Molecular, behavioral and anatomical sophistication in spider webs: insights from spinning gland RNA-seq experiments in primitive and modern spiders.

RNA-seq experiments conducted in 454 sequencers were carried out to produce 87.000 short-reads representing the transcriptome of two spiders'spinning glands. We produced sequences from (i) Actinopus sp., a spider from the Mygalomorphae clade, and (ii) Gasteracantha cancriformis, an Orbicularia spider. Mygalomorphae spiders are known to retain a number of primitive morphological and behavioral characters. They use mixtures of a primitive web, soil, and plants only to cover a burrow they make on the ground for shelter and predation. On the other hand, Orbicularia spiders show a number of derivative spider's characters and they are capable to build different and complex silks used in a variety of situations. It is interesting to note that the complexity of web production, usage and behavior in these spiders is reflected both by (i) the variety of the repertoire of protein molecules (spidroins) they use to make their webs and (ii) the complexity of their anatomical spinning gland apparatu used to produce silk. Here we have first conducted a broad analysis of the spinning gland transcriptome in both spiders producing unigenes and categorizing annotated genes in biological functions. Then we started to analyze the number and variety of spider silk proteins and families found in different spider clades. We have shown that spiders using web only for a limited number of situations present a less sophisticated morphological spinning apparatus and produce a small repertoire of spidroin molecules. Phylogenetic analyses were conductedin the 3'region of spidroins and we try to relate (i) the evolution of silk protein families,(ii) the evolutionary complexification of silk production behavior and web usage, and (iii) the appearance of new specialized spinning glands along the evolution of specimens and clades in the Araneae order

Aerosol-Assisted CVD-Grown PdO Nanoparticle-Decorated Tungsten Oxide Nanoneedles Extremely Sensitive and Selective to Hydrogen

We report for the first time the successful synthesis of palladium (Pd) nanoparticle (NP)-decorated tungsten trioxide (WO3) nanoneedles (NNs) via a two-step aerosol-assisted chemical vapor deposition approach. Morphological, structural, and elemental composition analysis revealed that a Pd(acac)2 precursor was very suitable to decorate WO3 NNs with uniform and well-dispersed PdO NPs. Gas-sensing results revealed that decoration with PdO NPs led to an ultrasensitive and selective hydrogen (H2) gas sensor (sensor response peaks at 1670 at 500 ppm of H2) with low operating temperature (150 °C). The response of decorated NNs is 755 times higher than that of bare WO3 NNs. Additionally, at a temperature near that of the ambient temperature (50 °C), the response of this sensor toward the same concentration of H2 was 23, which is higher than that of some promising sensors reported in the literature. Finally, humidity measurements showed that PdO/WO3 sensors displayed low-cross-sensitivity toward water vapor, compared to bare WO3 sensors. The addition of PdO NPs helps to minimize the effect of ambient humidity on the sensor response

Safety and effectiveness of Copaiba Oleoresin (C. reticulata Ducke) on inflammation and tissue repair of oral wounds in rats.

In traditional communities of the Brazilian Amazon, the copaiba oleoresin (C. reticulata Ducke) is widely known for its therapeutic activity, especially its wound healing and anti-inflammatory actions. Our study aimed to evaluate these effects in oral lesions and the safety of the dosage proposed. A punch biopsy wound was induced on the ventral surface of the tongue of forty-five male Wistar rats under anesthesia. Animals were randomly allocated to one of three groups based on the treatment: control, corticoid and copaiba. A daily dose of each treatment and vehicle was administrated by oral gavage for three consecutive days. Sample collections took place on the third, seventh and 15th days post-wounding for clinical and histopathological analyses. Blood was collected on the third and seventh days for kidneys and liver function tests. Semi-quantitative analyses were performed based on scores of inflammation and reepithelization. Tissue collagen deposition was detected by PicroSirius red staining. Copaiba-treated wounds revealed a smaller wound area, decreased of acute inflammatory reaction and enhanced reepithelization. The levels of kidney and liver function tests did not reveal presence of damage post-treatments. Our findings suggest that copaiba oleoresin is a safe and effective alternative therapy for inflammation and tissue repair of oral wounds in this animal model
