Spatial Nano-Morphology of the Prolamellar Body in Etiolated Arabidopsis thaliana Plants With Disturbed Pigment and Polyprenol Composition

The prolamellar body (PLB) is a periodic bicontinuous membrane structure based on tubular tetrahedral units. PLBs are present in plant etioplasts and, upon illumination, directly transform into the lamellar thylakoid networks within chloroplasts. Efficient tubular-lamellar rearrangement and later formation of the photosynthetically active thylakoid membranes are crucial steps in the development of plant autotrophy. PLB membranes are mainly composed of galactolipids, carotenoids, and protochlorophyllide (Pchlide), the chlorophyll precursor, bound in a complex with NADPH and Pchlide oxidoreductase. Although the PLB structure has been studied for over 50 years, the direct role of particular membrane components in the formation of the PLB paracrystalline network remains elusive. Moreover, despite the numerous literature data regarding the PLB geometry, their reliable comparative analysis is complicated due to variable experimental conditions. Therefore, we performed comprehensive ultrastructural and low-temperature fluorescence analysis of wild type Arabidopsis thaliana (Arabidopsis) seedlings grown in different conditions typical for studies on etiolated seedlings. We established that the addition of sucrose to the growing media significantly affected the size and compactness of the PLB. The etiolation period was also an important factor influencing the PLB structural parameters and the ratio of free to complex-bound Pchlide. Thus, a reliable PLB structural and spectral analysis requires particular attention to the applied experimental conditions. We investigated the influence of the pigment and polyprenol components of the etioplast membranes on the formation of the PLB spatial structure. The PLB 3D structure in several Arabidopsis mutants (ccr1-1, lut5-1, szl1-1npq1-2, aba1-6, pif1, cpt7) with disturbed levels of particular pigments and polyprenols using electron tomography technique was studied. We found that the PLB nano-morphology was mainly affected in the pif1 and aba1-6 mutants. An increased level of Pchlide (pif1) resulted in the substantial shift of the structural balance between outer and inner PLB water channels and overall PLB compactness compared to wild type plants. The decrease in the relative content of β-branch xanthophylls in aba1-6 plants was manifested by local disturbances in the paracrystalline structure of the PLB network. Therefore, proper levels of particular etioplast pigments are essential for the formation of stable and regular PLB structure

The relevance of surface curvature in biology is supported by numerous experimental and theoretical investigations in recent years. In this review, first, a brief introduction to the key ideas of surface curvature in the context of biological systems is given and the challenges that arise when measuring surface curvature are discussed. Giving an overview of the emergence of curvature in biological systems, its significance at different length scales becomes apparent. On the other hand, summarizing current findings also shows that both single cells and entire cell sheets, tissues or organisms respond to curvature by modulating their shape and their migration behavior. Finally, the interplay between the distribution of morphogens or micro-organisms and the emergence of curvature across length scales is addressed with examples demonstrating these key mechanistic principles of morphogenesis. Overall, this review highlights that curved interfaces are not merely a passive by-product of the chemical, biological, and mechanical processes but that curvature acts also as a signal that co-determines these processes.A.P.G.C. and P.R.F. acknowledge the funding from Fundação para a Ciência e Tecnologia (Portugal), through IDMEC, under LAETA project UIDB/50022/2020. Identyfikacja wielokryterialnego modelu istotności pompowni melioracyjnych na terenie Polski

    No full text
Niniejsza praca omawia istotny problem związany z klasyfikacją i właściwym zdefiniowaniem wielkości pompowni melioracyjnych. Do rozwiązania problemu wykorzystano jedną z wielokryterialnych metod podejmowania decyzji (MCDM), opartą na mechanizmach logiki rozmytej, noszącą nazwę metody obiektów charakterystycznych (COMET)

TECHNO-ECONOMIC ANALYSIS OF THE OF LAND RECLAMATION PUMPING STATIONS’ ENERGY EFFICIENCY

    No full text
The energy efficiency of the operation and maintenance of land reclamation pumping stations were conducted at this paper. The study included 12 pump stations located in the southern part of the region of Wielkopolska. All objects are administered by the Great Poland of Land Melioration and Water Units Board in Poznan, District Branch in Leszno. It was found that the work of half of the analyzed pumps is very expensive, and maintenance cost incomparable to the results achieved. Operating costs of the analyzed drainage pumping stations account for 86% of total operating costs, of which 40% is energy cost, and the remaining 14% to the upkeep. Small relative time of operating pumps for leeve and channel pumping stations indicates a for a leap of work, which proves their inefficient use. The use of a larger number of pumps with less power would preserve the continuity and stability of their work

TECHNICAL STATE ASSESSMENT OF THE DRAINAGE FACILITIES IN THE ZAGOROW POLDER

    No full text
At this work the assessment of state of water drainage conditions and the needs of its maintenance was carried out, as the basis for rational water management in usage of agricultural of Zagorow polder area. Based on field research, it was found that the condition of the 80.8% of the length of canals and ditches, and 40.7% of hydraulic and communication structures is unacceptable and does not meet the accepted criteria. The main reason of such condition of drainage facilities is a long-term exploitation absence or insufficient maintenance work frequency. To restore the proper state of maintenance facilities, in most cases the routine operation is sufficient. Before to the work we should conduct a detailed analysis regarding the necessity of the equipment for the smooth functioning of the drainage system. The analysis should address the current needs of the drainage area taking into account the economic conditions and the needs of environmental protection. On the polders often characterized by large natural values it is recommended to apply the principles of reasonable compromise between maintaining adequate drainage facilities and state of conservation of the natural environment

Published as: Behnke, M., Buchwald, M., Bykowski, A. et al. Psychophysiology of positive and negative emotions, dataset of 1157 cases and 8 biosignals. Sci Data 9, 10 (2022). https://doi.org/10.1038/s41597-021-01117-0 Subjective experience and physiological activity are fundamental components of emotion. There is an increasing interest in the link between experiental and physiological processes across different disciplines, e.g., psychology, economics, or computer science. However, the findings largely rely on sample sizes that have been modest at best (limiting the statistical power) and capture only some concurrent biosignals (e.g., making it problematic to seek specificity of some effects). We present a novel publicly available dataset of psychophysiological responses to positive and negative emotions that offers some improvement over other databases. This database involves recording of 1157 healthy participants, collected across seven studies, a continuous recordof self-reported affect along with several biosignals (electrocardiogram, impedance cardiogram, electrodermal activity, hemodynamic measures, e.g., blood pressure), respiration trace, and skin temperature. We experimentally elicited a wide range of positive and negative emotions, including amusement, anger, disgust, excitement, fear, gratitude, sadness, tenderness, and threat. Psychophysiology of positive and negative emotions (POPANE) database is currently the largest and the most comprehensive psychophysiological dataset on elicited emotions.
