    The future of the Amazon: new perspectives from climate, ecosystem and social sciences

    The potential loss or large-scale degradation of the tropical rainforests has become one of the iconic images of the impacts of twenty-first century environmental change and may be one of our century's most profound legacies. In the Amazon region, the direct threat of deforestation and degradation is now strongly intertwined with an indirect challenge we are just beginning to understand: the possibility of substantial regional drought driven by global climate change. The Amazon region hosts more than half of the world's remaining tropical forests, and some parts have among the greatest concentrations of biodiversity found anywhere on Earth. Overall, the region is estimated to host about a quarter of all global biodiversity. It acts as one of the major ‘flywheels’ of global climate, transpiring water and generating clouds, affecting atmospheric circulation across continents and hemispheres, and storing substantial reserves of biomass and soil carbon. Hence, the ongoing degradation of Amazonia is a threat to local climate stability and a contributor to the global atmospheric climate change crisis. Conversely, the stabilization of Amazonian deforestation and degradation would be an opportunity for local adaptation to climate change, as well as a potential global contributor towards mitigation of climate change. However, addressing deforestation in the Amazon raises substantial challenges in policy, governance, sustainability and economic science. This paper introduces a theme issue dedicated to a multidisciplinary analysis of these challenges

    Immediate Cytotoxicity But Not Degranulation Distinguishes Effector and Memory Subsets of CD8+ T Cells

    CD8+ T cells play a central role in the resolution and containment of viral infections. A key effector function of CD8+ T cells is their cytolytic activity toward infected cells. Here, we studied the regulation of cytolytic activity in naive, effector, and central versus effector memory CD8+ T cells specific for the same glycoprotein-derived epitope of lymphocytic choriomeningitis virus. Our results show that the kinetics of degranulation, assessed by a novel flow cytometric based assay, were identical in effector and both subsets of memory CD8+ T cells, but absent in naive CD8+ T cells. However, immediate cytolytic activity was most pronounced in effector T cells, low in effector memory T cells, and absent in central memory T cells, correlating with the respective levels of cytolytic effector molecules present in lytic granules. These results indicate that an inherent program of degranulation is a feature of antigen-experienced cells as opposed to naive CD8+ T cells and that the ability of CD8+ T cells to induce target cell apoptosis/death is dependent on granule protein content rather than on the act of degranulation itself. Furthermore, these results provide a potential mechanism by which central memory CD8+ T cell–mediated death of antigen-presenting cells within the lymph node is avoided

    Regional disparities and seasonal differences in climate risk to rice labour.

    The 880 million agricultural workers of the world are especially vulnerable to increasing heat stress due to climate change, affecting the health of individuals and reducing labour productivity. In this study, we focus on rice harvests across Asia and estimate the future impact on labour productivity by considering changes in climate at the time of the annual harvest. During these specific times of the year, heat stress is often high compared to the rest of the year. Examining climate simulations of the Coupled Model Intercomparison Project 6 (CMIP6), we identified that labour productivity metrics for the rice harvest, based on local wet-bulb globe temperature, are strongly correlated with global mean near-surface air temperature in the long term (p ≪ 0.01, R 2 > 0.98 in all models). Limiting global warming to 1.5 °C rather than 2.0 °C prevents a clear reduction in labour capacity of 1% across all Asia and 2% across Southeast Asia, affecting the livelihoods of around 100 million people. Due to differences in mechanization between and within countries, we find that rice labour is especially vulnerable in Indonesia, the Philippines, Bangladesh, and the Indian states of West Bengal and Kerala. Our results highlight the regional disparities and importance in considering seasonal differences in the estimation of the effect of climate change on labour productivity and occupational heat-stress

    p66Shc activation promotes increased oxidative phosphorylation and renders CNS cells more vulnerable to amyloid beta toxicity

    A key pathological feature of Alzheimer’s disease (AD) is the accumulation of the neurotoxic amyloid beta (Aβ) peptide within the brains of affected individuals. Previous studies have shown that neuronal cells selected for resistance to Aβ toxicity display a metabolic shift from mitochondrial-dependent oxidative phosphorylation (OXPHOS) to aerobic glycolysis to meet their energy needs. The Src homology/collagen (Shc) adaptor protein p66Shc is a key regulator of mitochondrial function, ROS production and aging. Moreover, increased expression and activation of p66Shc promotes a shift in the cellular metabolic state from aerobic glycolysis to OXPHOS in cancer cells. Here we evaluated the hypothesis that activation of p66Shc in CNS cells promotes both increased OXPHOS and enhanced sensitivity to Aβ toxicity. The effect of altered p66Shc expression on metabolic activity was assessed in rodent HT22 and B12 cell lines of neuronal and glial origin respectively. Overexpression of p66Shc repressed glycolytic enzyme expression and increased both mitochondrial electron transport chain activity and ROS levels in HT22 cells. The opposite effect was observed when endogenous p66Shc expression was knocked down in B12 cells. Moreover, p66Shc activation in both cell lines increased their sensitivity to Aβ toxicity. Our findings indicate that expression and activation of p66Shc renders CNS cells more sensitive to Aβ toxicity by promoting mitochondrial OXPHOS and ROS production while repressing aerobic glycolysis. Thus, p66Shc may represent a potential therapeutically relevant target for the treatment of AD

    Potential for Carbon Credits from Conservation Management: Price and Potential for Multi-Habitat Nature-Based Carbon Sequestration in Dorset, UK

    Carbon offsetting is currently a major tool in managing carbon emissions and informing sustainability plans of organisations in the drive to net-zero. This study aims to identify the offsetting potential of existing conservation schemes, and whether carbon offsetting credits could provide finance these conservation activities. The results from Dorset, in the UK, indicate that many existing conservation schemes in woodlands, heathlands, and grasslands cannot only enhance biodiversity but also capture significant amounts of carbon, and while habitats differ by region and country, the general results should be applicable elsewhere. We show that the cost per additional tonne of carbon sequestered as a result of conservation activities varies considerably between different conservation projects. On average, across the conservation projects we studied, the cost of this offsetting is GBP 80 per tonne CO2e sequestered and ranging between GBP 120 and GBP 0, depending on the project and whether existing biodiversity grants would be available. However, this figure was based on adapting and refining the existing conservation projects and did not involve expensive factors, such as purchase of land, which make the prices potentially unrealistic, especially in a Global North context. While the costs identified are higher than many offsetting schemes at present, it could present a useful option for those wishing to localise their offsetting. The concept is highly scalable and could remove significant amounts of carbon dioxide. Combining the approach with biodiversity credits or other credit schemes could make the higher costs more attractive to potential buyers
