    The Capsicum terpenoid biosynthetic module is affected by spider-mite herbivory

    In response to herbivory, Capsicum annuum leaves adapt their specialized metabolome that may protect the plant against herbivore feeding either directly or indirectly through volatile metabolites acting as cues for natural enemies of the herbivore. The volatile blend of spider-mite infested leaves differs from non-challenged leaves predominantly by a higher contribution of mono- and sesquiterpenes. In addition to these terpenoids released into the headspace, the terpenoid composition of the leaves alters upon herbivory. All this suggests an important role for terpenoids and their biosynthetic machinery in the defence against herbivory. Here, we show that the C. annuum genome contains a terpene synthase (TPS) gene family of 103 putative members of which structural analysis revealed that 27 encode functional enzymes. Transcriptome analysis showed that several TPS loci were differentially expressed upon herbivory in leaves of two C. annuum genotypes, that differ in susceptibility towards spider mites. The relative expression of upstream biosynthetic genes from the mevalonate and the methylerythritol phosphate pathway also altered upon herbivory, revealing a shift in the metabolic flux through the terpene biosynthetic module. The expression of multiple genes potentially acting downstream of the TPSs, including cytochrome P450 monooxygenases, UDP-glucosyl transferases, and transcription factors strongly correlated with the herbivory-induced TPS genes. A selection of herbivory-induced TPS genes was functionally characterized through heterologous expression and the products that these enzymes catalysed matched with the volatile and non-volatile terpenoids induced in response to herbivory

    All the choice and all the responsibilities: an exploration of the agency perceived by women with children around their childbearing

    Findings from an exploratory study of agency around childbearing perceived by women with children from a feminist perspective are presented. Contemporary women‘s childbearing has confusedly been portrayed as chosen but constrained and also a duty. Most recently women have been considered to have a duty to reproduce to stimulate economic growth. Furthermore, a plethora of fertility theories have been put forward to explain recent declining fertility but these have found to be incapable of explaining fertility trends and the complexity of childbearing negotiations. Amongst these deliberations women‘s interests tend to get lost. This research attempts to reverse that tendency. Taking part in this research were 26 women each of whom had at least one child of nine years of age or younger from in and around Orange, NSW. Data was collected using in-depth semi-structured interviews and focus groups. As agency is ill-defined within sociology, seven criteria for recognising agency were constructed from a review of sociological theory. These criteria were used as a series of lenses through which the data were viewed; allowing for the explanatory powers of theories to be compared. The findings show that women‘s agency around childbearing was intermittent and imperfect but not completely absent. Important to the discourse of choice prevalent in the popular media and expounded by economic rationalists, the concept of choice was shown to be simplistic. Rather the women described complex negotiations between biological factors, social influences and personal preferences. This research demonstrates that despite making some progress in workforce participation, women‘s expected role in the home curtailed what they could achieve. The thesis furthers understanding of women‘s childbearing agency, has implications for public policy, provides insights into the relevance of sociological theories to women who have children and provides a novel methodological approach for assessing agency

    Probing Non-Standard Neutrino Interactions with Neutrino Factories

    We discuss the sensitivity reach of a neutrino factory measurement to non-standard neutrino interactions (NSI), which may exist as a low-energy manifestation of physics beyond the Standard Model. We use the muon appearance mode \nu_e --> \nu_\mu and consider two detectors, one at 3000 km and the other at 7000 km. Assuming the effects of NSI at the production and the detection are negligible, we discuss the sensitivities to NSI and the simultaneous determination of \theta_{13} and \delta by examining the effects in the neutrino propagation of various systems in which two NSI parameters \epsilon_{\alpha \beta} are switched on. The sensitivities to off-diagonal \epsilon's are found to be excellent up to small values of \theta_{13}. We demonstrate that the two-detector setting is powerful enough to resolve the \theta_{13}-NSI confusion problem. We believe that the results obtained in this paper open the door to the possibility of using neutrino factory as a discovery machine for NSI while keeping its primary function of performing precision measurements of the lepton mixing parameters.Comment: 47 pages, 22 figures.     Mechanical stress and inflammation have opposite effects on Wnt signaling in human chondrocytes

    Dysregulation of Wingless and Int-1 (Wnt) signaling has been strongly associated with development and progression of osteoarthritis (OA). Here, we set out to investigate the independent effects of either mechanical stress (MS) or inflammation on Wnt signaling in human neocartilage pellets, and to relate this Wnt signaling to OA pathophysiology. OA synovium-conditioned media (OAS-CM) was collected after incubating synovium from human end-stage OA joints for 24 h in medium. Cytokine levels in the OAS-CM were determined with a multiplex immunoassay (Luminex). Human neocartilage pellets were exposed to 20% MS, 2% OAS-CM or 1 ng/mL Interleukin-1 & beta; (IL-1 & beta;). Effects on expression levels of Wnt signaling members were determined by reverse transcription-quantitative polymerase chain reaction. Additionally, the expression of these members in articular cartilage from human OA joints was analyzed in association with joint space narrowing (JSN) and osteophyte scores. Protein levels of IL-1 & beta;, IL-6, IL-8, IL-10, tumor necrosis factor & alpha;, and granulocyte-macrophage colony-stimulating factor positively correlated with each other. MS increased noncanonical WNT5A and FOS expression. In contrast, these genes were downregulated upon stimulation with OAS-CM or IL-1 & beta;. Furthermore, Wnt inhibitors DKK1 and FRZB decreased in response to OAS-CM or IL-1 & beta; exposure. Finally, expression of WNT5A in OA articular cartilage was associated with increased JSN scores, but not osteophyte scores. Our results demonstrate that MS and inflammatory stimuli have opposite effects on canonical and noncanonical Wnt signaling in human neocartilage. Considering the extent to which MS and inflammation contribute to OA in individual patients, we hypothesize that targeting specific Wnt pathways offers a more effective, individualized approach.Orthopaedics, Trauma Surgery and Rehabilitatio

    Balloon atrial septostomy for transposition of the great arteries: safety and experience with the Z-5 balloon catheter

    BackgroundBalloon atrial septostomy (BAS) is an emergent and essential cardiac intervention to enhance intercirculatory mixing at atrial level in deoxygenated patients diagnosed with transposition of the great arteries (TGA) and restrictive foramen ovale. The recent recall of several BAS catheters and the changes in the European legal framework for medical devices (MDR 2017/745), has led to an overall scarcity of BAS catheters and raised questions about the use, safety, and experience of the remaining NuMED Z-5 BAS catheter.AimsTo evaluate and describe the practice and safety of the Z-5 BAS catheter, and to compare it to the performance of other BAS catheters.MethodsA retrospective single-center cohort encompassing all BAS procedures performed with the Z-5 BAS catheter in TGA patients between 1999 and 2022.ResultsA total of 182 BAS procedures were performed in 179 TGA-newborns at Day 1 (IQR 0–5) days after birth, with median weight of 3.4 (IQR 1.2–5.7) kg. The need for BAS was urgent in 90% of patients. The percentage of BAS procedures performed at bedside increased over time from 9.8% (before 2010) to 67% (2017–2022). Major complication rate was 2.2%, consisting of cerebral infarction (1.6%) and hypovolemic shock (0.5%). The rate of minor complications was 9.3%, including temporary periprocedural AV-block (3.8%), femoral vein thrombosis (2.7%), transient intracardiac thrombus (0.5%), and atrial flutter (2.2%). BAS procedures performed at bedside and in the cardiac catheterization laboratory had similar complication rates.ConclusionsBAS using the Z-5 BAS catheter is both feasible and safe at bedside and at the cardiac catheterization laboratory with minimal major complications.Thoracic Surger
