    The chemokine CXCL13 in acute neuroborreliosis

    Objective Recent studies have suggested an important role of the B cell chemoattractant CXCL13 in acute neuroborreliosis (NB). Our aim was to confirm the diagnostic role of CXCL13 and to evaluate its relevance as a therapy response and disease activity marker in NB. Methods CXCL13 was measured in cerebrospinal fluid (CSF) and serum of patients with NB (n = 28), systemic borreliosis (SB, n = 9), Guillaine-Barre syndrome (GBS, n = 11), Bell's palsy (BP, n = 19), other cranial nerve palsies (CNP, n = 5), cephalgia (C, n = 20), bacterial CNS infections (B-CNS-I, n = 16) and viral CNS infections (V-CNS-I, n = 18). For follow-up studies, serial sample pairs were evaluated from 25 patients with NB (n = 56), 11 with B-CNS-I (n = 25) and 14 with V-CNS-I (n = 36). Results CSF-CXCL13 was significantly elevated in NB compared with other neurological diseases (p<0.001). Using receiver operating characteristic analysis, 337 ng/g was determined as a cut-off with a sensitivity of 96.4% and a specificity of 96.9%. Of all the parameters investigated, CSF CXCL13 showed the fastest response to antibiotic therapy, decreasing significantly (p = 0.008) within 1 week. In untreated patients, CSF CXCL13 was elevated in patients with a short duration of disease. Borrelia burgdorferi antibody index showed no significant (p = 0.356) change over follow-up. Conclusions The study confirms the relevance of CXCL13 as a diagnostic biomarker of NB and suggests that CSF CXCL13 in NB is linked to duration of disease and could be a marker of disease activity and response to antibiotic therapy

    Canine parvovirus detected from a serval (Leptailurus serval) in South Africa

    Canine parvovirus first emerged in domestic dogs (Canis familiaris), most likely as a variant of the feline panleucopaenia virus. Relatively recently, canine parvovirus-2a and canine parvovirus-2b infections have been identified in both symptomatic and asymptomatic domestic cats, while canine parvovirus infections have also been demonstrated in wild felids. This report documents the first known case of canine parvovirus-2b detected in unvaccinated serval (Leptailurus serval) from South Africa. The serval presented with clinical signs of vomiting, anorexia and diarrhoea that responded to symptomatic treatment. Two weeks later, severe leucopaenia, thrombocytopenia and death occurred. Typical enteric histological lesions of parvovirus infection were not observed on histopathological examination of the small intestine; however, histological lesions consistent with septicaemia were present. Canine parvovirus was detected in formalin-fixed paraffin-embedded small intestine using polymerase chain reaction. Phylogenetic analysis of the sequence of the canine parvovirus viral capsid protein gene showed similarities between the sample from the serval and canine parvovirus-2b isolates from domestic dogs in Argentina and South Africa. A case of canine parvovirus-2b in a domestic dog from South Africa in 2012 that fell within the same clade as the serval sample appears distantly related because of the long branch length. The significance of these findings is explored. More extensive surveys of canine parvovirus in domestic and wild felids and canids are needed to understand the epidemiology of canine parvovirus in non-domestic felids in South Africa

    TDP-43 in the hypoglossal nucleus identifies amyotrophic lateral sclerosis in behavioral variant frontotemporal dementia

    The hypoglossal nucleus was recently identified as a key brain region in which the presence of TDP-43 pathology could accurately discriminate TDP-43 proteinopathy cases with clinical amyotrophic lateral sclerosis (ALS). The objective of the present study was to assess the hypoglossal nucleus in behavioral variant frontotemporal dementia (bvFTD), and determine whether TDP-43 in this region is associated with clinical ALS. Twenty-nine cases with neuropathological FTLD-TDP and clinical bvFTD that had not been previously assessed for hypoglossal TDP-43 pathology were included in this study. Of these 29 cases, 41% (n = 12) had a dual diagnosis of bvFTD-ALS at presentation, all 100% (n = 12) of which demonstrated hypoglossal TDP-43 pathology. Of the 59% (n = 17) cohort that presented with pure bvFTD, 35% (n = 6) were identified with hypoglossal TDP-43 pathology. Review of the case files of all pure bvFTD cases revealed evidence of possible or probable ALS in 5 of the 6 hypoglossal-positive cases (83%) towards the end of disease, and this was absent from all cases without such pathology. In conclusion, the present study validates grading the presence of TDP-43 in the hypoglossal nucleus for the pathological identification of bvFTD cases with clinical ALS, and extends this to include the identification of cases with possible ALS at end-stage

    A low-power data acquisition system for geomagnetic observatories and variometer stations

    A modern geomagnetic observatory must provide data of high stability, continuity, and resolution. The INTERMAGNET network has therefore specified quantitative criteria to ensure a high quality standard of geomagnetic observatories. Here, we present a new data acquisition system which was designed to meet these criteria, in particular with respect to 1 Hz data. This system is based on a Raspberry Pi embedded PC and runs a C+ +  data acquisition software. As a result, the data acquisition system is modular, cheap, and flexible, and it can be operated in remote areas with limited power supply. In addition, the system is capable of near-real-time data transmission, using a reverse SSH tunnel to work with any network available. The system hardware was successfully tested at the Niemegk observatory for a period of 1 year and subsequently installed at the Tatuoca observatory in Brazil

    Bartonellae of the Namaqua rock mouse, Micaelamys namaquensis (Rodentia : Muridae) from South Africa

    The aim of this study was to determine Bartonella prevalence and diversity in Namaqua rock mice, Micaelamys namaquensis, a species endemic to South Africa, which can attain pest status. A total of 100 heart samples collected monthly from March to December were screened for Bartonella genome presence using three primer sets targeting the citrate synthase (gltA) gene, the NADH dehydrogenase gamma subunit (nuoG) gene and the RNA polymerase b-subunit-encoding gene (rpoB). An overall prevalence of 44% was obtained, with no statistically significant differences or correlations between infection rates and rodent sex, month of capture or season of capture. Phylogenetic analysis of 34 unambiguous gltA sequences revealed the presence of three discrete Bartonella lineages in M. namaquensis, one of which corresponds to Bartonella elizabethae, a species with known zoonotic potential.The study was funded by grants awarded to ADSB and NCB by the South African National Research Foundation (NRF)http://www.elsevier.com/locate/vetmicab201
