    Managing risk and consistency in the raw material supply chain for single use systems

    As the bioprocessing industry moves increasingly towards single use systems (SUS), security of supply at the raw material level becomes imperative to quality. Long term supply of consistent, high quality materials desired for single use film is directly attributed to the stability, continuity and quality of raw material supply from the film manufacturer. This presentation will focus on the strategic selection of raw materials for a single use platform film in the bioprocessing industry to minimize risk associated with change notifications. Utilizing a quality by design approach, a single use film was formulated with no added Irgafos 168, enhanced abuse resistance and a low extractables and leachables profile. There are several factors that are critically important to understanding quality and risk of supply in raw material sourcing. This presentation will outline the raw material selection process established to ensure supply continuity and high quality desired for use in bioprocessing films. Although a buoyant small business sector is vital to the success of the UK economy, it is well established that most small businesses never grow or, at best, achieve only modest growth. Accordingly, understanding the factors that drive and shape small business performance is a key concern for both academics and policymakers. By increasing our understanding of these factors, this innovative project can make a major contribution to entrepreneurship research and to the evidence base underpinning enterprise policy

    A diabetes, em particular a diabetes mellitus tipo 2, constitui um distúrbio metabólico que se carateriza por glicemias aumentadas resultantes de diminuição da libertação e/ou ação da insulina. Os inibidores das α-glucosidases surgem como agentes de elevado interesse terapêutico uma vez que podem contribuir para a diminuição da absorção intestinal de glicose e, consequentemente, para um desagravamento da hiperglicemia. Alguns compostos da família iminociclitol são inibidores das α- glucosidases [1] e têm vindo a ser sintetizados diversos compostos novos que podem ser promissores e cuja ação sobre a atividade α-glucosidase de mamífero ainda não foi estudada. Neste trabalho procurou-se estudar a ação de alguns novos compostos da família iminociclitol (figura 1) na atividade α-glucosidase de enterócitos de mamífero, tendo-se também avaliado o grau de toxidade.     WII-FIT FOR BALANCE AND GAIT IN SKILLED NURSING FACILITY: A RETROSPECTIVE STUDY

    Background: Falls in elderly are a major public health problem. Poor balance and gait abnormalities are risk factors for falls. Exercise improves gait and balance in elderly. However, it is difficult to engage patients in exercise programs. Wii-Fit might bridge this gap by providing high level of engagement at an affordable price. Wii-Fit is a Nintendo game used for balance, yoga, aerobics, and strength training. It is a TV based self-directed activity. Virtual trainers talk the user through the activity and track progress while visual and auditory feedback improve engagement. Anecdotal reports suggest improvement in balance and social benefits with Wii-Fit, but no systematic studies are available. Objective: To compare the effect of Wii-Fit augmentation to physical therapy alone in subjects undergoing rehabilitation in skilled nursing facility (SNF) using a retrospective chart review. Methods: 100 charts were reviewed of which seventeen patients were noted to have used Wii-Fit during their rehabilitation. These subjects were matched with seventeen controls who did physical therapy alone. Results: There were no baseline differences between the groups. The mean age was 77.7 years, with 11 females in each group. All the subjects were Caucasians. The average length of stay was 30 days in each group. Mean duration of the Wii-Fit use was 4-weeks. Wii-Fit augmentation group improved significantly in Activities of Daily Living (p=0.008), balance (p=0.0001), and assistance with gait (p=0.05) when compared to the control group. Conclusion: Wii-Fit can be used safely in a SNF, and it may improve balance, gait, and functional status

    3-Hydroxypyrrolidine and (3,4)-dihydroxypyrrolidine derivatives: inhibition of rat intestinal α-glucosidase

    Thirteen pyrrolidine-based iminosugar derivatives have been synthesized and evaluated for inhibition of α-glucosidase from rat intestine. The compounds studied were the non-hydroxy, mono-hydroxy and dihydroxypyrrolidines. All the compounds were N-benzylated apart from one. Four of the compounds had a carbonyl group in the 2,5-position of the pyrrolidine ring. The most promising iminosugar was the trans-3,4-dihydroxypyrrolidine 5 giving an IC50 of 2.97 ± 0.046 and a KI of 1.18 mM. Kinetic studies showed that the inhibition was of the mixed type, but predominantly competitive for all the compounds tested. Toxicological assay results showed that the compounds have low toxicity. Docking studies showed that all the compounds occupy the same region as the DNJ inhibitor on the enzyme binding site with the most active compounds establishing similar interactions with key residues. Our studies suggest that a rotation of ∼90° of some compounds inside the binding pocket is responsible for the complete loss of inhibitory activity. Despite the fact that activity was found only in the mM range, these compounds have served as simple molecular tools for probing the structural features of the enzyme, so that inhibition can be improved in further studies

    An All-Cryogenic THz Transmission Spectrometer

    This paper describes a THz transmission spectrometer for the spectral range of 2-65 cm^-1 (100 GHz to 2 THz) with a spectral resolution of at least 1.8 cm^-1 (50 GHz) where the source, sample, and detector are all fully contained in a cryogenic environment. Cyclotron emission from a two-dimensional electron gas heated with an electrical current serves as a magnetic field tunable source. The spectrometer is demonstrated at 4.2 K by measuring the resonant cyclotron absorption of a second two dimensional electron gas. Unique aspects of the spectrometer are that 1) an ultra-broadband detector is used and 2) the emitter is run quasi-continuously with a chopping frequency of only 1 Hz. Since optical coupling to room temperature components is not necessary, this technique is compatible with ultra-low temperature (sub 100 mK) operation.Comment: 7 pages, 5 figures.     Complications and failure modes of coronary embolic protection devices: Insights from the MAUDE database

    BACKGROUND: There is limited data on complications associated with the use of coronary embolic protection devices (EPDs). METHODS: We queried the Manufacturer and User Facility Device Experience database between November 2010 and November 2020 for reports on coronary EPDs: Spider FX (Medtronic, Minneapolis, MN) and Filterwire EZ (Boston Scientific, Natick, MA). RESULTS: We retrieved 119 reports on coronary EPD failure (Spider FX n = 33 and Filterwire EZ n = 86), most of which (78.2%) occurred during saphenous vein graft interventions. The most common failure mode was inability to retrieve the EPD (49.6%), with the filter trapped against stent struts in 76.2% of the cases. Other device complications included filter fracture (28.6%), failure to cross (7.6%), failure to deploy (7.6%), and failure to recapture the filter (3.4%). Filter fracture (54.5 vs. 29.1%) and failure to recapture (9.1 vs. 2.1%) were more commonly reported, while failure to deploy the filter (0 vs. 10.5%) was less commonly reported with the Spider-FX. CONCLUSIONS: The most common modes of failure of coronary EPDs are the failure of retrieval (49.6%), followed by the filter fracture (28.6%). When using EPDs, careful attention to the technique is essential to avoid failures and subsequent complications
