Risk of elevated nitrate concentrations below forest in the region of Munich (South Bavaria) : regionalisation at the basis of remote sensing data and nested samples
Eine zielgerichtete Bewirtschaftung der WĂ€lder im Hinblick auf die fortschreitende N-SĂ€ttigung fordert entsprechende Geoinformationen. Auf der Basis der Ergebnisse der Nitratinventur Bayern (Gensior et al 2003b, Mellert et al. 2005a) wurde eine Karte des Risikos erhöhter Nitratkonzentrationen fĂŒr das Land Bayern erstellt (Mellert 2005c). Die Bayernkarte liefert Informationen ĂŒber die durchschnittliche Situation in den forstlichen Wuchsgebieten und dient der Identifizierung von Problemregionen. Als Ăbersichtskarte kann sie jedoch die BedĂŒrfnisse auf regionaler Ebene, z.B. fĂŒr ein Wuchsgebiet, kaum befriedigen. Hierzu ist eine rĂ€umliche PrĂ€zisierung der Geodaten in einem detaillierten MaĂstab erforderlich. Die bayernweite Regionalisierung basiert auf der in den Jahren 2001/2002 durchgefĂŒhrten Nitratinventur im 8 x 8 km Raster (Level-I/BZE) an 399 Punkten im Flachland. Die Anzahl von Inventurpunkten in den einzelnen Wuchsgebieten ist daher sehr begrenzt. Zur Informationsverdichtung der kleinmaĂstĂ€bigen Bayernkarte auf den gröĂeren MaĂstab der Karte fĂŒr den GroĂraum MĂŒnchen wurden detaillierte Informationen aus einer 1998 durchgefĂŒhrten Sickerwasserstudie (Rothe & Mellert 2004) herangezogen. Im vorliegenden Beitrag wird die Möglichkeit eines Downscalings durch ein genestets Verfahren vorgestellt. Die auf einem logistischem Regressionsmodell basierende Regionalisierung auf bayerischer Ebene (Meller et al. 2005c) wird hierbei mit den regionalen Daten durch ein multiples Regressionsverfahren verknĂŒpft. Dank einer ins Projekt integrierten Pilotstudie zur Fernerkundung von Waldtypen konnte eine geeignete Waldkarte fĂŒr den Raum MĂŒnchen durch Klassifikation von Landsat-Daten bereit gestellt werden.A nested model design at two different spatial scales is presented to predict the risk of elevated nitrate concentrations below the main rooting zone of forests in the region of Munich. The procedure combines a logistic regression on the Bavarian scale with a general linear model (Mellert et al. 2005c) are used as a link between both scales. This link of information on two spatial levels and the identification of predictors at different scales are the advantages of this approach, compared to a sole and independent model. At the Bavarian scale the ammonium deposition and the precipitation as well as the stand type and the site conditions predict the risk of elevated nitrate concentration. At the regional scale the degree of forestation and the stand age contribute considerably to explain the variance of the nitrate concentrations. Among the sources of nitrogen input into the forests especially regional ammonium emissions by agriculture seem to contribute to the occurrence of high nitrate concentrations. The central outcome of this study is a map indicating the risk of elevated nitrate concentrations. The results suggests that forestry could contribute to retard nitrogen saturation and nitrate leaching by conversation of spruce into mixed stands with a high percentage of broad-leaved species
Population pharmacokinetics of liposomal irinotecan in patients with cancer and exposureâsafety analyses in patients with metastatic pancreatic cancer
Pharmacokinetics; Liposomal irinotecan; SafetyFarmacocinĂ©tica; IrinotecĂĄn liposomal; SeguridadFarmacocinĂštica; Irinotecan liposomal; SeguretatLiposomal irinotecan is a liposomal formulation of irinotecan, which prolongs circulation of irinotecan and its active metabolite SN-38. A population pharmacokinetic (PK) model was developed based on data from seven studies (N = 440). Adequacy of the model was assessed using multiple methods, including visual predictive check. Associations between PK exposure and the incidence of diarrhea (grade â„3) and neutropenia adverse events (AEs) (grade â„3) at first event in patients with metastatic pancreatic ductal adenocarcinoma (mPDAC) were investigated using logistic regression based on data from two studies (the phase III NAPOLI-1 [N = 260] and phase I/II NCT02551991 [N = 56] trials). The PKs of total irinotecan was described by a two-compartment model with first-order elimination, with SN-38 formed directly by a first-order constant from the central compartment of irinotecan or after using a transit compartment. Clearance was 17.9 L/week (0.107 L/h) and 19,800 L/week (118 L/h) for total irinotecan and SN-38, respectively. The UGT1A1*28 7/7 homozygous genotype had no significant impact on SN-38 clearance. Model evaluation was satisfactory for both irinotecan and SN-38. The incidence of diarrhea (grade â„3) at first event was significantly higher with increasing average concentrations of total irinotecan and SN-38; there was no significant association between an increased risk of neutropenia AEs (grade â„3) at first event and average SN-38 concentrations. In summary, the PKs of total irinotecan and SN-38 after administration of liposomal irinotecan were well-described by the model. The UGT1A1*28 status had no significant impact on the PKs of liposomal irinotecan
Overview of model-building strategies in population PK/PD analyses: 2002-2004 literature survey.
AIMS: A descriptive survey of published population pharmacokinetic and/or pharmacodynamic (PK/PD) analyses from 2002 to 2004 was conducted and an evaluation made of how model building was performed and reported. METHODS: We selected 324 articles in Pubmed using defined keywords. A data abstraction form (DAF) was then built comprising two parts: general characteristics including article identification, context of the analysis, description of clinical studies from which the data arose, and model building, including description of the processes of modelling. The papers were examined by two readers, who extracted the relevant information and transmitted it directly to a MySQL database, from which descriptive statistical analysis was performed. RESULTS: Most published papers concerned patients with severe pathology and therapeutic classes suffering from narrow therapeutic index and/or high PK/PD variability. Most of the time, modelling was performed for descriptive purposes, with rich rather than sparse data and using NONMEM software. PK and PD models were rarely complex (one or two compartments for PK; E(max) for PD models). Covariate testing was frequently performed and essentially based on the likelihood ratio test. Based on a minimal list of items that should systematically be found in a population PK-PD analysis, it was found that only 39% and 8.5% of the PK and PD analyses, respectively, published from 2002 to 2004 provided sufficient detail to support the model-building methodology. CONCLUSIONS: This survey allowed an efficient description of recent published population analyses, but also revealed deficiencies in reporting information on model building
Extreme Ultraviolet Reflective Grating Characterization and Simulationsfor the Aspera SmallSat Mission
The Aspera SmallSat mission is designed to detect and map the warm-hot gaseous component of the halos of nearby galaxies through long-slit spectroscopy of the ionized O VI emission line (103.2 nm) for the first time. The Aspera Rowland circle type spectrograph uses a toroidal grating coated with a multilayer film consisting of aluminum, lithium fluoride, and magnesium fluoride capping to optimize reflectivity in the extreme ultraviolet (EUV) waveband from 103 to 104nm. We discuss the grating characterization test setup at the University of Arizona (UA), which will validate the multilayer coating and grating efficiency in a UV vacuum chamber. We also simulate the reflectivity of the multilayer thin film coating using IMD IDL software to compare simulated results with measured reflectivity. Additionally, non-sequential ray trace simulations and 3D CAD modeling are used for verification of the test setup. Finally, the implications of the differences between the measured and simulated reflectivity and grating efficiencies are considered, including impact to the mission

Computing normalised prediction distribution errors to evaluate nonlinear mixed-effect models: The npde add-on package for R.
International audiencePharmacokinetic/pharmacodynamic data are often analysed using nonlinear mixed-effect models, and model evaluation should be an important part of the analysis. Recently, normalised prediction distribution errors (npde) have been proposed as a model evaluation tool. In this paper, we describe an add-on package for the open source statistical package R, designed to compute npde. npde take into account the full predictive distribution of each individual observation and handle multiple observations within subjects. Under the null hypothesis that the model under scrutiny describes the validation dataset, npde should follow the standard normal distribution. Simulations need to be performed before hand, using for example the software used for model estimation. We illustrate the use of the package with two simulated datasets, one under the true model and one with different parameter values, to show how npde can be used to evaluate models. Model estimation and data simulation were performed using NONMEM version 5.1
