EVALUATING USE OF OUTLOOK INFORMATION IN GRAIN SORGHUM STORAGE DECISIONS
This study examines grain sorghum storage decisions in the Texas Coastal Bend region. Decisions involving use and non-use of outlook information are compared using stochastic dominance criteria. Results indicate outlook information is of value to most classes of decision-makers. The value of outlook information, however, is contingent upon producers' risk preferences. The methodology presented could be used to evaluate a more extensive set of marketing strategies for grain sorghum as well as for other crops.Crop Production/Industries,
Cloning, purification and characterization of the 6-phospho-3-hexulose isomerase YckF from Bacillus subtilis
The enzyme 6-phospho-3-hexulose isomerase (YckF) from Bacillus subtilis has been prepared and crystallized in a form suitable for X-ray crystallographic analysis. Crystals were grown by the hanging-drop method at 291 K using polyethylene glycol 2000 monomethylether as precipitant. They diffract beyond 1.7 A using an in-house Cu Kalpha source and belong to either space group P6(5)22 or P6(1)22, with unit-cell parameters a = b = 72.4, c = 241.2 A, and have two molecules of YckF in the asymmetric unit
Water and sanitation infrastructure for health: The impact of foreign aid
<p>Abstract</p> <p>Background</p> <p>The accessibility to improved water and sanitation has been understood as a crucial mechanism to save infants and children from the adverse health outcomes associated with diarrheal disease. This knowledge stimulated the worldwide donor community to develop a specific category of aid aimed at the water and sanitation sector. The actual impact of this assistance on increasing population access to improved water and sanitation and reducing child mortality has not been examined.</p> <p>Methods</p> <p>We performed a country-level analysis of the relationship between water and sanitation designated official development assistance (WSS-ODA) per capita, water and sanitation coverage, and infant and child mortality in low-income countries as defined by the World Bank. We focused our inquiry to aid effectiveness since the establishment of the Millennium Development Goals (MDGs).</p> <p>Results</p> <p>Access to improved water has consistently improved since 2002. Countries receiving the most WSS-ODA ranged from odds ratios of 4 to 18 times more likely than countries in the lowest tertile of assistance to achieve greater gains in population access to improved water supply. However, while there were modestly increased odds of sanitation access, these were largely non-significant. The countries with greatest gains in sanitation were 8-9 times more likely to have greater reductions in infant and child mortality.</p> <p>Conclusions</p> <p>Official development assistance is importantly impacting access to safe water, yet access to improved sanitation remains poor. This highlights the need for decision-makers to be more intentional with allocating WSS-ODA towards sanitation projects.</p
Novel penalised likelihood reconstruction of PET in the assessment of histologically verified small pulmonary nodules
OBJECTIVES: Investigate the effect of a novel Bayesian penalised likelihood (BPL) reconstruction algorithm on analysis of pulmonary nodules examined with 18F-FDG PET/CT, and to determine its effect on small, sub-10-mm nodules. METHODS: 18F-FDG PET/CTs performed for nodule evaluation in 104 patients (121 nodules) were retrospectively reconstructed using the new algorithm, and compared to time-of-flight ordered subset expectation maximisation (OSEM) reconstruction. Nodule and background parameters were analysed semi-quantitatively and visually. RESULTS: BPL compared to OSEM resulted in statistically significant increases in nodule SUV(max) (mean 5.3 to 8.1, p < 0.00001), signal-to-background (mean 3.6 to 5.3, p < 0.00001) and signal-to-noise (mean 24 to 41, p < 0.00001). Mean percentage increase in SUV(max) (%ΔSUV(max)) was significantly higher in nodules ≤10 mm (n = 31, mean 73 %) compared to >10 mm (n = 90, mean 42 %) (p = 0.025). Increase in signal-to-noise was higher in nodules ≤10 mm (224 %, mean 12 to 27) compared to >10 mm (165 %, mean 28 to 46). When applying optimum SUV(max) thresholds for detecting malignancy, the sensitivity and accuracy increased using BPL, with the greatest improvements in nodules ≤10 mm. CONCLUSION: BPL results in a significant increase in signal-to-background and signal-to-noise compared to OSEM. When semi-quantitative analyses to diagnose malignancy are applied, higher SUV(max) thresholds may be warranted owing to the SUV(max) increase compared to OSEM. KEY POINTS: • Novel Bayesian penalised likelihood PET reconstruction was applied for lung nodule evaluation. • This was compared to current standard of care OSEM reconstruction. • The novel reconstruction generated significant increases in lung nodule signal-to-background and signal-to-noise. • These increases were highest in small, sub-10-mm pulmonary nodules. • Higher SUV(max)thresholds may be warranted when using semi-quantitative analyses to diagnose malignancy

Large-scale discovery of enhancers from human heart tissue.
Development and function of the human heart depend on the dynamic control of tissue-specific gene expression by distant-acting transcriptional enhancers. To generate an accurate genome-wide map of human heart enhancers, we used an epigenomic enhancer discovery approach and identified ∼6,200 candidate enhancer sequences directly from fetal and adult human heart tissue. Consistent with their predicted function, these elements were markedly enriched near genes implicated in heart development, function and disease. To further validate their in vivo enhancer activity, we tested 65 of these human sequences in a transgenic mouse enhancer assay and observed that 43 (66%) drove reproducible reporter gene expression in the heart. These results support the discovery of a genome-wide set of noncoding sequences highly enriched in human heart enhancers that is likely to facilitate downstream studies of the role of enhancers in development and pathological conditions of the heart
Weather Avoidance Guidelines for NASA Global Hawk High-Altitude Unmanned Aircraft Systems (UAS)
The current Global Hawk flight rules would probably not have been effective in the single event of greatest concern (the Emily encounter). The cloud top had not reached 50,000 ft until minutes before the encounter. The TOT and lightning data would not have been available until near the overflight time since this was a rapidly growing cell. This case would have required a lastminute diversion when lightning became frequent. Avoiding such a cell probably requires continual monitoring of the forward camera and storm scope, whether or not cloud tops have been exceeding specific limits. However, the current overflight rules as strictly interpreted would have prohibited significant fractions of the successful Global Hawk overpasses of Karl and Matthew that proved not to be hazardous. Many other high altitude aircraft (ER2 and Global Hawk) flights in NASA tropical cyclone field programs have successfully overflown deep convective clouds without incident.The convective cell that caused serious concern about the safety of the ER2 in Emily was especially strong for a tropical cyclone environment, probably as strong or stronger than any that was overflown by the ER2 in 20 previous flights over tropical cyclones. Specifically, what made that cell a safety concern was the magnitude of the vertical velocity of the updraft, at least 20 m/s (4000 ft/minute) at the time the ER2 overflew it. Such a strong updraft can generate strong gravity waves at and above the tropopause, posing a potential danger to aircraft far above the maximum altitude of the updraft itself or its associated cloud top. Indeed, the ER2 was probably at least 9000 ft above that cloud top. Cloudtop height, by itself, is not an especially good indicator of the intensity of convection and the likelihood of turbulence. Nor is overflying high cloud tops (i.e. > 50,000 ft) of particular concern unless there is other evidence of very strong convective updrafts beneath those tops in the path of the aircraft. center dot Lightning, especially lightning with a high flash rate, is well correlated with convective intensity. Lightning with a minimal flash rate (say 13 flashes per minute) is indicative of updraft speeds of about 10 m/s in the mixed phase region where charge is being separated, generally at altitudes about 2025 kft in a hurricane. That is still stronger than typical updrafts (more like 5 m/s). An unresolved issue is whether there is a high and instantaneous correlation between vertical velocity in the middle troposphere (necessary for lightning generation) and near cloud top (more direct concern for overflights)
18F-FDG PET/CT assessment of histopathologically confirmed mediastinal lymph nodes in non-small cell lung cancer using a penalised likelihood reconstruction
Purpose To investigate whether using a Bayesian penalised likelihood reconstruction (BPL) improves signal-to-background (SBR), signal-to-noise (SNR) and SUVmax when evaluating mediastinal nodal disease in non-small cell lung cancer (NSCLC) compared to ordered subset expectation maximum (OSEM) reconstruction. Materials and methods 18F-FDG PET/CT scans for NSCLC staging in 47 patients (112 nodal stations with histopathological confirmation) were reconstructed using BPL and compared to OSEM. Node and multiple background SUV parameters were analysed semi-quantitatively and visually. Results Comparing BPL to OSEM, there were significant increases in SUVmax (mean 3.2–4.0, p<0.0001), SBR (mean 2.2–2.6, p<0.0001) and SNR (mean 27.7–40.9, p<0.0001). Mean background SNR on OSEM was 10.4 (range 7.6–14.0), increasing to 12.4 (range 8.2–16.7, p<0.0001). Changes in background SUVs were minimal (largest mean difference 0.17 for liver SUVmean, p<0.001). There was no significant difference between either algorithm on receiver operating characteristic analysis (p=0.26), although on visual analysis, there was an increase in sensitivity and small decrease in specificity and accuracy on BPL. Conclusion BPL increases SBR, SNR and SUVmax of mediastinal nodes in NSCLC compared to OSEM, but did not improve the accuracy for determining nodal involvement
