    Faint young Sun paradox remains

    The Sun was fainter when the Earth was young, but the climate was generally at least as warm as today; this is known as the `faint young Sun paradox'. Rosing et al. [1] claim that the paradox can be resolved by making the early Earth's clouds and surface less reflective. We show that, even with the strongest plausible assumptions, reducing cloud and surface albedos falls short by a factor of two of resolving the paradox. A temperate Archean climate cannot be reconciled with the low level of CO2 suggested by Rosing et al. [1]; a stronger greenhouse effect is needed.Comment: 3 pages, no figures.     Optimized high-throughput microRNA expression profiling provides novel biomarker assessment of clinical prostate and breast cancer biopsies

    BACKGROUND: Recent studies indicate that microRNAs (miRNAs) are mechanistically involved in the development of various human malignancies, suggesting that they represent a promising new class of cancer biomarkers. However, previously reported methods for measuring miRNA expression consume large amounts of tissue, prohibiting high-throughput miRNA profiling from typically small clinical samples such as excision or core needle biopsies of breast or prostate cancer. Here we describe a novel combination of linear amplification and labeling of miRNA for highly sensitive expression microarray profiling requiring only picogram quantities of purified microRNA. RESULTS: Comparison of microarray and qRT-PCR measured miRNA levels from two different prostate cancer cell lines showed concordance between the two platforms (Pearson correlation R(2 )= 0.81); and extension of the amplification, labeling and microarray platform was successfully demonstrated using clinical core and excision biopsy samples from breast and prostate cancer patients. Unsupervised clustering analysis of the prostate biopsy microarrays separated advanced and metastatic prostate cancers from pooled normal prostatic samples and from a non-malignant precursor lesion. Unsupervised clustering of the breast cancer microarrays significantly distinguished ErbB2-positive/ER-negative, ErbB2-positive/ER-positive, and ErbB2-negative/ER-positive breast cancer phenotypes (Fisher exact test, p = 0.03); as well, supervised analysis of these microarray profiles identified distinct miRNA subsets distinguishing ErbB2-positive from ErbB2-negative and ER-positive from ER-negative breast cancers, independent of other clinically important parameters (patient age; tumor size, node status and proliferation index). CONCLUSION: In sum, these findings demonstrate that optimized high-throughput microRNA expression profiling offers novel biomarker identification from typically small clinical samples such as breast and prostate cancer biopsies

    The association between insulin resistance and cytokines in adolescents: the role of weight status and exercise

    Increased adiposity is associated with insulin resistance (IR) and an inflammatory response in adults. We tested the hypotheses that cytokines associated with adiposity are also correlated with IR in early adolescents and that these relationships are moderated by weight status, levels of vigorous physical activity (VPA), or maximal aerobic power (pVO2max). Body mass, stature, and a fasting blood sample were obtained from 120 mid-pubertal adolescents (60 girls & 60 boys). Habitual VPA was obtained by a survey. Predicted VO2max was determined using a cycle-ergometer test. Weight status was based on body mass index percentiles (normal weight = BMI 95th %tile). Glucose, insulin, adiponectin, resistin, tumor necrosis factor-α, and interleukin-6 were measured, and IR index was based on the Homeostatic Model Assessment (HOMA). Adiponectin, resistin and TNF-α were associated with IR in all adolescents (R2=0.329, p0.050). Exercise did not moderate the association between these cytokines and IR. However, higher levels of VPA and/or pVO2max were associated with higher adiponectin, lower resistin and lower TNF- α in at least one of the genders. Our results indicate that the pathophysiology of obesity is already established in early adolescents. Increased adiposity, resulting in reduced adiponectin and increased resistin and TNF-α may link these cytokines with insulin resistance in adolescents

    RoSETZ: Roman Survey of the Earth Transit Zone -- a SETI-optimized survey for habitable-zone exoplanets

    In this White Paper for Nancy Grace Roman Space Telescope (Roman) science, we propose the Roman Survey of the Earth Transit Zone (RoSETZ), a transit search for rocky planets within the habitable zones (HZs) of stars located within the Earth Transit Zone (ETZ). The ETZ holds special interest in the search for extra-terrestrial intelligence (SETI) - observers on planets within the ETZ can see Earth as a transiting planet. RoSETZ would augment the Roman Galactic Bulge Time Domain Survey (GBTDS) as an additional field located 5\sim 5~degrees away from other GBTDS fields. Our simulations show that RoSETZ alone can find from 120 to 630 Earth-sized HZ planets around K- and M-type hosts, with the range reflecting different survey design assumptions. These yields are 5-20 times the number currently known. Such a sample will transform our knowledge of ``Eta-Earth'' (η\eta_{\oplus}) -- the occurrence of Earth-sized HZ planets -- and would be the first catalogue of exoplanets selected in a manner optimized according to the Mutual Detectability targetted-SETI strategy. If it can be accommodated alongside the existing GBTDS design, we favour a RoSETZ-Max design that is observed for the duration of the GBTDS. If not, we show that a slimmed-down RoSETZ-Lite design, occupying two GBTDS seasons, would not significantly impact overall GBTDS exoplanet yields, even if time allocated to it had to come from time allocations to other fields. We argue that the angular separation of RoSETZ from other GBTDS fields permits self-calibration of systematic uncertainties that would otherwise hamper exoplanet demographic modelling of both microlensing and transit datasets. Other science possible with RoSETZ data include studies of small solar system bodies and high resolution 3D extinction mapping.Comment: 20 pages.     Exoplanet diversity in the era of space-based direct imaging missions

    Community White Paper: submitted to the National Academy of Sciences Exoplanet Science StrategyThis white paper discusses the diversity of exoplanets that could be detected by future observations, so that comparative exoplanetology can be performed in the upcoming era of large space-based flagship missions. The primary focus will be on characterizing Earth-like worlds around Sun-like stars. However, we will also be able to characterize companion planets in the system simultaneously. This will not only provide a contextual picture with regards to our Solar system, but also presents a unique opportunity to observe size dependent planetary atmospheres at different orbital distances. We propose a preliminary scheme based on chemical behavior of gases and condensates in a planet's atmosphere that classifies them with respect to planetary radius and incident stellar flux

    Gene Expression and Biological Pathways in Tissue of Men with Prostate Cancer in a Randomized Clinical Trial of Lycopene and Fish Oil Supplementation

    Studies suggest that micronutrients may modify the risk or delay progression of prostate cancer; however, the molecular mechanisms involved are poorly understood. We examined the effects of lycopene and fish oil on prostate gene expression in a double-blind placebo-controlled randomized clinical trial.Eighty-four men with low risk prostate cancer were stratified based on self-reported dietary consumption of fish and tomatoes and then randomly assigned to a 3-month intervention of lycopene (n = 29) or fish oil (n = 27) supplementation or placebo (n = 28). Gene expression in morphologically normal prostate tissue was studied at baseline and at 3 months via cDNA microarray analysis. Differential gene expression and pathway analyses were performed to identify genes and pathways modulated by these micronutrients.Global gene expression analysis revealed no significant individual genes that were associated with high intake of fish or tomato at baseline or after 3 months of supplementation with lycopene or fish oil. However, exploratory pathway analyses of rank-ordered genes (based on p-values not corrected for multiple comparisons) revealed the modulation of androgen and estrogen metabolism in men who routinely consumed more fish (p = 0.029) and tomato (p = 0.008) compared to men who ate less. In addition, modulation of arachidonic acid metabolism (p = 0.01) was observed after 3 months of fish oil supplementation compared with the placebo group; and modulation of nuclear factor (erythroid derived-2) factor 2 or Nrf2-mediated oxidative stress response for either supplement versus placebo (fish oil: p = 0.01, lycopene: p = 0.001).We did not detect significant individual genes associated with dietary intake and supplementation of lycopene and fish oil.
