    Quantification of endogenous auxin levels and polyphenoloxidase enzymatic activity in olive (Olea europaea L.)

    A actividade enzimática de polifenoloxidases foi avaliada em folhas e na zona apical, média e basal de ramos de duas cultivares de oliveira (Olea europaea L.), comuns no Alentejo ( Galega vulgar e Cobrançosa ), mostrando que a actividade enzimática nas folhas foi muito superior à encontrada em tecidos de ramos do ano. Maior actividade enzimática foi também detectada na variedade Cobrançosa versus Galega vulgar . As condições óptimas para a determinação da actividade enzimática foram: pH= 5.5 e T= 40 ºC, com 20 mM de 4-metilcatecol em tampão acetato. Nestas condições o KM determinado foi: 2,60 e 3,48 mM com o método de Michaelis-Menten e Lineweaver-Burk, respectivamente. A melhor recuperação das auxinas AIA (ácido indol-3-acético) e AIB (ácido indolbutírico) em material vegetal foi conseguida através da extracção das amostras com acetona. A separação, identificação e quantificação do AIA e AIB em padrões, material vegetal dopado (tecidos de oliveira dopados com uma concentração conhecida de padrão) e não dopado, foi efectuada por técnicas cromatográficas (HPLC-DAD e LCMS), mostrando os resultados taxas de recuperação superiores a 40% para o AIA e 60% para o AIB

    Internal Transcribed Spacer 2 (nu ITS2 rRNA) Sequence-Structure Phylogenetics: Towards an Automated Reconstruction of the Green Algal Tree of Life

    L). Some have advocated the use of the nuclear-encoded, internal transcribed spacer two (ITS2) as an alternative to the traditional chloroplast markers. However, the ITS2 is broadly perceived to be insufficiently conserved or to be confounded by introgression or biparental inheritance patterns, precluding its broad use in phylogenetic reconstruction or as a DNA barcode. A growing body of evidence has shown that simultaneous analysis of nucleotide data with secondary structure information can overcome at least some of the limitations of ITS2. The goal of this investigation was to assess the feasibility of an automated, sequence-structure approach for analysis of IT2 data from a large sampling of phylum Chlorophyta.Sequences and secondary structures from 591 chlorophycean, 741 trebouxiophycean and 938 ulvophycean algae, all obtained from the ITS2 Database, were aligned using a sequence structure-specific scoring matrix. Phylogenetic relationships were reconstructed by Profile Neighbor-Joining coupled with a sequence structure-specific, general time reversible substitution model. Results from analyses of the ITS2 data were robust at multiple nodes and showed considerable congruence with results from published phylogenetic analyses.Our observations on the power of automated, sequence-structure analyses of ITS2 to reconstruct phylum-level phylogenies of the green algae validate this approach to assessing diversity for large sets of chlorophytan taxa. Moreover, our results indicate that objections to the use of ITS2 for DNA barcoding should be weighed against the utility of an automated, data analysis approach with demonstrated power to reconstruct evolutionary patterns for highly divergent lineages

    Regional differences in prediction models of lung function in Germany

    <p>Abstract</p> <p>Background</p> <p>Little is known about the influencing potential of specific characteristics on lung function in different populations. The aim of this analysis was to determine whether lung function determinants differ between subpopulations within Germany and whether prediction equations developed for one subpopulation are also adequate for another subpopulation.</p> <p>Methods</p> <p>Within three studies (KORA C, SHIP-I, ECRHS-I) in different areas of Germany 4059 adults performed lung function tests. The available data consisted of forced expiratory volume in one second, forced vital capacity and peak expiratory flow rate. For each study multivariate regression models were developed to predict lung function and Bland-Altman plots were established to evaluate the agreement between predicted and measured values.</p> <p>Results</p> <p>The final regression equations for FEV<sub>1 </sub>and FVC showed adjusted r-square values between 0.65 and 0.75, and for PEF they were between 0.46 and 0.61. In all studies gender, age, height and pack-years were significant determinants, each with a similar effect size. Regarding other predictors there were some, although not statistically significant, differences between the studies. Bland-Altman plots indicated that the regression models for each individual study adequately predict medium (i.e. normal) but not extremely high or low lung function values in the whole study population.</p> <p>Conclusions</p> <p>Simple models with gender, age and height explain a substantial part of lung function variance whereas further determinants add less than 5% to the total explained r-squared, at least for FEV1 and FVC. Thus, for different adult subpopulations of Germany one simple model for each lung function measures is still sufficient.</p

    Polyurethane scaffold with in situ swelling capacity for nucleus pulposus replacement

    Nucleus pulposus (NP) replacement offers a minimally invasive alternative to spinal fusion or total disc replacement for the treatment of intervertebral disc (IVD) degeneration. This study aimed to develop a cytocompatible {NP} replacement material, which is feasible for non-invasive delivery and tunable design, and allows immediate mechanical restoration of the IVD. A bi-phasic polyurethane scaffold was fabricated consisting of a core material with rapid swelling property and a flexible electrospun envelope. The scaffold was assessed in a bovine whole {IVD} organ culture model under dynamic load for 14 days. Nucleotomy was achieved by incision through the endplate without damaging the annulus fibrosus. After implantation of the scaffold and in situ swelling, the dynamic compressive stiffness and disc height were restored immediately. The scaffold also showed favorable cytocompatibility for native disc cells. Implantation of the scaffold in a partially nucleotomized {IVD} down-regulated catabolic gene expression, increased proteoglycan and type {II} collagen intensity and decreased type I collagen intensity in remaining {NP} tissue, indicating potential to retard degeneration and preserve the {IVD} cell phenotype. The scaffold can be delivered in a minimally invasive manner, and the geometry of the scaffold post-hydration is tunable by adjusting the core material, which allows individualized design.
