    To Be "High" and "Fine": Quilts, Art, and Power, 1971-1991

    This thesis examines the status of quilts in the Western art world from 1971-1991 as point of entry into the complex process of classifying "art." Each chapter focuses on one of four art-world groups that represent the key contributors to the growing debate over quilts as art. Analysis of their strategies reveals the how quilts-as-art proponents advanced multiple agendas. The quilt became a symbol of power: for art museums and curators to demonstrate cultural hegemony, for art critics to legitimize their role as arbiters of taste, for feminist scholars to expose the oppression of women, and for collectors and dealers to enhance their economic and social status. At the dawn of the 1990s, the art world had granted only limited acceptance to quilts. Although the art world's prevailing gender- and class-based hierarchy remained largely intact, quilts' partial move into the art world had implications for the quiltmaking community

    Discovery and Development of a Small Molecule Library with Lumazine Synthase Inhibitory Activity

    (E)-5-Nitro-6-(2-hydroxystyryl)pyrimidine-2,4(1H,3H)-dione (9) was identified as a novel inhibitor of Schizosaccharomyces pombe lumazine synthase by high-throughput screening of a 100,000 compound library. The Ki of 9 vs. Mycobacterium tuberculosis lumazine synthase was 95 μM. Compound 9 is a structural analog of the lumazine synthase substrate, 5-amino-6-(D-ribitylamino)-2,4-(1H,3H)pyrimidinedione (1). This indicates that the ribitylamino side chain of the substrate is not essential for binding to the enzyme. Optimization of the enzyme inhibitory activity through systematic structure modification of the lead compound 9 led to (E)-5-nitro-6-(4-nitrostyryl)pyrimidine-2,4(1H,3H)-dione (26), which has a Ki of 3.7 μM vs. M. tuberculosis lumazine synthase

    This project is an innovative initiative involving the College of Engineering at Rowan University and the Center for Aquatic Sciences (CAS) at Adventure Aquarium in Camden, New Jersey to enhance STEM education at all levels. Three other educational institutions will have a direct impact from our proposed activities. These include the Rowan College at Gloucester County (RCGC), Rowan College at Burlington County (RCBC) and the STEM Academy at Glassboro High School. The project uses algae to introduce engineering students to the Global Grand Challenges of the 21st Century with a strong focus on combining critical reflection through activities that invest in the humanities especially literary readings, sociology, environmental justice and public policy. While everyone recognizes algae as a photosynthetic organism that is ubiquitous, it is rare that that students make a connection to the prospect of this microbe playing a significant role in impacting the future of this world. The use of algae can range from biofuels, carbon sequestration, nutrient removal to use in the nutraceutical industry. The algae production facility is the backbone of CAS activities at the Adventure Aquarium as it is food for various zooplankton, filter feeders or larval fishes. Students are engaged in the scientific discovery process using both exciting hands-on activities and cyberlearning experiences that introduce chemical, mechanical, electrical engineering, civil and environmental principles such as mass and energy balances; fluid flow; work, energy, and efficiency; water quality and treatment; unit physical, chemical and biological processes; microbiology; power and electrical signal processing. The algae theme also adds to the need for an understanding of biological systems, ecosystems, pollution, alternate energy and sustainable development. A strong unique focus of this project is in combining critical reflection through activities that invest in the humanities

    Production of small cysteine-rich effector proteins in Escherichia coli for structural and functional studies

    Although the lifestyles and infection strategies of plant pathogens are diverse, a prevailing feature is the use of an arsenal of secreted proteins, known as effectors, which aid in microbial infection. In the case of eukaryotic filamentous pathogens, such as fungi and oomycetes, effector proteins are typically dissimilar, at the protein sequence level, to known protein families and functional domains. Consequently, we currently have a limited understanding of how fungal and oomycete effectors promote disease. Protein biochemistry and structural biology are two methods that can contribute greatly to the understanding of protein function. Both techniques are dependent on obtaining proteins that are pure and functional, and generally require the use of heterologous recombinant protein expression systems. Here, we present a general scheme and methodology for the production and characterization of small cysteine-rich (SCR) effectors utilizing Escherichia coli expression systems. Using this approach, we successfully produced cysteine-rich effectors derived from the biotrophic fungal pathogen Melampsora lini and the necrotrophic fungal pathogen Parastagonospora nodorum. Access to functional recombinant proteins facilitated crystallization and functional experiments. These results are discussed in the context of a general workflow that may serve as a template for others interested in understanding the function of SCR effector(s) from their plant pathogen(s) of interest.This research was supported by the Australian Research Council (ARC) Discovery Projects DP120100685 and DP120103558.     Trajectories of depression and anxiety symptom severity during psychological therapy for common mental health problems

    BACKGROUND: There is substantial variation in patient symptoms following psychological therapy for depression and anxiety. However, reliance on endpoint outcomes ignores additional interindividual variation during therapy. Knowing a patient's likely symptom trajectories could guide clinical decisions. We aimed to identify latent classes of patients with similar symptom trajectories over the course of psychological therapy and explore associations between baseline variables and trajectory class. METHODS: Patients received high-intensity psychological treatment for common mental health problems at National Health Service Improving Access to Psychological Therapies services in South London (N = 16 258). To identify trajectories, we performed growth mixture modelling of depression and anxiety symptoms over 11 sessions. We then ran multinomial regressions to identify baseline variables associated with trajectory class membership. RESULTS: Trajectories of depression and anxiety symptoms were highly similar and best modelled by four classes. Three classes started with moderate-severe symptoms and showed (1) no change, (2) gradual improvement, and (3) fast improvement. A final class (4) showed initially mild symptoms and minimal improvement. Within the moderate-severe baseline symptom classes, patients in the two showing improvement as opposed to no change tended not to be prescribed psychotropic medication or report a disability and were in employment. Patients showing fast improvement additionally reported lower baseline functional impairment on average. CONCLUSIONS: Multiple trajectory classes of depression and anxiety symptoms were associated with baseline characteristics. Identifying the most likely trajectory for a patient at the start of treatment could inform decisions about the suitability and continuation of therapy, ultimately improving patient outcomes

    The development and validation of the Healthy Computing Questionnaire for Children (HCQC)

    BACKGROUND: School children may be at risk of physical health problems associated with desktop, laptop and tablet computer use. There are some interventions designed to educate children about ergonomic practices when using computers. A common limitation of past intervention studies has been the lack of a valid and reliable questionnaire to determine effectiveness of the intervention. OBJECTIVE: The aim of this study was to develop a valid and reliable self-report measure to assess primary school children’s knowledge, attitudes and behaviours related to healthy computer use. METHODS: A mixed methods approach was used to develop the questionnaire and test its psychometric properties. A convenience sample of 440 primary school children in Year 5 (aged 9–11 years) was used in the development and validation of the questionnaire. RESULTS: The final questionnaire comprised 56 items across the three subscales of knowledge, attitudes and behaviour. The questionnaire was shown to have good content validity and adequate test-retest reliability. Internal consistency was adequate for the attitude items, but low for the knowledge items. CONCLUSIONS: This study produced a valid and reliable tool, using a health promotion framework, for measuring primary school children’s knowledge, attitudes and behaviours related to healthy computing
