggest that this subgroup of AML could be genetically heterogeneous. But lack of genetic markers makes it difficult to further study this subgroup of AML. Using paired-end RNAseq method, we performed a transcriptome analysis in 45 AML cases including 29 normal karyotype AML, 8 abnormal karyotype AML and 8 AML without karyotype informaiton. Our study identified 134 fusion transcripts, all of which were formed between the partner genes adjacent in the same chromosome and distributed at different frequencies in the AML cases. Seven fusions are exclusively present in normal karyotype AML, and the rest fusions are shared between the normal karyotype AML and abnormal karyotype AML. CIITA, a master regulator of MHC class II gene expression and truncated in B-cell lymphoma and Hodgkin disease, is found to fuse with DEXI in 48% of normal karyotype AML cases. The fusion transcripts formed between adjacent genes highlight the possibility that certain such fusions could be involved in oncological process in AML, and provide a new source to identify genetic markers for normal karyotype AML
Isolation and characterization of a novel arenavirus harbored by Rodents and Shrews in Zhejiang province, China
AbstractTo determine the biodiversity of arenaviruses in China, we captured and screened rodents and shrews in Wenzhou city, Zhejiang province, a locality where hemorrhagic fever diseases are endemic in humans. Accordingly, arenaviruses were detected in 42 of 351 rodents from eight species, and in 12 of 272 Asian house shrews (Suncus murinus), by RT-PCR targeting the L segment. From these, a single arenavirus was successfully isolated in cell culture. The virion particles exhibited a typical arenavirus morphology under transmission electron microscopy. Comparison of the S and L segment sequences revealed high levels of nucleotide (>32.2% and >39.6%) and amino acid (>28.8% and >43.8%) sequence differences from known arenaviruses, suggesting that it represents a novel arenavirus, which we designated Wenzhou virus (WENV). Phylogenetic analysis revealed that all WENV strains harbored by both rodents and Asian house shrews formed a distinct lineage most closely related to Old World arenaviruses

The accelerated scaling attractor solution of the interacting agegraphic dark energy in Brans-Dicke theory
We investigate the interacting agegraphic dark energy in Brans-Dicke theory
and introduce a new series general forms of dark sector coupling. As examples,
we select three cases involving a linear interaction form (Model I) and two
nonlinear interaction form (Model II and Model III). Our conclusions show that
the accelerated scaling attractor solutions do exist in these models. We also
find that these interacting agegraphic dark energy modes are consistent with
the observational data. The difference in these models is that nonlinear
interaction forms give more approached evolution to the standard CDM
model than the linear one. Our work implies that the nonlinear interaction
