PEAK RATE OF TRUNK ENERGY OUTFLOW DIFFERS BETWEEN PITCH TYPES IN SOFTBALL PITCHERS
In softball players, it is unclear how certain pitch types may increase the risk of injury. The purpose of this study was to determine energy flow differences in the trunk and upper-arm segments between pitch types. Twenty-three softball pitchers participated. Absolute values of trunk energy inflow (IF) and outflow (OF), and upper arm IF, as well as segment energy flow when normalized to pitch speed were assessed in three pitch types. Differences between trunk energy OF were found between fastballs compared to curveballs and dropballs. When normalized to pitch speed, trunk energy OF only differed between fastballs and dropballs. For the upper arm, absolute differences were found between the fastball and curveball. Similar rates of humerus IF between the fastball and dropball and less trunk outflow in the dropball may indicate increased upper extremity demands in the dropball
Radial and circumferential flow surveys at the inlet and exit of the Space Shuttle Main Engine High Pressure Fuel Turbine Model
The main objective of this test was to obtain detailed radial and circumferential flow surveys at the inlet and exit of the SSME High Pressure Fuel Turbine model using three-hole cobra probes, hot-film probes, and a laser velocimeter. The test was designed to meet several objectives. First, the techniques for making laser velocimeter, hot-film probe, and cobra probe measurements in turbine flows were developed and demonstrated. The ability to use the cobra probes to obtain static pressure and, therefore, velocity had to be verified; insertion techniques had to be established for the fragile hot-film probes; and a seeding method had to be established for the laser velocimetry. Once the measurement techniques were established, turbine inlet and exit velocity profiles, temperature profiles, pressure profiles, turbulence intensities, and boundary layer thicknesses were measured at the turbine design point. The blockage effect due to the model inlet and exit total pressure and total temperature rakes on the turbine performance was also studied. A small range of off-design points were run to obtain the profiles and to verify the rake blockage effects off-design. Finally, a range of different Reynolds numbers were run to study the effect of Reynolds number on the various measurements
EFFICIENCY INDEX USED TO ASSESS SHOULDER STRESS IN COLLEGE SOFTBALL PITCHERS THROUGHOUT A SIMULATED GAME
Shoulder distraction forces in softball pitching are known to have a positive impact on performance yet a negative impact on musculoskeletal health. The purpose of this study was to assess changes in shoulder stress across innings pitched using Efficiency Arm-Stress Index (EASI) scores. Motion capture was used on collegiate softball pitchers pitching a simulated game. Peak shoulder distraction force was obtained using inverse dynamics procedures and used to calculate an EASI score (fastball velocity divided by peak shoulder distraction force in percent body weight). A RM·ANOVA revealed inning had no effect on EASI score (F[6,7]=1.28, p=0.286). Understanding a pitcher’s efficiency score may help shape individual pitching loads. Future work should investigate clinically meaningful changes in efficiency scores and mechanisms behind low efficiencies

The paediatric flat foot and general anthropometry in 140 Australian school children aged 7 - 10 years
<p>Abstract</p> <p>Background</p> <p>Many studies have found a positive relationship between increased body weight and flat foot posture in children.</p> <p>Methods</p> <p>From a study population of 140 children aged seven to 10 years, a sample of 31 children with flat feet was identified by screening with the FPI-6. Basic anthropometric measures were compared between subjects with and without flat feet as designated.</p> <p>Results</p> <p>The results of this study, in contrast to many others, question the association of flat feet and heavy children. A significant relationship between foot posture and weight (FPI (L) r = -0.186 (p < 0.05), FPI(R) r = -0.194 (p < 0.05), waist girth (FPI (L) r = -0.213 (p < 0.05), FPI(R) r = -0.228 (p < 0.01) and BMI (FPI (L) r = -0.243 (p < 0.01), FPI(R) r = -0.263 (p < 0.01) was identified, but was both weak and inverse.</p> <p>Conclusions</p> <p>This study presents results which conflict with those of many previous investigations addressing the relationship between children's weight and foot posture. In contrast to previous studies, the implication of these results is that heavy children have less flat feet. Further investigation is warranted using a standardized approach to assessment and a larger sample of children to test this apparent contradiction.</p
Creatine Protects against Excitoxicity in an In Vitro Model of Neurodegeneration
Creatine has been shown to be neuroprotective in aging, neurodegenerative conditions and brain injury. As a common molecular background, oxidative stress and disturbed cellular energy homeostasis are key aspects in these conditions. Moreover, in a recent report we could demonstrate a life-enhancing and health-promoting potential of creatine in rodents, mainly due to its neuroprotective action. In order to investigate the underlying pharmacology mediating these mainly neuroprotective properties of creatine, cultured primary embryonal hippocampal and cortical cells were challenged with glutamate or H2O2. In good agreement with our in vivo data, creatine mediated a direct effect on the bioenergetic balance, leading to an enhanced cellular energy charge, thereby acting as a neuroprotectant. Moreover, creatine effectively antagonized the H2O2-induced ATP depletion and the excitotoxic response towards glutamate, while not directly acting as an antioxidant. Additionally, creatine mediated a direct inhibitory action on the NMDA receptor-mediated calcium response, which initiates the excitotoxic cascade. Even excessive concentrations of creatine had no neurotoxic effects, so that high-dose creatine supplementation as a health-promoting agent in specific pathological situations or as a primary prophylactic compound in risk populations seems feasible. In conclusion, we were able to demonstrate that the protective potential of creatine was primarily mediated by its impact on cellular energy metabolism and NMDA receptor function, along with reduced glutamate spillover, oxidative stress and subsequent excitotoxicity
Genome-Wide Gene-Environment Study Identifies Glutamate Receptor Gene GRIN2A as a Parkinson's Disease Modifier Gene via Interaction with Coffee
Our aim was to identify genes that influence the inverse association of coffee with the risk of developing Parkinson's disease (PD). We used genome-wide genotype data and lifetime caffeinated-coffee-consumption data on 1,458 persons with PD and 931 without PD from the NeuroGenetics Research Consortium (NGRC), and we performed a genome-wide association and interaction study (GWAIS), testing each SNP's main-effect plus its interaction with coffee, adjusting for sex, age, and two principal components. We then stratified subjects as heavy or light coffee-drinkers and performed genome-wide association study (GWAS) in each group. We replicated the most significant SNP. Finally, we imputed the NGRC dataset, increasing genomic coverage to examine the region of interest in detail. The primary analyses (GWAIS, GWAS, Replication) were performed using genotyped data. In GWAIS, the most significant signal came from rs4998386 and the neighboring SNPs in GRIN2A. GRIN2A encodes an NMDA-glutamate-receptor subunit and regulates excitatory neurotransmission in the brain. Achieving P2df = 10−6, GRIN2A surpassed all known PD susceptibility genes in significance in the GWAIS. In stratified GWAS, the GRIN2A signal was present in heavy coffee-drinkers (OR = 0.43; P = 6×10−7) but not in light coffee-drinkers. The a priori Replication hypothesis that “Among heavy coffee-drinkers, rs4998386_T carriers have lower PD risk than rs4998386_CC carriers” was confirmed: ORReplication = 0.59, PReplication = 10−3; ORPooled = 0.51, PPooled = 7×10−8. Compared to light coffee-drinkers with rs4998386_CC genotype, heavy coffee-drinkers with rs4998386_CC genotype had 18% lower risk (P = 3×10−3), whereas heavy coffee-drinkers with rs4998386_TC genotype had 59% lower risk (P = 6×10−13). Imputation revealed a block of SNPs that achieved P2df<5×10−8 in GWAIS, and OR = 0.41, P = 3×10−8 in heavy coffee-drinkers. This study is proof of concept that inclusion of environmental factors can help identify genes that are missed in GWAS. Both adenosine antagonists (caffeine-like) and glutamate antagonists (GRIN2A-related) are being tested in clinical trials for treatment of PD. GRIN2A may be a useful pharmacogenetic marker for subdividing individuals in clinical trials to determine which medications might work best for which patients
Plasticity in D1-Like Receptor Expression Is Associated with Different Components of Cognitive Processes
Dopamine D1-like receptors consist of D1 (D1A) and D5 (D1B) receptors and play a key role in working memory. However, their possibly differential contribution to working memory is unclear. We combined a working memory training protocol with a stepwise increase of cognitive subcomponents and real-time RT-PCR analysis of dopamine receptor expression in pigeons to identify molecular changes that accompany training of isolated cognitive subfunctions. In birds, the D1-like receptor family is extended and consists of the D1A, D1B, and D1D receptors. Our data show that D1B receptor plasticity follows a training that includes active mental maintenance of information, whereas D1A and D1D receptor plasticity in addition accompanies learning of stimulus-response associations. Plasticity of D1-like receptors plays no role for processes like response selection and stimulus discrimination. None of the tasks altered D2 receptor expression. Our study shows that different cognitive components of working memory training have distinguishable effects on D1-like receptor expression
α-Synuclein Genetic Variants Predict Faster Motor Symptom Progression in Idiopathic Parkinson Disease
Currently, there are no reported genetic predictors of motor symptom progression in Parkinson’s disease (PD). In familial PD, disease severity is associated with higher α-synuclein (SNCA) expression levels, and in postmortem studies expression varies with SNCA genetic variants. Furthermore, SNCA is a well-known risk factor for PD occurrence. We recruited Parkinson’s patients from the communities of three central California counties to investigate the influence of SNCA genetic variants on motor symptom progression in idiopathic PD. We repeatedly assessed this cohort of patients over an average of 5.1 years for motor symptom changes employing the Unified Parkinson’s Disease Rating Scale (UPDRS). Of 363 population-based incident PD cases diagnosed less than 3 years from baseline assessment, 242 cases were successfully re-contacted and 233 were re-examined at least once. Of subjects lost to follow-up, 69% were due to death. Adjusting for covariates, risk of faster decline of motor function as measured by annual increase in motor UPDRS exam score was increased 4-fold in carriers of the REP1 263bp promoter variant (OR 4.03, 95%CI:1.57–10.4). Our data also suggest a contribution to increased risk by the G-allele for rs356165 (OR 1.66; 95%CI:0.96–2.88), and we observed a strong trend across categories when both genetic variants were considered (p for trend = 0.002). Our population-based study has demonstrated that SNCA variants are strong predictors of faster motor decline in idiopathic PD. SNCA may be a promising target for therapies and may help identify patients who will benefit most from early interventions. This is the first study to link SNCA to motor symptom decline in a longitudinal progression study
