    A commercial approach to the design and fabrication of an economical space power system is presented. Cost reductions are projected through the conceptual design of a 2 kW space power system built with the capability for having serviceability. The approach to system costing that is used takes into account both the constraints of operation in space and commercial production engineering approaches. The cost of this power system reflects a variety of cost/benefit tradeoffs that would reduce system cost as a function of system reliability requirements, complexity, and the impact of rigid specifications.     Trajectory-Based Dynamic Map Labeling

    In this paper we introduce trajectory-based labeling, a new variant of dynamic map labeling, where a movement trajectory for the map viewport is given. We define a general labeling model and study the active range maximization problem in this model. The problem is NP-complete and W[1]-hard. In the restricted, yet practically relevant case that no more than k labels can be active at any time, we give polynomial-time algorithms.     The use of wastewater analysis in forensic intelligence: drug consumption comparison between Sydney and different European cities

Wastewater analysis offers objective and complementary information to illicit drug agencies by monitoring patterns of illicit drug consumption. In this study, wastewater samples from three different wastewater treatment plants in Sydney, Australia were collected in March 2016. Ten targeted drugs were analysed and temporal and geographical analyses were performed to obtain a better understanding of the type and amount of illicit drugs consumed in Sydney in comparison with similar studies conducted around Australia and in Europe. Among the targeted drugs, methamphetamine was consumed the most, followed by cocaine and 3,4-methylenedioxymethamphetamine (MDMA). Weekly patterns were observed where a peak during the weekend was present. The geographical analysis showed differences between the regions targeted. This observation may be related to socio-demographic aspects. The comparison of our study to other data sources from Australia showed a high consumption of methamphetamine in Sydney and Western Australia. The comparison between Sydney and different European cities revealed a difference in consumption, which is in line with traditional market indicators. The information obtained through wastewater analysis provides complementary information regarding illicit drug consumption, the size, and the evolution of the illicit drug market. This, ultimately, will assist authorities in making informed decisions

    Lorentz Violation in Extra Dimensions

    In theories with extra dimensions it is well known that the Lorentz invariance of the D=4+nD=4+n-dimensional spacetime is lost due to the compactified nature of the nn dimensions leaving invariance only in 4d. In such theories other sources of Lorentz violation may exist associated with the physics that initiated the compactification process at high scales. Here we consider the possibility of capturing some of this physics by analyzing the higher dimensional analog of the model of Colladay and Kostelecky. In that scenario a complete set of Lorentz violating operators arising from spontaneous Lorentz violation, that are not obviously Planck-scale suppressed, are added to the Standard Model action. Here we consider the influence of the analogous set of operators which break Lorentz invariance in 5d within the Universal Extra Dimensions picture. We show that such operators can greatly alter the anticipated Kaluza-Klein(KK) spectra, induce electroweak symmetry breaking at a scale related to the inverse compactification radius, yield sources of parity violation in, e.g., 4d QED/QCD and result in significant violations of KK-parity conservation produced by fermion Yukawa couplings, thus destabilizing the lightest KK particle.     Retrospective suspect and non-target screening combined with similarity measures to prioritize MDMA and amphetamine synthesis markers in wastewater

    3,4-Methylenedioxymethamphetamine (MDMA) and amphetamine are commonly used psychoactive stimulants. Illegal manufacture of these substances, mainly located in the Netherlands and Belgium, generates large amounts of chemical waste which is disposed in the environment or released in sewer systems. Retrospective analysis of high-resolution mass spectrometry (HRMS) data was implemented to detect synthesis markers of MDMA and amphetamine production in wastewater samples. Specifically, suspect and non-target screening, combined with a prioritization approach based on similarity measures between detected features and mass loads of MDMA and amphetamine was implemented. Two hundred and thirty-five 24 h-composite wastewater samples collected from a treatment plant in the Netherlands between 2016 and 2018 were analyzed by liquid chromatography coupled to high-resolution mass spectrometry. Samples were initially separated into two groups (i.e., baseline consumption versus dumping) based on daily loads of MDMA and amphetamine. Significance testing and fold-changes were used to find differences between features in the two groups. Then, associations between peak areas of all features and MDMA or amphetamine loads were investigated across the whole time series using various measures (Euclidian distance, Pearson's correlation coefficient, Spearman's rank correlation coefficient, distance correlation and maximum information coefficient). This unsupervised and unbiased approach was used for prioritization of features and allowed the selection of 28 presumed markers of production of MDMA and amphetamine. These markers could potentially be used to detect dumps in sewer systems, help in determining the synthesis route and track down the waste in the environment

    Preoperative inhibition in patients with irresectable locally advanced stage III melanoma

    Aim: Neoadjuvant treatment of locally advanced disease with BRAF inhibitors is expected to increase the likelihood of a R0 resection. We present six patients with stage III unresectable melanoma, neoadjuvantly treated with BRAF inhibitors. Methods: Patients with unresectable, BRAF-mutated, stage III melanoma, were treated with BRAF inhibitors between 2012 and 2015. Unresectability was determined based on clinical and/or radiological findings. At maximal response, resection was performed. The specimen was reviewed to determine the degree of response. Results: In five of six patients a radical resection was achieved. Postoperative complications were unremarkable. In five of six resected specimens, vital tumor tissue was found. Conclusion: Neoadjuvant BRAF inhibitor treatment of locally advanced melanoma is feasible and has the potential to facilitate an R0 resection

    Carriers of a novel frame-shift insertion in WNT16a possess elevated pancreatic expression of TCF7L2

    BACKGROUND: The discovery of TCF7L2 as a global type 2 diabetes (T2D) gene has sparked investigations to explore the clinical utility of its variants for guiding the development of new diagnostic and therapeutic strategies. However, interpreting the resulting associations into function still remains unclear. Canonical Wnt signaling regulates β-catenin and its binding with TCF7L2, which in turn is critical for the production of glucagon-like peptide-1 (GLP-1). This study examines the role of a novel frame-shift insertion discovered in a conserved region of WNT16a, and it is proposed that this mutation affects T2D susceptibility in conjunction with gene variants in TCF7L2. RESULTS: Our results predicted that the insertion would convert the upstream open reading frame in the Wnt16a mRNA to an alternative, in-frame translation initiation site, resulting in the prevention of nonsense-mediated decay, leading to a consequent stabilization of the mutated WNT16a message. To examine the role of Wnt16a in the Wnt signaling pathway, DNA and serum samples from 2,034 individuals (48% with T2D) from the Sikh Diabetes Study were used in this investigation. Prevalence of Wnt16a insertion did not differ among T2D cases (33%) and controls (32%). However, there was a 3.2 fold increase in Wnt16a mRNA levels in pancreatic tissues from the insertion carriers and a significant increase (70%, p < 0.0001) in luciferase activity in the constructs carrying the insertion. The expression of TCF7L2 mRNA in pancreas was also elevated (~23-fold) among the insertion carriers (p=0.003). CONCLUSIONS: Our results suggest synergistic effects of WNT16a insertion and the at-risk ‘T’ allele of TCF7L2 (rs7903146) for elevating the expression of TCF7L2 in human pancreas which may affect the regulation of downstream target genes involved in the development of T2D through Wnt/β-catenin/TCF7L2 signaling pathway. However, further studies would be needed to mechanistically link the two definitively
