    Satellite investigations of ice dynamics and supraglacial lake development in Greenland

    This thesis aims to improve the current understanding of the processes which control the flow variability of Greenland Ice Sheet (GrIS) outlet glaciers. The most recent Intergovernmental Governmental Panel on Climate Change (IPCC) report (Meehl et al., 2007) identifies that a critical limitation to forecasts of sea-level rise are uncertainties in modelling the ice dynamics of the GrIS. Using Synthetic Aperture Radar (SAR) feature tracking, seasonal velocities of land- and marine- terminating glaciers in a region in the northeast of Greenland are measured. Records of air temperature in conjunction with seasonal observations of supraglacial lake development, sea ice conditions and ice front positions, derived from SAR imagery, are used to investigate the controls on the observed variations in ice velocity. A clear link between ice velocities and glacier hydrology is found. These findings are consistent with observations from other glaciers in Greenland and are suggestive of a universal hydrological forcing of ice velocity for the whole of the GrIS ablation zone. Lake drainage events have been identified as a key factor in linking atmospheric changes, glacier hydrology and ice velocities in Greenland. For modelling purposes, a means of parameterising the distribution and evolution of supraglacial lakes is therefore needed. Assuming that water will pond in surface depressions, this thesis assesses the ability of using Digital Elevation Models (DEMs) for this purpose. High resolution DEMs are created using Interferometric SAR (InSAR) for two, separate regions of the GrIS. The positions and areal extent of surface depressions are compared with those of lakes observed in optical satellite imagery. The level of correspondence between the two datasets is found to be poor as a result of the resolution of the DEMs and the physical differences between surface depressions and lakes (e.g. lakes may not fill the capacity of the depression). An alternative method for parameterising the seasonal distribution of supra-glacial lakes, by extrapolating trends observed in current lake distributions, is investigated. The locations and evolution of lakes in the west of Greenland during the summer of 2003 are mapped using 47 Moderate Resolution Imaging Spectroradiometer (MODIS) images. Clear trends are identified in the distributions of lakes with elevation and are linked to the seasonal melt-cycle and to changes in ice thickness and its influence on surface depressions, tensile stresses and hydrofracturing. It may be possible to extrapolate these trends to other regions and higher elevations on the ice sheet, thereby enabling the distribution of lakes to be parameterised in ice sheet models. The findings of this thesis help to contribute to the understanding of the interaction between climate and ice dynamics in the context of the GrIS

    The Effectiveness of Nutritional Education Interventions on Dietary Intake in Young Black Males: A Near-Empty Systematic Review

    The incidence of several diet and lifestyle-related diseases, previously seen only in adults, is increasing in prevalence in young people. The Black population, and particularly Black males, are at high risk of developing lifestyle-related diseases. Adolescence and young adulthood are considered a transitional period with increasing independence and responsibility, along with the development of lifelong lifestyle habits. This systematic review aimed to establish which methods and approaches to nutritional education interventions are the most effective in improving the nutritional/dietary intake in healthy young Black males. Eligibility criteria were designed using PICOS and included controlled trials of nutrition education interventions designed to improve dietary intake in healthy young Black or mixed-race males aged 14–21 years old. Medline, Cinahl and Scopus were searched in April 2021, resulting in 20,375 records being screened, and subsequently 72 full-text articles were reviewed. Risk of bias was assessed using the ROBINS-I tool. One study met the eligibility criteria. Results are presented in a narrative format as meta-analysis was not possible. This systematic review revealed a lack of evidence on the effectiveness of nutritional education interventions in this high-risk population. Limitations are noted and recommendations have been made

    The Value of Health Technology Assessment: a mixed methods framework

    Whilst much research has been undertaken on establishing what factors influence improved decision-making including good governance structures, expertise, political and institutional factors, resources and participation, how such influences on decision-making interact with local context and health systems, leading to impact on health outcomes, is less understood. The focus of our research is on the impact of Health Technology Assessment (HTA) as a tool for priority-setting with its explicit consideration of costs and benefits. Where evaluations have been undertaken, they mainly focus on processes or outcomes at the decision-making level, with impact on health outcomes rarely measured. Even in countries where HTA programmes are well established, evidence which identifies their outcomes and impact in terms of health gains is limited. For countries with greater capacity constraints, how decision-making interacts with ‘context’ leading to health outcomes is even less explored and arguably of critical importance. This research aims to provide a methodological framework and evidence base to: quantify the returns on investment in HTA; and produce explanatory programme theory that considers individual, interpersonal, institutional and systems-level components and their interactions on the mechanisms by which HTA impact can be optimised

    Dimerization of ABCG2 Analysed by Bimolecular Fluorescence Complementation

    ABCG2 is one of three human ATP binding cassette transporters that are functionally capable of exporting a diverse range of substrates from cells. The physiological consequence of ABCG2 multidrug transport activity in leukaemia, and some solid tumours is the acquisition of cancer multidrug resistance. ABCG2 has a primary structure that infers that a minimal functional transporting unit would be a homodimer. Here we investigated the ability of a bimolecular fluorescence complementation approach to examine ABCG2 dimers, and to probe the role of individual amino acid substitutions in dimer formation. ABCG2 was tagged with fragments of venus fluorescent protein (vYFP), and this tagging did not perturb trafficking or function. Co-expression of two proteins bearing N-terminal and C-terminal fragments of YFP resulted in their association and detection of dimerization by fluorescence microscopy and flow cytometry. Point mutations in ABCG2 which may affect dimer formation were examined for alterations in the magnitude of fluorescence complementation signal. Bimolecular fluorescence complementation (BiFC) demonstrated specific ABCG2 dimer formation, but no changes in dimer formation, resulting from single amino acid substitutions, were detected by BiFC analysis

    The impact of COVID-19 on opportunities for adolescent girls and the role of girls\u27 groups

    Covid-19 has disrupted lives, networks, and institutions across social, economic, and health dimensions around the world. This report examines how the pandemic has affected adolescent girls and young women in particular, and explores how group-based programs for girls in low- and middle-income countries have been affected by and are responding to the pandemic. The pandemic has affected adolescent girls and young women across multiple outcomes, including education, livelihoods, sexual and reproductive health, sexual- and gender-based violence, and early and forced marriage. These heightened barriers to economic and social opportunities have the potential to derail progress toward gender equality unless recognized and addressed. Insights shared by 12 girl-centered programs revealed that girls’ groups offer a unique opportunity to identify and deliver interventions to vulnerable communities during crises, and many programs have seen an expansion in their roles during the pandemic. This report urges decisionmakers, donors, and researchers to invest in generating data and evidence to assess how Covid-19 is affecting adolescent girls and young women, and to evaluate the potential roles and effects of girls’ groups in the immediate and longer-term

    A Novel Form of Chondrocyte Stress is Triggered by a COMP Mutation Causing Pseudoachondroplasia

    Pseudoachondroplasia (PSACH) results from mutations in cartilage oligomeric matrix protein (COMP) and the p.D469del mutation within the type III repeats of COMP accounts for approximately 30% of PSACH. To determine disease mechanisms of PSACH in vivo, we introduced the Comp D469del mutation into the mouse genome. Mutant animals were normal at birth but grew slower than their wild-type littermates and developed short-limb dwarfism. In the growth plates of mutant mice chondrocyte columns were reduced in number and poorly organized, while mutant COMP was retained within the endoplasmic reticulum (ER) of cells. Chondrocyte proliferation was reduced and apoptosis was both increased and spatially dysregulated. Previous studies on COMP mutations have shown mutant COMP is co-localized with chaperone proteins, and we have reported an unfolded protein response (UPR) in mouse models of PSACH-MED (multiple epiphyseal dysplasia) harboring mutations in Comp (T585M) and Matn3, Comp etc (V194D). However, we found no evidence of UPR in this mouse model of PSACH. In contrast, microarray analysis identified expression changes in groups of genes implicated in oxidative stress, cell cycle regulation, and apoptosis, which is consistent with the chondrocyte pathology. Overall, these data suggest that a novel form of chondrocyte stress triggered by the expression of mutant COMP is central to the pathogenesis of PSACH.
