Retinoid X Receptor and Peroxisome Proliferator-Activated Receptor-Gamma Agonists Cooperate to Inhibit Matrix Metalloproteinase Gene Expression
We recently described the ability of retinoid X receptor (RXR) ligand LG100268 (LG268) to inhibit interleukin-1-beta (IL-1-β)-driven matrix metalloproteinase-1 (MMP-1) and MMP-13 gene expression in SW-1353 chondrosarcoma cells. Other investigators have demonstrated similar effects in chondrocytes treated with rosiglitazone, a ligand for peroxisome proliferator-activated receptor-gamma (PPARγ), for which RXR is an obligate dimerization partner. Laser Ablation Mass Spectrometer (LAMS) as a Standoff Analyzer in Space Missions for Airless Bodies
A laser ablation mass spectrometer (LAMS) based on a time-of-flight (TOF) analyzer with adjustable drift length is proposed as a standoff elemental composition sensor for space missions to airless bodies. It is found that the use of a retarding potential analyzer in combination with a two-stage reflectron enables LAMS to be operated at variable drift length. For field-free drift lengths between 33 cm to 100 cm, at least unit mass resolution can be maintained solely by adjustment of internal voltages, and without resorting to drastic reductions in sensitivity. Therefore, LAMS should be able to be mounted on a robotic arm and analyze samples at standoff distances of up to several tens of cm, permitting high operational flexibility and wide area coverage of heterogeneous regolith on airless bodies

Co). The novel magnetic relaxation, which is quite different from those
in normal spin glasses and superparamagnets but very resembles qualitatively
those in single-molecule magnet Mn-Ac even if they possess different
structures, might be attributed to the presence of frustration that is
incrementally unveiled by the external magnetic field. The two distinct
transitions in [GdFe] were presumed from DC and AC susceptibility as well as

Quantifying the increasing sensitivity of power systems to climate variability
Large quantities of weather-dependent renewable energy generation are expected in power systems under climate change mitigation policies, yet little attention has been given to the impact of long term climate variability. By combining state-ofthe-art multi-decadal meteorological records with a parsimonious representation of a power system, this study characterises the impact of year-to-year climate variability on multiple aspects of the power system of Great Britain (including coal, gas and
nuclear generation), demonstrating why multi-decadal approaches are necessary. All aspects of the example system are impacted by inter-annual climate variability, with the impacts being most pronounced for baseload generation. The impacts of inter-annual climate variability increase in a 2025 wind-power scenario, with a 4-fold increase in
the inter-annual range of operating hours for baseload such as nuclear. The impacts on peak load and peaking-plant are comparably small. Less than 10 years of power supply and demand data are shown to be insuffcient for providing robust power system planning guidance. This suggests renewable integration studies - widely used in policy, investment and system design - should adopt a more robust approach to climate characterisation

Dragonfly: Investigating the Surface Composition of Titan
Dragonfly is a rotorcraft lander mission, selected as a finalist in NASA's New Frontiers Program, that is designed to sample materials and determine the surface composition in different geologic settings on Titan. This revolutionary mission concept would explore diverse locations to characterize the habitability of Titan's environment, to investigate how far prebiotic chemistry has progressed, and to search for chemical signatures that could be indicative of water-based and/or hydrocarbon-based life. Here we describe Dragonfly's capabilities to determine the composition of a variety of surface units on Titan, from elemental components to complex organic molecules. The compositional investigation ncludes characterization of local surface environments and finely sampled materials. The Dragonfly flexible sampling approach can robustly accommodate materials from Titan's most intriguing surface environments
Curiosity's Sample Analysis at Mars (SAM) Investigation: Overview of Results from the First 120 Sols on Mars
During the first 120 sols of Curiosity s landed mission on Mars (8/6/2012 to 12/7/2012) SAM sampled the atmosphere 9 times and an eolian bedform named Rocknest 4 times. The atmospheric experiments utilized SAM s quadrupole mass spectrometer (QMS) and tunable laser spectrometer (TLS) while the solid sample experiments also utilized the gas chromatograph (GC). Although a number of core experiments were pre-programmed and stored in EEProm, a high level SAM scripting language enabled the team to optimize experiments based on prior runs
MMP-13 stimulates osteoclast differentiation and activation in tumour breast bone metastases
INTRODUCTION:
The increased bone degradation in osteolytic metastases depends on stimulation of mature osteoclasts and on continuous differentiation of new pre-osteoclasts. Metalloproteinases (MMP)-13 is expressed in a broad range of primary malignant tumours and it is emerging as a novel biomarker. Recent data suggest a direct role of MMP-13 in dissolving bone matrix complementing the activity of MMP-9 and other enzymes. Tumour-microenvironment interactions alter gene expression in malignant breast tumour cells promoting osteolytic bone metastasis. Gene expression profiles revealed that MMP-13 was among the up-regulated genes in tumour-bone interface and its abrogation reduced bone erosion. The precise mechanism remained not fully understood. Our purpose was to further investigate the mechanistic role of MMP-13 in bone osteolytic lesions.
METHODS:
MDA-MB-231 breast cancer cells that express MMP-13 were used as a model for in vitro and in vivo experiments. Conditioned media from MDA-MB-231 cells were added to peripheral blood mononuclear cultures to monitor pre-osteoclast differentiation and activation. Bone erosion was evaluated after injection of MMP-13-silenced MDA-MB-231 cells into nude mice femurs.
RESULTS:
MMP-13 was co-expressed by human breast tumour bone metastases with its activator MT1-MMP. MMP-13 was up-regulated in breast cancer cells after in vitro stimulation with IL-8 and was responsible for increased bone resorption and osteoclastogenesis, both of which were reduced by MMP inhibitors. We hypothesized that MMP-13 might be directly involved in the loop promoting pre-osteoclast differentiation and activity. We obtained further evidence for a direct role of MMP-13 in bone metastasis by a silencing approach: conditioned media from MDA-MB-231 after MMP-13 abrogation or co-cultivation of silenced cells with pre-osteoclast were unable to increase pre-osteoclast differentiation and resorption activity. MMP-13 activated pre-MMP-9 and promoted the cleavage of galectin-3, a suppressor of osteoclastogenesis, thus contributing to pre-osteoclast differentiation. Accordingly, MMP-13 abrogation in tumour cells injected into the femurs of nude mice reduced the differentiation of TRAP positive cells in bone marrow and within the tumour mass as well as bone erosion.
CONCLUSIONS:
These results indicate that within the inflammatory bone microenvironment MMP-13 production was up-regulated in breast tumour cells leading to increased pre-osteoclast differentiation and their subsequent activation
