    Developing serious games for cultural heritage: a state-of-the-art review

    Although the widespread use of gaming for leisure purposes has been well documented, the use of games to support cultural heritage purposes, such as historical teaching and learning, or for enhancing museum visits, has been less well considered. The state-of-the-art in serious game technology is identical to that of the state-of-the-art in entertainment games technology. As a result, the field of serious heritage games concerns itself with recent advances in computer games, real-time computer graphics, virtual and augmented reality and artificial intelligence. On the other hand, the main strengths of serious gaming applications may be generalised as being in the areas of communication, visual expression of information, collaboration mechanisms, interactivity and entertainment. In this report, we will focus on the state-of-the-art with respect to the theories, methods and technologies used in serious heritage games. We provide an overview of existing literature of relevance to the domain, discuss the strengths and weaknesses of the described methods and point out unsolved problems and challenges. In addition, several case studies illustrating the application of methods and technologies used in cultural heritage are presented

    Amphotericin B-loaded nanoparticles for local treatment of cutaneous leishmaniasis

    Cutaneous leishmaniasis (CL) is an infectious, parasitic disease caused by the protozoan Leishmania. Amphotericin B (AMB) is a macrolide polyene antibiotic presenting potent antifungal and antileishmanial activity, but due to poor water solubility at physiological pH, side effects, and toxicity, its therapeutic efficiency is limited. In the present study, poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NPs) loaded with AMB were generated to reduce drug toxicity and facilitate localized delivery over a prolonged time. AMB NPs were characterized for particle size, zeta potential, polydispersity index, and degree of aggregation. In vitro assessments demonstrated its sustained activity against Leishmania major promastigotes and parasite-infected macrophages. A single intralesional administration to infected BALB/c mice revealed that AMB NPs were more effective than AMB deoxycholate in terms of reducing lesion area. Taken together, these findings suggest thatAMB NPs improve AMB delivery and can be used for local treatment of CL.This research was funded by the GIP program of the Deutsche Forschungsgemeinschaft (DFG) German Research Foundation.     4D Numerical Analysis of Scaffolds: A New Approach

    A large range of biodegradable polymers are used to produce scaffoldsfor tissue engineering, which temporarily replace the biomechanical functions ofa biologic tissue while it progressively regenerates its capacities. However, the mechanicalbehavior of biodegradable materials during its degradation, which is an importantaspect of the scaffold design, is still an unexplored subject. For a biodegradablescaffold, performance will decrease along its degradation, ideally in accordanceto the regeneration of the biologic tissue, avoiding the stress shielding effect or thepremature rupture. In this chapter, a new numerical approach to predict the mechanicalbehavior of complex 3D scaffolds during degradation time (the 4th dimension)is presented. The degradation of mechanical properties should ideally be compatibleto the tissue regeneration. With this new approach, an iterative process of optimizationis possible to achieve an ideal solution in terms of mechanical behavior anddegradation time. The scaffold can therefore be pre-validated in terms of functionalcompatibility. An example of application of this approach is demonstrated at the endof this chapter
