    Water hyacinth (Eichhornia crassipes) and water lettuce (Pistia stratiotes) in the Great Lakes: Playing with fire?

    The Laurentian Great Lakes have been successfully invaded by at least 182 nonindigenous species. Here we report on two new species, water hyacinth Eichhornia crassipes and water lettuce Pistia stratiotes, that were found at a number of locations in Lake St. Clair and Detroit River during autumn 2010. Both species are commonly sold in the water garden and aquarium trade in southern Ontario and elsewhere. While it is not clear whether these species are established or can establish in the Great Lakes, the historic assumption that neither of these subtropical to tropical plants pose an invasion risk must be questioned in the light of changing environmental conditions associated with climate warming that may render Great Lakes\u27 habitats more suitable for these species and increase the likelihood of their successful establishment. © 2011 The Author(s)

    Identification of BRCA1 missense substitutions that confer partial functional activity: potential moderate risk variants?

    Introduction: Many of the DNA sequence variants identified in the breast cancer susceptibility gene BRCA1 remain unclassified in terms of their potential pathogenicity. Both multifactorial likelihood analysis and functional approaches have been proposed as a means to elucidate likely clinical significance of such variants, but analysis of the comparative value of these methods for classifying all sequence variants has been limited. Methods: We have compared the results from multifactorial likelihood analysis with those from several functional analyses for the four BRCA1 sequence variants A1708E, G1738R, R1699Q, and A1708V. Results: Our results show that multifactorial likelihood analysis, which incorporates sequence conservation, co-inheritance, segregation, and tumour immunohistochemical analysis, may improve classification of variants. For A1708E, previously shown to be functionally compromised, analysis of oestrogen receptor, cytokeratin 5/6, and cytokeratin 14 tumour expression data significantly strengthened the prediction of pathogenicity, giving a posterior probability of pathogenicity of 99%. For G1738R, shown to be functionally defective in this study, immunohistochemistry analysis confirmed previous findings of inconsistent 'BRCA1-like' phenotypes for the two tumours studied, and the posterior probability for this variant was 96%. The posterior probabilities of R1699Q and A1708V were 54% and 69%, respectively, only moderately suggestive of increased risk. Interestingly, results from functional analyses suggest that both of these variants have only partial functional activity. R1699Q was defective in foci formation in response to DNA damage and displayed intermediate transcriptional transactivation activity but showed no evidence for centrosome amplification. In contrast, A1708V displayed an intermediate transcriptional transactivation activity and a normal foci formation response in response to DNA damage but induced centrosome amplification. Conclusion: These data highlight the need for a range of functional studies to be performed in order to identify variants with partially compromised function. The results also raise the possibility that A1708V and R1699Q may be associated with a low or moderate risk of cancer. While data pooling strategies may provide more information for multifactorial analysis to improve the interpretation of the clinical significance of these variants, it is likely that the development of current multifactorial likelihood approaches and the consideration of alternative statistical approaches will be needed to determine whether these individually rare variants do confer a low or moderate risk of breast cancer

    Perception of Thermal Pain and the Thermal Grill Illusion Is Associated with Polymorphisms in the Serotonin Transporter Gene

    AIM: The main aim of this study was to assess if the perception of thermal pain thresholds is associated with genetically inferred levels of expression of the 5-HT transporter (5-HTT). Additionally, the perception of the so-called thermal grill illusion (TGI) was assessed. Forty-four healthy individuals (27 females, 17 males) were selected a-priori based on their 5-HTTLPR/rs25531 ('tri-allelic 5-HTTLPR') genotype, with inferred high or low 5-HTT expression. Thresholds for heat- and cold-pain were determined along with the sensory and affective dimensions of the TGI. RESULTS: Thresholds to heat- and cold-pain correlated strongly (rho  = -0.58, p<0.001). Individuals in the low 5-HTT-expressing group were significantly less sensitive to heat-pain (p = 0.02) and cold-pain (p = 0.03), compared to the high-expressing group. A significant gender-by-genotype interaction also emerged for cold-pain perception (p = 0.02); low 5-HTT-expressing females were less sensitive. The TGI was rated as significantly more unpleasant (affective-motivational dimension) than painful (sensory-discriminatory dimension), (p<0.001). Females in the low 5-HTT expressing group rated the TGI as significantly less unpleasant than high 5-HTT expressing females (p<0.05), with no such differences among men. CONCLUSION/SIGNIFICANCE: We demonstrate an association between inferred low 5-HTT expression and elevated thresholds to thermal pain in healthy non-depressed individuals. Despite the fact that reduced 5-HTT expression is a risk factor for chronic pain we found it to be related to hypoalgesia for threshold thermal pain. Low 5-HTT expression is, however, also a risk factor for depression where thermal insensitivity is often seen. Our results may thus contribute to a better understanding of the molecular underpinnings of such paradoxical hypoalgesia. The results point to a differential regulation of thermoafferent-information along the neuraxis on the basis of 5-HTT expression and gender. The TGI, suggested to rely on the central integration of thermoafferent-information, may prove a valuable tool in probing the affective-motivational dimension of these putative mechanisms

    Determinants of rapid response success for alien invasive species in aquatic ecosystems

    Alien invasive species (AIS) have received much attention for their harmful effects on health, ecology and the global economy. In response to this threat, many countries have adopted the Convention on Biological Diversity, which requires prevention or eradication of AIS. The best management approach is prevention, however when this fails and AIS establish, it is imperative that cost-efficient, rapid-response (RR) countermeasures be available. We performed a meta-analysis of case studies involving successful and failed RR to AIS in temperate aquatic ecosystems. We examined eight variables including ecosystem type (freshwater vs. marine), method type (chemical vs. mechanical), number of methods (multiple vs. single), taxonomy (animal vs. plant), population abundance (number of organisms), infestation extent (surface area of infestation), habitat size (surface area of management site), and project duration (length of project in number of months). Eradication success was significantly greater for plant (89 %) versus animal AIS (64 %) while suppression of AIS was most successful for projects using chemical versus mechanical methods and when conducted in small habitats. Managers should expect that taxonomy will be highly influential to the success of eradication-based RR, while both method type and management surface area influence suppression outcomes. © 2015, Springer International Publishing Switzerland

    Can tropical macrophytes establish in the Laurentian Great Lakes?

    Tropical macrophytes sold in the live garden trade are perceived as unlikely to invade temperate regions owing to climate mismatches. Here we study two tropical macrophytes (Pistia stratiotes and Eichhornia crassipes) not previously considered an invasion risk but which were recently discovered in the Great Lakes, and determine mechanisms that may be responsible for their continued presence including human introduction, reproduction through viable seeds and tolerance of winter conditions. Surveys conducted in 2011 and 2012 revealed recurrent presence of one or both species at some sites. Macrophytes in in situ enclosures failed to survive winter conditions, with plant health declining progressively prior to mortality. Water hyacinth seeds were field-collected, identified using Sanger sequencing, and germinated at 28°C with or without scarification. Germination was highest for scarified versus non-scarified seeds. Human introduction was observed at two sites, one involving both species, the other only water hyacinth. These species likely persist through a combination of annual reintroduction (both species) and possibly by production of viable seed (water hyacinth). Macrophytes, particularly water hyacinth, that were not previously viewed as a threat to the Great Lakes owing to environmental incompatibility may need to be reassessed. © 2015, Springer International Publishing Switzerland

    Reorganization and Preservation of Motor Control of the Brain in Spinal Cord Injury: A Systematic Review

    Reorganization of brain function in patients with CNS damage has been identified as one of the fundamental mechanisms involved in the recovery of sensori-motor function. Spinal cord injury (SCI) brain mapping studies during motor tasks aim for assessing the reorganization and preservation of brain networks involved in motor control. Revealing the activation of cortical and sub-cortical brain areas in patients with SCI can indicate principal patterns of brain reorganization when the neurotrauma is distal to the brain. This review assessed brain activation after SCI in terms of intensity, volume, and somatotopic localization, as well as preservation of activation during attempted and/or imagined movements. Twenty-five studies meeting the inclusion criteria could be identified in MEDLINE (1980 to January 2008). Relevant characteristics of studies (level of lesion, time after injury, motor task) and mapping techniques varied widely. Changes in brain activation were found in both cortical and subcortical areas of SCI subjects. In addition, several studies described a shift in the region of brain activation. These patterns appeared to be dynamic and influenced by the level, completeness and time after injury, as well as extent of clinical recovery. In addition, several aspects of reorganization of brain function following SCI resembled those reported in stroke. This review demonstrates that brain networks involved in different demands of motor control remain responsive even in chronic paralysis. These findings imply that therapeutic strategies aiming for restoring spinal cord function even in chronic SCI can build on a preserved competent brain control

    Redefining the subcellular location and transport of APC: new insights using a panel of antibodies

    Adenomatous polyposis coli (APC) is a tumour suppressor involved in colon cancer progression. We and others previously described nuclear–cytoplasmic shuttling of APC. However, there are conflicting reports concerning the localization of endogenous wild-type and tumour-associated, truncated APC. To resolve this issue, we compared APC localization using immunofluorescence (IF) microscopy and cell fractionation with nine different APC antibodies. We found that three commonly used APC antibodies showed nonspecific nuclear staining by IF and validated this conclusion in cells where APC was inactivated using small interfering RNA or Cre/Flox. Fractionation showed that wild-type and truncated APC from colon cancer cells were primarily cytoplasmic, but increased in the nucleus after leptomycin B treatment, consistent with CRM1-dependent nuclear export. In contrast to recent reports, our biochemical data indicate that APC nuclear localization is not regulated by changes in cell density, and that APC nuclear export is not prevented by truncating mutations in cancer. These results verify that the bulk of APC resides in the cytoplasm and indicate the need for caution when evaluating the nuclear accumulation of APC

    Are the Great Lakes at risk of new fish invasions from trans-Atlantic shipping?

    The 180+ invaders established in the Great Lakes include three fishes introduced by ballast water. Using previously published and new data, Snyder et al. (2014) identified a total of 10 possible \u27high risk\u27 fishes from Eastern Europe and assessed their respective abilities to survive open-ocean, ballast-water exchange during a trans-Atlantic voyage. Their study predicted that a subset of four species would not only survive ballast-water exchange but also invade and disrupt Great Lakes ecosystems. We have reassessed these results by incorporating information regarding opportunities for potential ballast water discharge by ships (i.e. propagule pressure) and a more robust requirement for salinity tolerance imposed by current ballast-water regulation. Our analysis reveals that only one species within the candidate group is likely to potentially pose a \u27high risk\u27 to the Great Lakes. © 2015 International Association for Great Lakes Research
