Introduction: T-cell immunoglobulin and mucin domain-3 (TIM-3) is a transmembrane molecule first identified as an immunoregulator. This molecule is also expressed on leukemic cells in acute myeloid leukemia and master cell survival and proliferation. In this study, we aimed to explore the effect of TIM-3 interaction with its ligand galectin-9 (Gal-9) on glucose and lipid metabolism in AML cell lines. Methods: HL-60 and THP-1 cell lines, representing M3 and M5 AML subtypes, respectively, were cultured under appropriate conditions. The expression of TIM-3 on the cell surface was ascertained by flow cytometric assay. We used real-time PCR to examine the mRNA expression of GLUT-1, HK-2, PFKFB-3, G6PD, ACC-1, ATGL, and CPT-1A; colorimetric assays to measure the concentration of glucose, lactate, GSH, and the enzymatic activity of G6PD; MTT assay to determine cellular proliferation; and gas chromatography–mass spectrometry (GC-MS) to designate FFAs. Results: We observed the significant upregulated expression of GLUT-1, HK-2, PFKFB-3, ACC-1, CPT-1A, and G6PD and the enzymatic activity of G6PD in a time-dependent manner in the presence of Gal-9 compared to the PMA and control groups in both HL-60 and THP-1 cell lines (p > 0.05). Moreover, the elevation of extracellular free fatty acids, glucose consumption, lactate release, the concentration of cellular glutathione (GSH) and cell proliferation were significantly higher in the presence of Gal-9 compared to the PMA and control groups in both cell lines (p < 0.05). Conclusion: TIM-3/Gal-9 ligation on AML cell lines results in aerobic glycolysis and altered lipid metabolism and also protects cells from oxidative stress, all in favor of leukemic cell survival and proliferation.Isfahan University of Medical Science

Design of a robotic hand with a biologically-inspired parallel actuation system for prosthetic applications

Trabajo presentado al 34th Annual Mechanisms and Robotics Conference celebrado en Quebec del 15 al 18 de agosto de 2010.This paper presents the design of a robotic hand for prosthetic applications. The main characteristic of this robotic hand is its biologically-inspired parallel actuation system, which is based on the behavior/strength space of the Flexor Digitorum Profundus (FDP) and the Flexor Digitorum Superficialis (FDS) muscles. The design separates the strength space of the FDS and FDP muscles into a lighter strength region where finer manipulation and general approach tasks are executed, and a higher strength region where the more robust grasps are achieved. Two parallel actuator types and kinematic structures are designed to complement the requirements of both strength space regions. This research was performed under an award/contract from Telemedicine Advanced Technology Research Center (TATRC), of the U.S. Army Medical Research and Materiel Command (USAMRMC) of the U.S. Department of Defense.Peer Reviewe

A Modern View of Anomalies in the Metal Groups of the Periodic System of D.I.Mendeleev

The article is devoted to the 150th anniversary of the Periodic Table of Chemical Elements by D.I.Mendeleev. The fundamental law of nature, discovered by D.I.Mendeleev has anomalies and paradoxes associated with certain groups of metals. When studying the physical and chemical properties of complex metal compounds, many discrepancies can be found, namely, the location of elements in groups, which primarily relate to metals with different valences. By studying the approaches and methods for predicting the arrangement of chemical elements, it can be established that D.I.Mendeleev eliminated many differences for some metals during the formation of the Periodic system of chemical elements. D.I.Mendeleev developed a principle that excludes such errors when finding and discovering new elements. Analytical studies conducted by a Russian scientist helped to calculate the atomic masses and describe the properties of three elements not known at that time – «eka-boron», «eka-silicon», «eka-aluminum», the existence of which was proved and confirmed by subsequent discoveries of scandium, germanium, boron, and gallium. The paper provides a significant assessment of the forecasting of metals in various groups of the periodic system. Changes in the properties of some metals significantly influenced their location in the table of D.I.Mendeleev

Study of the kinetics of the process of producing pellets from red mud in a hydrogen flow

    Get PDF
The reduction kinetics of serial phase transitions of iron oxides during reduction to a metallized state with different modes of technical hydrogen supply has been studied and substantiated. The results of the pellets formation when 3-5 % molasses is added to the red mud as a binding reagent are presented. The dependences of the reduction rate of iron oxides on the hydrogen flow rate are obtained. Based on the results of the experiments, a kinetic model was constructed, and with the help of X-ray phase and spectral analysis, it was proved that the agglomerates formed after heat treatment received high strength due to the adhesion of reduced iron particles with red mud particles. The use of a new type of charge materials in melting units will reduce the amount of emissions and dust fractions, as well as increase the metal yield

Expanding 1,2,4-triketone Toolbox for use as Aluorinated Building Blocks in the Synthesis of Pyrazoles, Pyridazinones and β-diketohydrazones

Fluorinated lithium β-diketonates bearing a methyl acetal group behave in the condensation reactions with hydrazines as trielectrophilic building blocks for the preparation of pyrazoles, pyridazinones and β-diketohydrazones. For the first time, solvent-induced regioisomeric and heterocyclic ring size-controlled formation was observed for 1,2,4-triketone analogues. Fluoroalkylated acetyl NH-pyrazoles or substituted 5-RF-pyrazoles were obtained from the acid-catalyzed cyclocondensation of lithium β-diketonates with (aryl)hydrazines in ethanol. In methanol solvent acetyl-containing 3-CF3-pyrazoles were isolated because of inverse nucleophilic attack of arylhydrazines. The use of aprotic acetonitrile in the condensation resulted in regioselective trifluorinated pyridazinones and fluorinated β-diketohydrazones formation via initial acetal fragment interaction with N,N-dinucleophile. © 2021 Elsevier B.V.This work carried out in the framework of the basic theme of the Russian Academy of Sciences (state registration № AAAA-A19-119011790132-7). Abstract: The reactions of functionalized lithium CF3-β-diketonate (LiL) with europium(III) and terbium(III) chlorides in acetonitrile afford heterobimetallic complexes with the general formula [LiL-nL4(H2O)](CH3CN) (Ln = Eu (IIIa) and Tb (IIIb)). It is found that Ln–Li tetrakis-β-diketonates can also be prepared by the crystallization from acetonitrile of the previously synthesized complexes [(LnL3)(LiL)(MeOH)] (I) and [(LnL3)(LiL)(H2O)] (II). For the first time, the single crystals of the compounds in a series of Ln–Li tetrakis-β-diketonates are characterized by X-ray diffraction analysis (CIF files CCDC nos. 1973632 (IIIa) and 1973633 (IIIb)). The photoluminescence properties of the synthesized heterometallic complexes in the solid state are studied. © 2020, Pleiades Publishing, Ltd.This work was supported by the Russian Foundation for Basic Research (project no. 18-33-20124), Council for Grants of President of the Russian Federation (grant no.
