Abstract A polyclonal antibody was raised against a Toxoplasma gondii 14-3-3^gluthatione S-transferase fusion protein obtained by cloning a 14-3-3 cDNA sequence determined from the T. gondii database. This antibody specifically recognized T. gondii 14-3-3 without any crossreaction with mammalian proteins. Immunofluorescence microscopy studies of the tachyzoites or the T. gondii-infected cells suggested cytosolic and membranous localizations of 14-3-3 protein. Different subcellular fractions were prepared for electrophoresis analysis and immunodetection. 14-3-3 proteins were found in the cytosol, the membrane fraction and Triton X-100-resistant membranes. Two 14-3-3 isoforms were detected. The major one was mainly cytoplasmic and to a lesser extent membrane-associated, whereas the minor isoform was associated with the detergent-resistant lipid rafts.

Toward an Observation of Volcanic ASH: Which Kind of Observation can be Made by Different Instruments

    No full text
In the first part, this presentation describes how an intercomparison was conducted between several lidars and ceilometers, the processing algorithms used, the difficulties encountered and the method used to design an optimum volcanic ash detection network. In the second part, we describe the work progress of a feasibility study concerning a WMO intercomparison for volcanic ash detection that is beginning with experts and colleagues from many others countries

Emotional labor: The role of organizational dehumanization

In a permanent quest for profit, employees can be reduced to a mere function or instrument, dissociated from their quality as individuals for the organization's ends. Experiencing such a feeling as an employee has been called organizational dehumanization. Scholars have recently suggested that organizational dehumanization may play a key role in the development of emotional labor. However, how organizational dehumanization and two main emotional labor strategies (i.e., surface and deep acting) are causally related remains unclear in this literature. In the present research, we argue that employees who experience organizational dehumanization and whose self is thus threatened then engage in surface acting to "conserve" their self or in deep acting to "give up" their self in service of the role. Overall, the combined results of three studies offer strong evidence that organizational dehumanization leads employees to perform more surface acting, but not more deep acting. Unexpectedly, our findings also indicate that deep acting reduces the perception of being dehumanized by the organization. In showing this, the present research sheds light on the potential dark side of deep acting, by suggesting that this strategy can change employees' perspectives in a way that may encourage them to stay in an organization that treats them as a means to an end

Pd-Catalyzed and Copper Assisted Regioselective Sequential C2 and C7 Arylation of Thiazolo[5,4-f]quinazolin-9(8H)-one with Aryl Halides

International audienceA selective functionalization of thiazolo[5,4-f]quinazolin-9(8H)-one has been developed through sequential activation of C–H bonds to furnish diarylated compounds. This strategy allows the regioselective C2 and C7 arylation by a judicious choice of coupling partners and bases, requiring no additional ligands or directing groups. Differently substituted N8-benzylated-2,7-diaryl-thiazoloquinazolin-9(8H)-ones were thereby obtained in a facile manner. A one-pot procedure was also performed. These protocols provide a synthetically useful route for late-stage functionalization of this highly valuable scaffold, required in drug discovery

Variability and vulnerability to comorbid conditions in addictive disorders

    No full text
Addictive disorders are a major burden worldwide, including high mortality rates. Part of this burden can be attributed to comorbid psychiatric disorders, especially suicidal behavior. Our project aims at characterizing the genetic and clinical vulnerability associated with the comorbidity between addictive and psychiatric disorders, focusing on suicidal risk
