Artificial insemination in South American camelids and wild equids
An overview of the present status of the use of artificial insemination (AI) in South American camelids and wild equids is offered. Technical aspects of semen collection, dilution and cryopreservation have limited the development and use of AI in camelid and equid species. To-date, efficiency is low but progress has been made and viable offspring have been produced through the use of AI in domestic South American camelids using both fresh and frozen semen. The origin, composition, and function of the viscous component of camelid seminal plasma remain a mystery and an obvious area for future research. A better understanding of the normal constituents of seminal plasma will enable the rational design of semen extenders suitable for camelids. Post-thaw sperm viability is very low, and studies are needed to address questions of optimal freezing and thawing procedures as well as the insemination dose. The basis for differences in reported pregnancy rates with sexed and frozen semen in domestic equids, and the ultimate success of AI in wild equids will require continued research into the ‘‘stallion effect’’, extenders and cryoprotectants, optimal volume and number of spermatozoa, temperatures during handling, processing an transport, and insemination techniques. In both camelids and equids, research on domestic species under controlled conditions provides and excellent opportunity to develop effective semen handling techniques for application in wild and endangered species of the respective families
Evidence to inform occupational therapy intervention with adults with intellectual disability: A scoping review
Importance: Occupational therapy practitioners use a range of habilitative and compensatory approaches to teach new skills or modify tasks and environments to address occupational performance among adults with intellectual disability (ID); therefore, they must identify and use available evidence to guide intervention planning. Objective: To summarize the scope of evidence that can inform occupational therapy intervention with adults with primary or comorbid ID. Data Sources: Articles published in peer-reviewed journals between January 2002 and January 2018 and indexed in PubMed, CINAHL, PsycINFO, and Scopus. Study Selection and Data Collection: A total of 159 articles met inclusion criteria and contained information on occupational therapy intervention with adults ages 18 yr or older with primary or coexisting ID. Findings: Fifty-seven of the 159 articles focused on intervention to address occupational performance outcomes (i.e., employment, self-care, leisure and social interaction, community participation). Conclusions and Relevance: A wide range of literature is available to support occupational therapy intervention with adults with ID. Occupational therapy practitioners may be less knowledgeable about this evidence because much of it is located in non-occupational therapy or international journals
Validation of a homologous canine relaxin radioimmunoassay and application with pregnant and non-pregnant Northern fur seals (\u3ci\u3eCallorhinus ursinus\u3c/i\u3e)
The primary objectives of this study were to validate a canine relaxin RIA for use in otariids and phocids and consider practical applications. For 6 captive Northern fur seal females, serum samples were grouped and examined according to pregnancy (n = 13), post-partum (n = 8) and non-pregnancy (n = 6), and, for 2 captive Northern fur seal males, serum samples were grouped and examined together regardless of age (2 mo–15 yrs, n = 6). Placental tissue was available for examination from one Northern fur seal, Steller sea lion and harbor seal. The validation process involved several steps using an acid-acetone extraction process to isolate a relaxin-containing fraction in pools of serum from each group of fur seals and placental tissue from each seal species. A relaxin-like substance was detected in extracts of pregnant, non-pregnant and male serum and placental tissue in a dose-responsive manner as increasing volumes of respective extracts or amounts of canine relaxin were introduced into the assay. In raw serum samples, mean immuno-reactive relaxin concentrations were higher (P \u3c 0.05) during pregnancy than post-partum and non-pregnancy, and lower (P \u3c 0.05) in male than female fur seals. During pregnancy, mean serum concentrations of relaxin progressively increased (P \u3c 0.05) over Months 4–10 and, in serial samples collected from the same fur seals before and after parturition, mean concentrations were higher (P \u3c 0.06) pre-partum than post-partum. In conclusion, validation of a homologous canine relaxin RIA for use in otariids and phocids resulted in the discovery of a relaxin-like substance in extracted and raw serum and placental tissue from Northern fur seals, a Steller sea lion and harbor seal. Distinctly higher immuno-reactive concentrations during pregnancy indicated the potential for relaxin to serve as a hormonal marker to differentiate between pregnant and non-pregnant or pseudopregnant pinnipeds

Evaluation of Stress Accompanying Immunocontraceptive Vaccination in Donkeys
The overpopulation of donkeys is recognized as a problem in many parts of the world. The main concerns with uncontrolled donkey populations are habitat degradation and competition for feed resources between donkeys and other species. One of the most effective and humane solutions is the use of immunocontraception. Therefore, this study sought to evaluate the stress imposed by the use of two formulations of a zona pellucida (ZP) vaccine, a recombinant (reZP) and a native porcine (pZP) vaccine, both formulated with a Freund’s adjuvant. The stress was objectively measured using fecal cortisol concentrations and physical examination parameters at fixed points before and after vaccination. We hypothesized that fewer changes in physical exam parameters and lower fecal cortisol concentrations would be stimulated in jennies treated with the reZP vaccine due to the selection of specific proteins. Twenty-five reproductively sound jennies were randomly assigned to reZP (n = 9), pZP (n = 8) or control (n = 8) groups. The vaccines were administered at five-week intervals. Physical exam parameters and body wall thickness of injection sites were recorded for each jenny for four days post-injections. Fecal samples were obtained every other day from day 0 (first vaccination) through day 6 and on days 35 to 41 after booster. Injection site reactions were common in all groups with the reZP and pZP groups being overrepresented. Lameness was observed in the pZP and reZP groups that were affected by injection site reactions and open abscesses. The present study showed an increase in fecal cortisol concentrations within 4 days after the first vaccination with ZP vaccines and, thereafter, a decrease in cortisol 35 days later after the second vaccination, especially in donkeys with open abscesses. Our results suggest that acute stress (increased cortisol) was induced after the first vaccination, and chronic stress (decreased cortisol) occurred thereafter in association with open abscesses. In conclusion, reZP and pZP formulated with Freund’s adjuvant induced local inflammatory reactions with a differential degree of acute and chronic stress in donkeys
Temporal interrelationships among luteolysis, FSH and LH concentrations and follicle deviation in mares
The effect of altered LH concentrations on the deviation in growth rates between the 2 largest follicles was studied in pony mares. The progestational phase was shortened by administration of PGF2α on Day 10 (Day 0=ovulation; n=9) or lengthened by daily administration of 100 mg of progesterone on Days 10 to 30 (n=11; controls, n=10). All follicles ≥5 mm were ablated on Day 10 in all groups to initiate a new follicular wave. The interovulatory interval was not altered by the PGF2α treatment despite a 4-day earlier decrease in progesterone concentrations. Time required for growth of the follicles of the new wave apparently delayed the interval to ovulation after luteolysis. The FSH concentrations of the first post-ablation FSH surge were not different among groups. A second FSH surge with an associated follicular wave began by Day 22 in 7 of 11 mares in the progesterone group and in 0 of 19 mares in the other groups, indicating reduced functional competence of the largest follicle. A prolonged elevation in LH concentrations began on the mean day of wave emergence (Day 11) in the prostaglandin group (19.2 ± 2.2 vs 9.0 ± 0.7 ng/mL in controls; P<0.05), an average of 4 d before an increase in the controls. Concentrations of LH in the progesterone group initially increased until Day 14 and then decreased so that by Day 18 the concentrations were lower (P<0.05) than in the control group (12.9 ± 1.6 vs 20.2 ± 2.6 ng/mL). Neither the early and prolonged increase nor the early decrease in LH concentrations altered the growth profile of the second-largest follicle, suggesting that LH was not involved in the initiation of deviation. However, the early decrease in LH concentrations in the progesterone group was followed by a smaller (P<0.05) diameter of the largest follicle by Day 20 (26.9 ± 1.7 mm) than the controls (30.3 ± 1.7 mm), suggesting that LH was necessary for continued growth of the largest follicle after deviation. Role of luteinizing hormone in follicle deviation based on manipulating progesterone concentrations in mares
The effects of several doses of progesterone on FSH and LH concentrations were used to study the role of the gonadotropins on deviation in growth rates of the two largest follicles during the establishment of follicle dominance. Progesterone was given to pony mares at a daily dose rate of 0 mg (controls), 30 mg (low dose), 100 mg (intermediate dose), and 300 mg (high dose). All follicles ≥ 6 mm were ablated at Day 10 (Day 0 = ovulation) to initiate a new follicular wave; prostaglandin F(2α) was given to induce luteolysis, and progesterone was given from Days 10 to 24. The low dose did not significantly alter any of the ovarian or gonadotropin end points. The high dose reduced (P < 0.05) the ablation-induced FSH concentrations on Day 11. Maximum diameter of the largest follicle (17.2 ± 0.6 mm) and the second- largest follicle (15.5 ± 0.9 mm) in the high-dose group was less (P < 0.04) than the diameter of the second-largest follicle in the controls (20.0 ± 1.0 mm) at the beginning of deviation (Day 16.7 ± 0.4). Thus, the growth of the two largest follicles was reduced by the high dose, presumably through depression of FSH, so that the follicles did not attain a diameter characteristic of deviation in the controls. The intermediate dose did not affect FSH concentrations. However, the LH concentrations increased in the control, low, and intermediate groups, but then decreased (P < 0.05) in the intermediate group to pretreatment levels. The LH decrease in the intermediate group occurred 2 days before deviation in the controls. The maximum diameter of the largest follicle was less (P < 0.0001) in the intermediate group (27.3 ± 1.8 mm) than in the controls (38.9 ± 1.5 mm), but the maximum diameter of the second-largest follicle was not different between the two groups (19.0 ± 1.1 vs. 20.3 ± 1.0 mm). Thus, the onset of deviation, as assessed by the second-largest follicle, was not delayed by the decrease in LH. Diameter of the largest follicle by Day 18 in the intermediate group (23.1 ± 1.6 mm) was less (P < 0.05) than in the controls (28.0 ± 1.0 mm). These results suggest that circulating LH was not involved in the initiation of dominance (inhibition of other follicles by the largest follicle) but was required for the continued growth of the largest follicle after or concurrently with its initial expression of dominance

Abstract Two experiments were designed to determine if the suppressive effect of estradiol treatment on ovarian follicles in progestogen-implanted heifers is mediated directly at the ovary or systemically, at a higher level. The purpose of Experiment 1 was to determine a minimal effective dose of Ž . estradiol-17b E-17b that would induce follicle regression in progestogen-implanted heifers. Beef Ž . heifers were implanted with progestogen on Day 2 Day 0 s ovulation and were assigned Ž . Ž . randomly to five groups: control sesame seed oil, n s 9 ; 0.1 mg of E-17b n s 8 ; 0.5 mg of Ž . Ž . Ž . Ž . E-17b n s 8 ; 1 mg of E-17b n s 8 ; or 5 mg of E-17b n s 8 by intramuscular im injection Ž . on Day 3. Treatment with 5 and 1 mg of E-17b resulted in smaller P -0.05 day-to-day diameter profiles of the dominant follicle compared with controls, whereas 0.1 mg of E-17b did not have an apparent effect on follicle growth. The effect of a dose of 0.5 mg was intermediate Ž . and tended P -0.06 to result in a smaller diameter profile of the dominant follicle compared Ž . with control heifers. Experiment 2 was designed to utilize a subminimal dose of E-17b 0.1 mg , locally, to determine whether estradiol treatment induces follicle regression through a direct action on the ovary. Beef heifers received a progestogen ear implant on Day 2 and were assigned Ž . Ž . randomly to five groups on Day 3: control sesame seed oil, n s 8 ; 5 mg of E-17b im n s 8 ; Ž . 0.1 mg of E-17b im n s 8 ; 0.1 mg of E-17b given into the wall of the uterus, near the tip of the Ž Ž . . horn ipsilateral to the dominant follicle intrauterine iu , n s 8 ; or 0.1 mg of E-17b given into Ž Ž . . the stroma of the ovary, immediately adjacent to the dominant follicle intraovarian io , n s 6 . Ž . Local iu and io treatments were given via a transvaginal ultrasound-guided needle injection. Treatment with 5 mg of E-17b im resulted in suppression of the dominant follicle of the first Ž . follicular wave and early emergence of the second follicular wave P -0.05 . Diameter profiles of the dominant follicle in heifers treated with 0.1 mg im or 0.1 mg iu differed from those of control heifers on Day 5, whereas diameter profiles of the dominant follicle in heifers treated with 0.1 mg io did not differ from the controls. Daily changes in diameter of the dominant follicle did Ž . not differ among the three groups treated with 0.1 mg of E-17b im, iu and io . Hourly changes in circulating concentrations of FSH and LH were not detected following estradiol treatment either before or after the results were combined for all estradiol-treated groups. Results are supportive of the hypothesis that the suppressive effect of estradiol in cattle is exerted indirectly through a systemic route rather than directly at the ovary. Although low plasma concentrations of FSH and LH were not detected, systemic treatments with high E-17b dosages resulted in follicular suppression whereas local treatments with subminimal dosages, within the ovary bearing the dominant follicle, were without effect.
