    Dog breeding and dog trade are strictly regulated by EU-legislation, as well as by national and regional legislations. Nevertheless, it is believed that part of the puppies traded within the EU are younger than the legal minimum age, with the risk of compromising their own health and wellbeing as well as public health. A proper age determination is necessary to expose potential malpractices. Combining multiple independent methods is essential to compensate for possible variations in one single estimation method. Assessment of the dental age and bone age, more specifically by evaluating the eruption status of the dentition and the radiographic appearance and formation of ossification centers are both eligible. However, current available data are not detailed enough. In order to improve the accuracy and reduce the error in age estimation, more breed specific data are required, more intermediate development stages must be described and possible developmental disorders and pathologies must be considered

    Angiopoietins differentially influence in vitro angiogenesis by endothelial cells of different origin

    Angiopoietins are important growth factors for vascular development and quiescence. They are promising targets for pro-or anti-angiogenic therapies in diverse pathologies, but the mechanisms of the ANGPT/TIE2 system are complex and not well understood. In the present study, the separate and combined effects of angiopoietin 1 and angiopoietin 2 were studied, using a recently developed in vitro angiogenesis model that allows both a quantitative and qualitative evaluation of the angiogenic cascade. This cell culture model was performed with microvascular endothelial cells (ECs) originating from different vascular beds, i.e. dermal ECs and cardiac ECs. In addition, the expression of the angiopoietins and the receptors, TIE1 and TIE2 was analyzed with RT-qPCR. This study revealed that the angiopoietins provoked a differential response in the two endothelial cultures. Both angiopoietin 1 as well as angiopoietin 2 elicited an angiogenic cascade in the dermal ECs but not in the cardiac ECs. In addition, the RT-qPCR data revealed marked differences in the endogenous expression pattern of these factors, indicating that the origin of endothelial cells might have an important impact on their angiogenic potential

    This article examines how the re-institutionalisation of transport policies starts with the everyday insurgency of fare evasion practices which exist at the fringe of social innovation. In the case of Brussels, fare evaders organised themselves at the heart of the digital world, as virtual communities to claim their rights to mobility and to re-common the public transport infrastructure by mobilising offline evasion actions. This perspective allows to understand fare evasion not as an illegal or criminal practice but as a symptom that embodies how transport policies fail under marketisation. The article concludes that fare evaders have the potential to be bottom-up actors, yet they are limited by socio-economic cleavages and the predominant transport policies

    Dialectical design dialogues: negotiating ethics in participatory planning by building a critical design atlas

    This article explores 'dialectical design dialogues' as an approach to engage with ethics in everyday urban planning contexts. It starts from Paulo Freire’s pedagogical view (1970/2017), in which dialogues imply the establishment of a horizontal relation between professionals and amateurs, in order to understand, question and imagine things in everyday reality, in this case, urban transformations, applied to participatory planning and enriched through David Harvey’s (2000, 2009) dialectical approach. A dialectical approach to design dialogues acknowledges and renegotiates contrasts and convergences of ethical concerns specific to the reality of concrete daily life, rather than artificially presenting daily life as made of consensus or homogeneity. The article analyses an atlas as a tool to facilitate dialectical design dialogues in a case study of a low-density residential neighbourhood in the city of Genk, Belgium. It sees the production of the atlas as a collective endeavour during which planners, authorities and citizens reflect on possible futures starting from a confrontation of competing uses and perspectives of neighbourhood spaces. The article contributes to the state-of-the-art in participatory urban planning in two ways: (1) by reframing the theoretical discussion on ethics by arguing that not only the verbal discourses around designerly atlas techniques but also the techniques themselves can support urban planners in dealing more consciously with ethics (accountability, morality and authorship) throughout urban planning processes, (2) by offering a concrete practice-based example of producing an atlas that supports the participatory articulation and negotiation of dialectical inquiry of ethics through dialogues in a 'real-time' urban planning process

    Ontogenetic development of the gastrointestinal tract of African lungfish larvae Protopterus aethiopicus (Heckel 1851) : a light microscopy study

    The organogenesis of the digestive system was described in the African lungfish (Protopterus aethiopicus) from 6 days post-hatching (6 DPH) to 17 days post-hatching (17 DPH) reared at 27 degrees C. To elucidate the position of the gastrointestinal tract in relation to the neural tube, notochord and yolk sac at 6 DPH, and to the vertebral column, lungs and kidneys at 17 DPH, larvae were mapped by means of computer-assisted 3D reconstructions starting from histological serial sections. The larvae showed a simple digestive tract, which appeared as a straight undifferentiated and closed tube at 6 DPH. Microscopical observation showed that yolk reserves were not completely depleted by 17 DPH. During the endogenous feeding period at least up to 17 DPH, the larval digestive system experienced a fast transformation with almost complete development of most digestive organs (pharyngeal teeth, intestinal vestibule, intestine and liver). Our findings suggest that, by 17 DPH, African lungfish larvae are ready to start exogenous feeding

    Intussusceptive pillar formation in developing porcine glomeruli

    Background/Aims: Intussusceptive angiogenesis (IA) is a dynamic process which contributes to vascular expansion and remodeling. Intraluminal pillars have long been the distinctive structural indicator of intussusceptive angiogenesis. However, the mechanism of their formation has not been fully elucidated yet. Methods: Using light and electron microscopy, we studied intussusceptive vascular growth in the developing porcine metanephric kidney. Results: We observed intraluminal pillars formed by endothelial cells in the vasculature of developing glomeruli. Their diameter was < 2.5 µm, consistent with the diameter of nascent pillars. TEM revealed that the majority of these pillars consisted only of endothelium. However, a central core of extracellular matrix (ECM) covered by endothelium, reminiscent of a more mature intussusceptive pillar, was also found in the lumen of a glomerular capillary. Perivascular cells or pericytes were not involved in the pillar structure during these stages of formation. Conclusion: This study shows ECM presence in a mature intussusceptive pillar without any perivascular cell involvement in the structure. This leads to the hypothesis that ECM deposition preceeds the participation of these cells in the formation of intraluminal pillars during IA in porcine metanephric glomerular capillaries

    The development of a juvenile porcine augmented renal clearance model through continuous infusion of lipopolysaccharides : an exploratory study

    Augmented renal clearance (ARC) as observed in the critically ill (pediatric) population can have a major impact on the pharmacokinetics and posology of renally excreted drugs. Although sepsis has been described as a major trigger in the development of ARC in human critically ill patients, mechanistic insights on ARC are currently lacking. An appropriate ARC animal model could contribute to reveal these underlying mechanisms. In this exploratory study, a state of ARC was induced in 8-week-old piglets. Conscious piglets were continuously infused over 36 h with lipopolysaccharides (LPS) from Escherichia coli (O111:B4) to induce sepsis and subsequently trigger ARC. To study the dose-dependent effect of LPS on the renal function, three different doses (0.75, 2.0, 5.0 mu g/kg/h) were administered (two male piglets/dose, one sham piglet), in combination with fluid administration (0.9% NaCl) at 6 ml/kg/h. Single boluses of renal markers, i.e., creatinine [40 mg/kg body weight (BW)], iohexol (64.7 mg/kg BW), and para-aminohippuric acid (PAH, 10 mg/kg BW) were administered intravenously to evaluate the effect of LPS on the renal function. Clinical parameters were monitored periodically. Blood sampling was performed to determine the effect on hematology, neutrophil gelatinase-associated lipocalin, and prostaglandin E-2 plasma levels. All piglets that were continuously infused with LPS displayed an elevated body temperature, heart rhythm, and respiratory rate similar to 1-3 h after start of the infusion. After infusion, considerably higher total body clearances of iohexol, creatinine, and PAH were observed, independent of the administration of LPS and/or its dose. Since also the sham piglet, receiving no LPS, demonstrated a comparable increase in renal function, the contribution of fluid administration to the development of ARC should be further evaluated
