    Selective advantage for multicellular replicative strategies: A two-cell example

    This paper develops a quasispecies model where cells can adopt a two-cell survival strategy. Within this strategy, pairs of cells join together, at which point one of the cells sacrifices its own replicative ability for the sake of the other cell. We develop a simplified model for the evolutionary dynamics of this process, allowing us to solve for the steady-state using standard approaches from quasispecies theory. We find that our model exhibits two distinct regimes of behavior: At low concentrations of limiting resource, the two-cell strategy outcompetes the single-cell survival strategy, while at high concentrations of limiting resource, the single-cell survival strategy dominates. Associated with the two solution regimes of our model is a localization to delocalization transition over the portion of the genome coding for the multicell strategy, analogous to the error catastrophe in standard quasispecies models. The existence of such a transition indicates that multicellularity can emerge because natural selection does not act on specific cells, but rather on replicative strategies. Within this framework, individual cells become the means by which replicative strategies are propagated.     A randomized controlled pilot trial of classroom-based mindfulness meditation compared to an active control condition in sixth-grade children

    The current study is a pilot trial to examine the effects of a nonelective, classroom-based, teacher-implemented, mindfulness meditation intervention on standard clinical measures of mental health and affect in middle school children. A total of 101 healthy sixth-grade students (55 boys, 46 girls) were randomized to either an Asian history course with daily mindfulness meditation practice (intervention group) or an African history course with a matched experiential activity (active control group). Self-reported measures included the Youth Self Report (YSR), a modified Spielberger State-Trait Anxiety Inventory, and the Cognitive and Affective Mindfulness Measure –Revised. Both groups decreased significantly on clinical syndrome subscales and affect but did not differ in the extent of their improvements. Meditators were significantly less likely to develop suicidal ideation or thoughts of self-harm than controls. These results suggest that mindfulness training may yield both unique and non-specific benefits that are shared by other novel activities

    Reducing Severity of Primary Dysmenorrhea with the Utilization of Acupressure

    Primary dysmenorrhea is a common gynecological condition associated with painful cramps during menstruation in the absence of other diseases. It impacts large populations of women worldwide yet is often disregarded and inadequately treated. The ways in which women are treated for this condition often bring forth issues including overuse of medications, drug dependence, adverse reactions from medication use, and increased medical expenses. The utilization of non-pharmacological options has been shown to be effective in managing pain related to primary dysmenorrhea, including the complementary and integrative treatment of acupressure. Guided by Jean Watson’s Human Caring Science Theory, this scholarly DNP-FNP project explored the use of acupressure to reduce the severity of primary dysmenorrhea in women. Women (N= 14) were asked to participate in an acupressure education class and preform acupressure at acupoints LI 4, SP 6, and SP 8 during one menstrual period. Pre- and post- intervention questionnaires were completed by the participants to measure the effectiveness of the acupressure intervention. The post intervention questionnaires indicated reduced levels of pain in relation to primary dysmenorrhea. These findings are pertinent to the role of the nurse practitioner as they suggest the importance of utilizing integrative modalities including acupressure as a means of treatment. The implications of this scholarly project can be utilized by all women experiencing pain related in primary dysmenorrhea and indicate the potential benefits of utilizing complementary and alternative modalities of health

    Differential expression of microRNAs in bovine papillomavirus type 1 transformed equine cells

    Bovine papillomavirus (BPV) types 1 and 2 play an important role in the pathogenesis of equine sarcoids (ES), the most common cutaneous tumour affecting horses. MicroRNAs (miRNAs), small non-coding RNAs that regulate essential biological and cellular processes, have been found dysregulated in a wide range of tumours. The aim of this study was to identify miRNAs associated with ES. Differential expression of miRNAs was assessed in control equine fibroblasts (EqPalFs) and EqPalFs transformed with the BPV-1 genome (S6-2 cells). Using a commercially available miRNA microarray, 492 mature miRNAs were interrogated. In total, 206 mature miRNAs were differentially expressed in EqPalFs compared with S6-2 cells. Aberrant expression of these miRNAs in S6-2 cells can be attributed to the presence of BPV-1 genomes. Furthermore, we confirm the presence of 124 miRNAs previously computationally predicted in the horse. Our data supports the involvement of miRNAs in the pathogenesis of ES

    Optimizing broad ion beam polishing of zircaloy-4 for electron backscatter diffraction analysis

    Microstructural analysis with electron backscatter diffraction (EBSD) involves sectioning and polishing to create a flat and preparation-artifact free surface. The quality of EBSD analysis is often dependant on this step, and this motivates us to explore how broad ion beam (BIB) milling can be optimised for the preparation of zircaloy-4 with different grain sizes. We systematically explore the role of ion beam angle, ion beam voltage, polishing duration and polishing temperature and how this changes the surface roughness and indexing quality. Our results provide a method to routinely prepare high-quality Zircaloy-4 surfaces, and methods to optimise BIB polishing of other materials for high-quality EBSD studies

    Application of small RNA technology for improved control of parasitic helminths

    Over the last decade microRNAs (miRNAs) and small interfering RNAs (siRNAs) have emerged as important regulators of post-transcriptional gene expression. miRNAs are short, non-coding RNAs that regulate a variety of processes including cancer, organ development and immune function. This class of small RNAs bind with partial complementarity to their target mRNA sequences, most often in the 3′UTR, to negatively regulate gene expression. In parasitic helminths, miRNAs are being increasingly studied for their potential roles in development and host-parasite interactions. The availability of genome data, combined with small RNA sequencing, has paved the way to profile miRNAs expressed at particular developmental stages for many parasitic helminths. While some miRNAs are conserved across species, others appear to be unique to specific parasites, suggesting important roles in adaptation and survival in the host environment. Some miRNAs are released from parasites, in exosomes or in protein complexes, and the potential effects of these on host immune function are being increasingly studied. In addition, release of miRNAs from schistosome and filarial parasites into host plasma can be exploited for the development of specific and sensitive diagnostic biomarkers of infection. Interfering with miRNA function, as well as silencing key components of the pathways they regulate, will progress our understanding of parasite development and provide a novel approach to therapeutic control. RNA interference (RNAi) by siRNAs has proven to be inconsistent in parasitic nematodes. However, the recent successes reported for schistosome and liver fluke RNAi, encourage further efforts to enhance delivery of RNA and improve in vitro culture systems and assays to monitor phenotypic effects in nematodes. These improvements are important for the establishment of reliable functional genomic platforms for novel drug and vaccine development. In this review we focus on the important roles of miRNAs and siRNAs in post-transcriptional gene regulation in veterinary parasitic helminths and the potential value of these in parasite diagnosis and control

    Gender Differences in Response to a School-Based Mindfulness Training Intervention for Early Adolescents

    Mindfulness training has been used to improve emotional wellbeing in early adolescents. However, little is known about treatment outcome moderators, or individual differences that may differentially impact responses to treatment. The current study focused on gender as a potential moderator for affective outcomes in response to school-based mindfulness training. Sixth grade students (N = 100) were randomly assigned to either the six weeks of mindfulness meditation or the active control group as part of a history class curriculum. Participants in the mindfulness meditation group completed short mindfulness meditation sessions four to five times per week, in addition to didactic instruction (Asian history). The control group received matched experiential activity in addition to didactic instruction (African history) from the same teacher with no meditation component. Self-reported measures of emotional wellbeing/affect, mindfulness, and self-compassion were obtained at pre and post intervention. Meditators reported greater improvement in emotional wellbeing compared to those in the control group. Importantly, gender differences were detected, such that female meditators reported greater increases in positive affect compared to females in the control group, whereas male meditators and control males displayed equivalent gains. Uniquely among females but not males, increases in self-reported self-compassion were associated with improvements in affect. These findings support the efficacy of school-based mindfulness interventions, and interventions tailored to accommodate distinct developmental needs of female and male adolescents

    Spatial synchronization and extinction of species under external forcing

    We study the interplay between synchronization and extinction of a species. Using a general model we show that under a common external forcing, the species with a quadratic saturation term in the population dynamics first undergoes spatial synchronization and then extinction, thereby avoiding the rescue effect. This is because the saturation term reduces the synchronization time scale but not the extinction time scale. The effect can be observed even when the external forcing acts only on some locations provided there is a synchronizing term in the dynamics. Absence of the quadratic saturation term can help the species to avoid extinction.Comment: 4 pages, 2 figure
