Influence of management and environment on Australian wheat: information for sustainable intensification and closing yield gaps
In the future, agriculture will need to produce more, from less land, more sustainably. But currently, in many places, actual crop yields are below those attainable. We quantified the ability for agricultural management to increase wheat yields across 179 Mha of potentially arable land in Australia. Using the Agricultural Production Systems Simulator (APSIM), we simulated the impact on wheat yield of 225 fertilization and residue management scenarios at a high spatial, temporal, and agronomic resolution from 1900 to 2010. The influence of management and environmental variables on wheat yield was then assessed using Spearman\u27s non-parametric correlation test with bootstrapping. While residue management showed little correlation, fertilization strongly increased wheat yield up to around 100 kg N ha−1 yr−1. However, this effect was highly dependent on the key environment variables of rainfall, temperature, and soil water holding capacity. The influence of fertilization on yield was stronger in cooler, wetter climates, and in soils with greater water holding capacity. We conclude that the effectiveness of management intensification to increase wheat yield is highly dependent upon local climate and soil conditions. We provide context-specific information on the yield benefits of fertilization to support adaptive agronomic decision-making and contribute to the closure of yield gaps. We also suggest that future assessments consider the economic and environmental sustainability of management intensification for closing yield gaps

Atomic excitation during recollision-free ultrafast multi-electron tunnel ionization
Modern intense ultrafast pulsed lasers generate an electric field of
sufficient strength to permit tunnel ionization of the valence electrons in
atoms. This process is usually treated as a rapid succession of isolated
events, in which the states of the remaining electrons are neglected. Such
electronic interactions are predicted to be weak, the exception being
recollision excitation and ionization caused by linearly-polarized radiation.
In contrast, it has recently been suggested that intense field ionization may
be accompanied by a two-stage `shake-up' reaction. Here we report a unique
combination of experimental techniques that enables us to accurately measure
the tunnel ionization probability for argon exposed to 50 femtosecond laser
pulses. Most significantly for the current study, this measurement is
independent of the optical focal geometry, equivalent to a homogenous electric
field. Furthermore, circularly-polarized radiation negates recollision. The
present measurements indicate that tunnel ionization results in simultaneous
excitation of one or more remaining electrons through shake-up. From an atomic
physics standpoint, it may be possible to induce ionization from specific
states, and will influence the development of coherent attosecond XUV radiation
sources. Such pulses have vital scientific and economic potential in areas such

Mechanisms of β-lactam resistance of Streptococcus uberis isolated from bovine mastitis cases.
A number of veterinary clinical pathology laboratories in New Zealand have been reporting emergence of increased minimum in inhibitory concentrations for β-lactams in the common clinical bovine mastitis pathogen Streptococcus uberis. The objective of this study was to determine the genetic basis of this increase in MIC for β-lactams amongst S. uberis. Illumina sequencing and determination of oxacillin MIC was performed on 265 clinical isolates. Published sequences of the five penicillin binding proteins pbp1a, pbp1b, pbp2a, pbp2b, and pbp2x were used to identify, extract and align these sequences from the study isolates. Amino acid substitutions resulting from single nucleotide polymorphisms (SNP) within these genes were analysed for associations with elevated (≥ 0.5 mg/L) oxacillin MIC together with a genome wide association study. The population structure of the study isolates was approximated using a phylogenetic tree generated from an alignment of the core genome. A total of 53 % of isolates had MIC ≥ 0.5 mg/L for oxacillin. A total of 101 substitutions within the five pbp were identified, of which 11 were statistically associated with an MIC ≥ 0.5 mg/L. All 140 isolates which exhibited an increased β-lactam MIC had SNPs leading to pbp2x E381K and Q554E substitutions. The phylogenetic tree indicated that the genotype and phenotype associated with the increased MIC for oxacillin were present in several different lineages suggesting that acquisition of this increased β-lactam MIC had occurred in multiple geographically distinct regions. Reanalysis of the data from the intervention studies from which the isolates were originally drawn found a tendency for the pbp2x E381K substitution to be associated with lower cure rates. It is concluded that there is geographically and genetically widespread presence of pbp substitutions associated with reduced susceptibility to β-lactam antimicrobials. Additionally, presence of pbp substitutions tended to be associated with poorer cure rate outcomes following antimicrobial therapy for clinical mastitis
